    The IMGT/HLA database

    It is 14 years since the IMGT/HLA database was first released, providing the HLA community with a searchable repository of highly curated HLA sequences. The HLA complex is located within the 6p21.3 region of human chromosome 6 and contains more than 220 genes of diverse function. Of these, 21 genes encode proteins of the immune system that are highly polymorphic. The naming of these HLA genes and alleles and their quality control is the responsibility of the World Health Organization Nomenclature Committee for Factors of the HLA System. Through the work of the HLA Informatics Group and in collaboration with the European Bioinformatics Institute, we are able to provide public access to these data through the website http://www.ebi.ac.uk/imgt/hla/. Regular updates to the website ensure that new and confirmatory sequences are dispersed to the HLA community and the wider research and clinical communities. This article describes the latest updates and additional tools added to the IMGT/HLA project

    Efficient processing of an antigenic sequence for presentation by MHC class I molecules depends on its neighboring residues in the protein

    Processing of endogenously synthesized proteins generates short peptides that are presented by MHC class I molecules to CD8 T lymphocytes. Here it is documented that not only the sequence of the presented peptide but also the residues by which it is flanked in the protein determine the efficiency of processing and presentation. This became evident when a viral sequence of proven antigenicity was inserted at different positions into an unrelated carrier protein. Not different peptides, but different amounts of the antigenic insert itself were retrieved by isolation of naturally processed peptides from cells expressing the different chimeric proteins. Low yield of antigenic peptide from an unfavorable integration site could be overcome by flanking the insert with oligo-alanine to space it from disruptive neighboring sequences. Notably, the degree of protection against lethal virus disease related directly to the amount of naturally processed antigenic peptide

    Making the city my own: uses and practices of mobile location technologies for exploration of a new city

    In this paper, we present an interview study of 13 recent newcomers to New York City, focusing on their early experiences of exploration and use of mobile location services and other tools for getting to know their new city. We describe their reasons and intentions behind exploratory practices using digital tools and emphasize how they make meaning out of new places in relation to technology tools as well as their previous places. Mobile location technologies make the process of finding specific places and exploring new neighborhoods a digital search task but discourage the notion of wandering and exploration. We point out missed opportunities for socio-technical systems supporting place making and place discovery and suggest that digital exploration tools should stay peripheral to the activities that people enjoy as tech-free but support a wider notion of search for salient characteristics of places

    The Turkey Ig-like receptor family: identification, expression and function.

    The chicken leukocyte receptor complex located on microchromosome 31 encodes the chicken Ig-like receptors (CHIR), a vastly expanded gene family which can be further divided into three subgroups: activating CHIR-A, bifunctional CHIR-AB and inhibitory CHIR-B. Here, we investigated the presence of CHIR homologues in other bird species. The available genome databases of turkey, duck and zebra finch were screened with different strategies including BLAST searches employing various CHIR sequences, and keyword searches. We could not identify CHIR homologues in the distantly related zebra finch and duck, however, several partial and complete sequences of CHIR homologues were identified on chromosome 3 of the turkey genome. They were designated as turkey Ig-like receptors (TILR). Using cDNA derived from turkey blood and spleen RNA, six full length TILR could be amplified and further divided according to the typical sequence features into one activating TILR-A, one inhibitory TILR-B and four bifunctional TILR-AB. Since the TILR-AB sequences all displayed the critical residues shown to be involved in binding to IgY, we next confirmed the IgY binding using a soluble TILR-AB1-huIg fusion protein. This fusion protein reacted with IgY derived from various gallinaceous birds, but not with IgY from other bird species. Finally, we tested various mab directed against CHIR for their crossreactivity with either turkey or duck leukocytes. Whereas no staining was detectable with duck cells, the CHIR-AB1 specific mab 8D12 and the CHIR-A2 specific mab 13E2 both reacted with a leukocyte subpopulation that was further identified as thrombocytes by double immunofluorescence employing B-cell, T-cell and thrombocyte specific reagents. In summary, although the turkey harbors similar LRC genes as the chicken, their distribution seems to be distinct with predominance on thrombocytes rather than lymphocytes

    Comparative Models in German Elections: Using the German Far-Right Party as a Proxy for Ethnic Conflict

    In 2017, the Alternative für Deutschland (AfD) became the first the far-right party to win seats in the Bundestag since 1933. By campaigning on ethnic division, the AfD saw an unprecedented rise in support, especially in East Germany. This paper tests two models of ethnic conflict within comparative politics, primordialism and constructivism, to see which better explains the result of the AfD’s 2017 election. By examining the rhetorical use of political advertisements, the Manifesto Project’s analysis of the AfD’s platform, and differences of support between East and West Germany, the paper finds that constructivism better shows that highly-educated elites within the AfD purposefully constructed ethnic division to gain political support. Clear implications are drawn for the future of the two ethnic models and for European far-right politics
