    Regulation of intracellular cyclic GMP concentration by light and calcium in electropermeabilized rod photoreceptors.

    Abstract This study examines the regulation of cGMP by illumination and by calcium during signal transduction in vertebrate retinal photoreceptor cells. We employed an electropermeabilized rod outer segment (EP-ROS) preparation which permits perfusion of low molecular weight compounds into the cytosol while retaining many of the features of physiologically competent, intact rod outer segments (ROS). When nucleotide-depleted EP-ROS were incubated with MgGTP, time- and dose-dependent increases in intracellular cGMP levels were observed. The steady state cGMP concentration in EP-ROS (0.007 mol cGMP per mol rhodopsin) approached the cGMP concentration in intact ROS. Flash illumination of EP-ROS in a 250-nM free calcium medium resulted in a transient decrease in cGMP levels; this occurred in the absence of changes in calcium concentration. The kinetics of the cGMP response to flash illumination of EP-ROS were similar to that of intact ROS. To further examine the effects of calcium on cGMP metabolism, dark-adapted EP-ROS were incubated with MgGTP containing various concentrations of calcium. We observed a twofold increase in cGMP steady state levels as the free calcium was lowered from 1 μM to 20 nM; this increase was comparable to the behavior of intact ROS. Measurements of guanylate cyclase activity in EP-ROS showed a 3.5-fold increase in activity over this range of calcium concentrations, indicating a retention of calcium regulation of guanylate cyclase in EP-ROS preparations. Flash illumination of EP-ROS in either a 50- or 250-nM free calcium medium revealed a slowing of the recovery time course at the lower calcium concentration. This observation conflicts with any hypothesis whereby a reduction in free calcium concentration hastens the recovery of cytoplasmic cGMP levels, either by stimulating guanylate cyclase activity or by inhibiting phosphodiesterase activity. We conclude that changes in the intracellular calcium concentration during visual transduction may have more complex effects on the recovery of the photoresponse than can be accounted for solely by guanylate cyclase activation

    Specification and [visual] modelling languages are expected to combine strong abstraction mechanisms with rigour, scalability, and parsimony. LePUS3 is a visual, object-oriented design description language axiomatized in a decidable subset of the first-order predicate logic. We demonstrate how LePUS3 is used to formally specify a structural design pattern and prove (‗verify‘) whether any JavaTM 1.4 program satisfies that specification. We also show how LePUS3 specifications (charts) are composed and how they are verified fully automatically in the Two-Tier Programming Toolkit

    Joint perceptual decision-making: a case study in explanatory pluralism.

    Traditionally different approaches to the study of cognition have been viewed as competing explanatory frameworks. An alternative view, explanatory pluralism, regards different approaches to the study of cognition as complementary ways of studying the same phenomenon, at specific temporal and spatial scales, using appropriate methodological tools. Explanatory pluralism has been often described abstractly, but has rarely been applied to concrete cases. We present a case study of explanatory pluralism. We discuss three separate ways of studying the same phenomenon: a perceptual decision-making task (Bahrami et al., 2010), where pairs of subjects share information to jointly individuate an oddball stimulus among a set of distractors. Each approach analyzed the same corpus but targeted different units of analysis at different levels of description: decision-making at the behavioral level, confidence sharing at the linguistic level, and acoustic energy at the physical level. We discuss the utility of explanatory pluralism for describing this complex, multiscale phenomenon, show ways in which this case study sheds new light on the concept of pluralism, and highlight good practices to critically assess and complement approaches

    The terms architecture, design, and implementation are typically used informally in partitioning software specifications into three coarse strata of abstraction. But these strata are not well-defined in either research or practice and often overlap causing confusion and needless discussion. To remedy this problem we formally define two criteria: the Intension and the Locality Criteria, and show that the intuitive discrimination between the three terms architecture, design, and implementation is qualitative and not merely quantitative.     EVALUATION OF MARKET ADVISORY SERVICE PERFORMANCE IN HOGS

    The purpose of this paper is to investigate the pricing performance of agricultural market advisory services in hogs. Pricing recommendations are available for all quarters from the beginning of 1995 through the end of 2001. The results show that average differences between advisory programs and market benchmarks are small in nominal terms for all three benchmarks, -0.41/bu.,0.41/bu., O.OO/cwt. and $-0.27/cwt. versus the cash, index and empirical benchmarks, respectively, and none of the average differences are significantly different from zero. Hence, advisory programs as a group do not outperform the market benchmarks in terms of average price. Advisory programs also do not outperform the market benchmarks in terms of average price and risk. Finally, there is little evidence that advisory programs with superior performance can be usefully selected based on past performance in the hog market.Marketing,

    Exploring the movement dynamics of deception

    Both the science and the everyday practice of detecting a lie rest on the same assumption: hidden cognitive states that the liar would like to remain hidden nevertheless influence observable behavior. This assumption has good evidence. The insights of professional interrogators, anecdotal evidence, and body language textbooks have all built up a sizeable catalog of non-verbal cues that have been claimed to distinguish deceptive and truthful behavior. Typically, these cues are discrete, individual behaviors—a hand touching a mouth, the rise of a brow—that distinguish lies from truths solely in terms of their frequency or duration. Research to date has failed to establish any of these non-verbal cues as a reliable marker of deception. Here we argue that perhaps this is because simple tallies of behavior can miss out on the rich but subtle organization of behavior as it unfolds over time. Research in cognitive science from a dynamical systems perspective has shown that behavior is structured across multiple timescales, with more or less regularity and structure. Using tools that are sensitive to these dynamics, we analyzed body motion data from an experiment that put participants in a realistic situation of choosing, or not, to lie to an experimenter. Our analyses indicate that when being deceptive, continuous fluctuations of movement in the upper face, and somewhat in the arms, are characterized by dynamical properties of less stability, but greater complexity. For the upper face, these distinctions are present despite no apparent differences in the overall amount of movement between deception and truth. We suggest that these unique dynamical signatures of motion are indicative of both the cognitive demands inherent to deception and the need to respond adaptively in a social context

