    Development of Bacillus thuringiensis CryIC resistance by Spodoptera exigua (Huebner) (Lepidoptera : Noctuidae)

    Selection of resistance in Spodoptera exigua (Hubner) to an HD-1 spore-crystal mixture, CryIC (HD-133) inclusion bodies, and trypsinized toxin from Bacillus thuringiensis' subsp, aizawai and B. thuringiensis subsp. entomocidus was attempted by using laboratory bioassays. No resistance to the HD-1 spore-crystal mixture could be achieved after 20 generations of selection. Significant levels of resistance (11-fold) to CryIC inclusion bodies expressed in Escherichia coli were observed after seven generations, Subsequent selection of the CryIC-resistant population with trypsinized CryIC toxin resulted, after 21 generations of CryIC selection, in a population of S. exigua that exhibited only 8% mortality at the highest toxin concentration tested (320 mu g/g), whereas the 50% lethal concentration was 4.30 mu g/g for the susceptible colony. Insects resistant to CryIC toxin from HD-133 also were resistant to trypsinized CryIA(b), CryIC from B. thuringiensis subsp. entomocidus, CryIE-CryIC fusion protein (G27), CryIH, and CryIIA. In vitro binding experiments with brush border membrane vesicles showed a twofold decrease in maximum CryIC binding, a fivefold difference in K-d, and no difference in the concentration of binding sites for the CryIC-resistant insects compared with those for the susceptible insects, Resistance to CryIC was significantly reduced by the addition of HD-1 spores, Resistance to the CryIC toxin was still observed 12 generations after CryIC selection was removed. These results suggest that, in S. exigua, resistance to a single protein is more likely to occur than resistance to spore crystal mixtures and that once resistance occurs, insects will be resistant to many other Cry proteins, These results have important implications for devising S. exigua resistance management strategies in the field

    Frameshift Mutagenesis and Microsatellite Instability Induced by Human Alkyladenine DNA Glycosylase

    Human alkyladenine DNA glycosylase (hAAG) excises alkylated purines, hypoxanthine, and etheno bases from DNA to form abasic (AP) sites. Surprisingly, elevated expression of hAAG increases spontaneous frameshift mutagenesis. By random mutagenesis of eight active site residues, we isolated hAAG-Y127I/H136L double mutant that induces even higher rates of frameshift mutation than does the wild-type hAAG; the Y127I mutation accounts for the majority of the hAAG-Y127I/H136L-induced mutator phenotype. The hAAG-Y127I/H136L and hAAG-Y127I mutants increased the rate of spontaneous frameshifts by up to 120-fold in S. cerevisiae and also induced high rates of microsatellite instability (MSI) in human cells. hAAG and its mutants bind DNA containing one and two base-pair loops with significant affinity, thus shielding them from mismatch repair; the strength of such binding correlates with their ability to induce the mutator phenotype.     The tax and price index

    A feature of this year's Budget was the shift in emphasis from direct taxation to indirect taxation. The annual yield of income tax has been reduced by £4½ billion while that of direct taxes was increased by a similar amount. It must be acknowledged that the new tax and price index has a number of deficiencies. It is only applicable to a proportion of the population, namely those who pay tax and whose gross income is less than £10,000 per annum. It takes no account of the social wage, nor of changes in benefits, such as the recent restructuring of child allowance. It smoothes out tax payments over a full year even though, for administrative reasons, these may fluctuate widely from month to month. This brief paper explores some of these deficiencies in more detail and assess their impact on taxation

    The Scottish economy [October 1979]

    The previous two issues of this Commentary have both indicated that the Scottish economy has been performing poorly since the mid 1970's. This is true in both an absolute and a relative sense. Manufacturing production only increased by 1.2% between 1976 and 1978 and, after dropping below 1975 levels in the first quarter of 1979, is unlikely to show any substantial improvement for the year as a whole. In an international context the 1975-1978 performance can best be described as appalling. Over the same period industrial production in Eire grew by 28%, in Japan and the US by 23% and in West Germany and France by 15%. Inertia in developing new markets and lack of competitiveness in existing markets both contributed substantially to the virtual stagnation of Scottish output. Because the problems are so diverse, so too must be the solutions

    Tourism in Scotland

    In 1978 13.2 million tourist and business travellers spent £523 million in Scotland, Foreign visitors, although only 8.5% of the total, contributed £158 million (ie 30.2%) of tourist expenditure. As such tourism is both an "invisible" export, since it is a means of earning foreign exchange, and a significant provider of jobs, many of which are created in the remoter rural areas. Despite its importance statistics relating specifically to tourism are not readily available from published sources. The "industry" actually combines the resources of a wide range of activities, most of which have economic and social links with other sectors of the economy independent from tourism. These include hotels and catering, transport and travel services as well as entertainment, sport and recreation facilities. Consequently the economic impact of tourism is generally assessed not in terms of employment and output but by more indirect measures such as numbers of visitors, or bednights spent in Scotland, as well as tourist expenditure and hotel occupancy data.     Moving tales, exploring narrative strategies for scalable locative audio drama.

