    The reproductive alterations induced by maternally inherited alpha-proteo-bacteria Wolbachia to their hosts is a well-documented phenomenon. In Nesolynx thymus, a gregarious hymenopterous ecto-pupal parasitoid of the uzifly, Exorista sorbillans, diagnostic PCR assay using specific primers revealed the presence of Wolbachia. Following genetic crossing experiments, we observed a female biased sex ratio of 1 : 9.5 at 25 degrees C and 1 : 3 male to female ratio when the populations were exposed to heat shock 33 degrees C for six hours. Furthermore, we found infection polymorphism, where female parasitoids are infected by Wolbachia but males are not infected. Infected eggs develop into females, whereas uninfected eggs develop parthenogenetically into males. The results are discussed in the context of the possible mechanism of sex-ratio bias caused by Wolbachia

    Despite advances in concept extraction from free text, finding meaningful health related information from online patient forums still poses a significant challenge. Here we demonstrate how structured information can be extracted from posts found in such online health related forums by forming relationships between a drug/treatment and a symptom or side effect, including the polarity/sentiment of the patient. In particular, a rule-based natural language processing (NLP) system is deployed, where information in sentences is linked together though anaphora resolution. Our NLP relationship extraction system provides a strong baseline, achieving an F1 score of over 80% in discovering the said relationships that are present in the posts we analysed

    The name “eukaryote” is derived from Greek, meaning “true kernel”, and describes the domain of organisms whose cells have a nucleus. The nucleus is thus the defining feature of eukaryotes and distinguishes them from prokaryotes (Archaea and Bacteria), whose cells lack nuclei. Despite this, we discuss the intriguing possibility that organisms on the path from the first eukaryotic common ancestor to the last common ancestor of all eukaryotes did not possess a nucleus at all—at least not in a form we would recognize today—and that the nucleus in fact arrived relatively late in the evolution of eukaryotes. The clues to this alternative evolutionary path lie, most of all, in recent discoveries concerning the structure of the nuclear pore complex. We discuss the evidence for such a possibility and how this impacts our views of eukaryote origins and how eukaryotes have diversified subsequent to their last common ancestor

    Novel synthetic inhibitors of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase activity that inhibit tumor cell proliferation and are structurally unrelated to existing statins

    Pilot-scale libraries of eight-membered medium ring lactams (MRLs) and related tricyclic compounds (either seven-membered lactams, thiolactams or amines) were screened for their ability to inhibit the catalytic activity of human recombinant 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase in vitro. A dozen of the synthetic compounds mimic the inhibition of purified HMG-CoA reductase activity caused by pravastatin, fluvastatin and sodium salts of lovastatin, mevastatin and simvastatin in this cell-free assay, suggesting direct interaction with the rate-limiting enzyme of cholesterol biosynthesis. Moreover, several MRLs inhibit the metabolic activity of L1210 tumor cells in vitro to a greater degree than fluvastatin, lovastatin, mevastatin and simvastatin, whereas pravastatin is inactive. Although the correlation between the concentration-dependent inhibitions of HMG-CoA reductase activity over 10 min in the cell-free assay and L1210 tumor cell proliferation over 4 days in culture is unclear, some bioactive MRLs elicit interesting combinations of statin-like (IC50: 7.4-8.0 µM) and anti-tumor (IC50: 1.4-2.3 µM) activities. The HMG-CoA reductase-inhibiting activities of pravastatin and an MRL persist in the presence of increasing concentrations of NADPH. But increasing concentrations of HMG-CoA block the HMG-CoA reductase-inhibiting activity of pravastatin without altering that of an MRL, suggesting that MRLs and existing statins may have different mechanisms of enzyme interaction and inhibition. When tested together, suboptimal concentrations of synthetic MRLs and existing statins have additive inhibitory effects on HMG-CoA reductase activity. Preliminary molecular docking studies with MRL-based inhibitors indicate that these ligands fit sterically well into the HMG-CoA reductase statin-binding receptor model and, in contrast to mevastatin, may occupy a narrow channel housing the pyridinium moiety on NADP+

Herein we describe a targeted carrier system that can deliver siRNA to cancer cells overexpressing the human epidermal growth factor 2 (HER2) receptor.