    Factors influencing adoption of conservation tillage in Australian cropping regions

    The purpose of this research is to improve understanding of conservation tillage adoption decisions by identifying key biophysical and socio-economic factors influencing no-till adoption by grain growers across four Australian cropping regions. The study is based on interviews with 384 grain growers using a questionnaire aimed at eliciting perceptions relating to a range of possible long- and short-term agronomic interactions associated with the relative economic advantage of shifting to a no-tillage cropping system. Together with other farm and farmer-specific variables, a dichotomous logistic regression analysis was used to identify opportunities for research and extension to facilitate more rapid adoption decisions. The broader systems approach to considering conservation tillage adoption identified important determinants of adoption not associated with soil conservation and erosion prevention benefits. Most growers recognised the erosion-reducing benefits of no-till but it was not an important factor in explaining whether a grower was an adopter or non-adopter. Perceptions associated with shorter-term crop production benefits under no-till, such as the relative effectiveness of pre-emergent herbicides and the ability to sow crops earlier on less rainfall were influential.     24 \textmu m length spin relaxation length in boron nitride encapsulated bilayer graphene

    We have performed spin and charge transport measurements in dual gated high mobility bilayer graphene encapsulated in hexagonal boron nitride. Our results show spin relaxation lengths λs\lambda_s up to 13~\textmu m at room temperature with relaxation times τs\tau_s of 2.5~ns. At 4~K, the diffusion coefficient rises up to 0.52~m2^2/s, a value 5 times higher than the best achieved for graphene spin valves up to date. As a consequence, λs\lambda_s rises up to 24~\textmu m with τs\tau_s as high as 2.9~ns. We characterized 3 different samples and observed that the spin relaxation times increase with the device length. We explain our results using a model that accounts for the spin relaxation induced by the non-encapsulated outer regions.Comment: 5 pages and 4 figure

    Seismic stratigraphy of the Ontong Java Plateau

    The Ontong Java Plateau, a large, deep-water carbonate plateau in the western equatorial Pacific, is an ideal location for studying responses of carbonate sedimentation to the effects of changing paleoceanographic conditions. These carbonate responses are often reflected in the physical properties of the sediment, which in turn control the appearance of seismic reflection profiles. Seismic stratigraphy analyses, correlating eight reflector horizons to each drill site, have been conducted in an attempt to map stratigraphic data. Accurate correlation of seismic stratigraphic data to drilling results requires conversion of traveltime to depth in meters. Synthetic seismogram models, using shipboard physical properties data, have been generated in an attempt to provide this correlation. Physical properties, including laboratory-measured and well-log data, were collected from sites drilled during Deep Sea Drilling Project Legs 30 and 89, and Ocean Drilling Program Leg 130, on the top and flank of the Ontong Java Plateau. Laboratory-measured density is corrected to in-situ conditions by accounting for porosity rebound resulting from removal of the sediment from its overburden. The correction of laboratory-measured compressional velocity to in situ appears to be largely a function of increases in elastic moduli (especially shear rigidity) with depth of burial, more than a function of changes in temperature, pressure, or density (porosity rebound). Well-log velocity and density data for the ooze intervals were found to be greatly affected by drilling disturbance; hence, they were disregarded and replaced by lab data for these intervals. Velocity and density data were used to produce synthetic seismograms. Correlation of seismic reflection data with synthetic data, and hence with depth below seafloor, at each drill site shows that a single velocity-depth function exists for sediments on the top and flank of the Ontong Java Plateau. A polynomial fit of this function provides an equation for domain conversion: Depth (mbsf) = 44.49 + 0.800(traveltime[ms]) + 3.308 × 10 4 (traveltime[ms]2 ) Traveltime (ms) = -35.18 + 1.118(depth[mbsf]) - 1.969 × KT* (depth[mbsf]2 ) Seismic reflection profiles down the flank of the plateau undergo three significant changes: (1) a drastic thinning of the sediment column with depth, (2) changes in the echo-character of the profile (development of seismic facies), and (3) loss of continuous, coherent reflections. Sediments on the plateau top were largely deposited by pelagic processes, with little significant postdepositional or syndepositional modification. Sediments on the flank of the plateau are also pelagic, but they have been modified by faulting, erosion, and mass movement. These processes result in disrupted and incoherent reflectors, development of seismic facies, and redistribution of sediment on the flank of the plateau. Seismic stratigraphic analyses have shown that the sediment section decreases in thickness by as much as 65% between water depths of 2000 m water depth (at the top of the plateau) and 4000 m (near the base of the plateau). Thinning is attributed to increasing carbonate dissolution with depth. If this assumption is correct, then changes in the relative thicknesses of seismostratigraphic units at each drill site are indicative of changes in the position of the lysocline and the dissolution gradient between the lysocline and the carbonate compensation depth. We think that a shallow lysocline in the early Miocene caused sediment thinning. A deepening of the lysocline in the late-early Miocene caused relative thickening at each site. Within the middle Miocene, a sharp rise in lysoclinal depth occurs, concurrent with a steepening of the dissolution gradient. These events result in sediment thinning at all four sites. The thicker sections in the late Miocene likely correspond to a deepening of the lysocline, and a subsequent rise in the lysocline again hinders accumulation of sediment in the very late Miocene and Pliocene