    This paper reports on a recent collaboration between the Lansdown Centre for Electronic Arts at Middlesex University and the BBC Radio Drama Department, which was designed to investigate the narrative possibilities of locative media in a drama context. The locative drama Scratch is the first outcome of an ongoing research project, Locating Drama, whose aim is to investigate and develop narrative strategies that take full advantage of the current generation of GPS enabled portable computing devices for audio drama. In particular, we are exploring content and modes of interaction, which, while based on location awareness are not in any way site-specific allowing users to experience the drama in a location of their choice. We will refer to this approach as translocational as it allows the translation of locative media experiences to a wide variety of spaces. The translocational approach is of particular interest to broadcasters as it is more scalable than a site-specific paradigm, opening the possibility of downloadable location-aware podcasts featuring professionally authored content for a wide audience

    In the January issue of the Commentary is was pointed out that the mildly favourable prospects for world economic growth in 1979 were overshadowed by some political uncertainties, notably the prospect of disruptions to the supply of oil and the growth of protectionism. While the volume of world trade, as measured by the utilisation rates of dry cargo shipping, has continued to improve since the trough of June, 1978, there have been further less favourable political developments. This brief considers the impact of these developments and analyses the latest economic data

    Spheres of Practice for the Co-design of Wearables

    As expectations within the area of smart textiles increasingly become informed and driven by technological developments, the disciplinary boundaries and relationship between user and technological innovation will unavoidably transform. The authors venture that new paradigms of collaborative practice will inevitably develop between design and science, to more fully realize both the opportunities and contexts that wearable textiles offer. Drawing on previous work by the authors namely Molecular Imprinted Textiles (MIT - 2009/10), Future Textile Visions (FTV - 2010/11), Design Specks: Connecting People with Speckled Computing (2012/13), Second Skin (2013/14), and The S*** Word: Designing the Empathic Underwardrobe (2014), a model is proposed to more clearly understand and navigate between design, technology and application, and more importantly, between our cultural understanding of the user and the wearer. This paper reflects on a series of projects that inform a methodological approach: a process of asking questions; developing scenarios; exploring materials and making; generating concepts and building prototypes. Each project involved collaborations between design, academics, users and industry, and a form of co-design, where knowledge exchange was central, design was the intermediary, and the goal was to understand the drivers and the stakeholders. Simultaneously, this research sought to better understand and communicate the development of more empathic textile and fashion artifacts, and solutions. Co-design in this context is seen as a core approach to shifting the balance from technology as merely adjunct, or as a hook for marketers and users, to a more informed and harmonised position, where technology sits proximally and comfortably. The notion of interdisciplinary understanding, which tracks across domains of product, fashion and textiles, presents an approach where the application is still emerging. Through analysis of this progressive series of projects, the authors suggest that there is an opportunity to explore the inherent connectedness that textiles might offer for the integration and embedding of technology within material as a means to embrace these affordance opportunities. Central to this notion is the realisation of opportunities arising from dialogue and collaborative making (i.e. co-design), and for exploring the transformative notions of the user and the wearer. This paper led the authors to pose a set of questions that align to a four stage design process: Research, Define, Develop, Reflect, to frame findings and insights, and to outline the potential for future opportunities of working with technology to achieve the making and wearing of desirable materializations on the body

    Using trained dogs and organic semi-conducting sensors to identify asymptomatic and mild SARS-CoV-2 infections: an observational study

    BACKGROUND: A rapid, accurate, non-invasive diagnostic screen is needed to identify people with SARS-CoV-2 infection. We investigated whether organic semi-conducting (OSC) sensors and trained dogs could distinguish between people infected with asymptomatic or mild symptoms, and uninfected individuals, and the impact of screening at ports-of-entry. METHODS: Odour samples were collected from adults, and SARS-CoV-2 infection status confirmed using RT-PCR. OSC sensors captured the volatile organic compound (VOC) profile of odour samples. Trained dogs were tested in a double-blind trial to determine their ability to detect differences in VOCs between infected and uninfected individuals, with sensitivity and specificity as the primary outcome. Mathematical modelling was used to investigate the impact of bio-detection dogs for screening. RESULTS: About, 3921 adults were enrolled in the study and odour samples collected from 1097 SARS-CoV-2 infected and 2031 uninfected individuals. OSC sensors were able to distinguish between SARS-CoV-2 infected individuals and uninfected, with sensitivity from 98% (95% CI 95–100) to 100% and specificity from 99% (95% CI 97–100) to 100%. Six dogs were able to distinguish between samples with sensitivity ranging from 82% (95% CI 76–87) to 94% (95% CI 89–98) and specificity ranging from 76% (95% CI 70–82) to 92% (95% CI 88–96). Mathematical modelling suggests that dog screening plus a confirmatory PCR test could detect up to 89% of SARS-CoV-2 infections, averting up to 2.2 times as much transmission compared to isolation of symptomatic individuals only. CONCLUSIONS: People infected with SARS-CoV-2, with asymptomatic or mild symptoms, have a distinct odour that can be identified by sensors and trained dogs with a high degree of accuracy. Odour-based diagnostics using sensors and/or dogs may prove a rapid and effective tool for screening large numbers of people. Trial Registration NCT04509713 (clinicaltrials.gov)
