Identification of novel Coxiella burnetii genotypes from Ethiopian ticks
Background:
Coxiella burnetii
, the etiologic agent of Q fever, is a highly infectious
zoonotic bacterium. Genetic information about the strains of this worldwide
distributed agent circulating on the African continent is limited. The aim of the
present study was the genetic characterization of
C. burnetii
DNA samples
detected in ticks collected from Ethiopian cattle and their comparison with other
genotypes found previously in other parts of the world.
Methodology/Principal Findings:
A total of 296 tick samples were screened by
real-time PCR targeting the IS
1111
region of
C. burnetii
genome and from the 32
positive samples, 8 cases with sufficient
C. burnetii
DNA load (
Amblyomma
cohaerens
,n
5
6;
A. variegatum
,n
5
2) were characterized by multispacer sequence
typing (MST) and multiple-locus variable-number tandem repeat analysis (MLVA).
One novel sequence type (ST), the proposed ST52, was identified by MST. The
MLVA-6 discriminated the proposed ST52 into two newly identified MLVA
genotypes: type 24 or AH was detected in both
Amblyomma
species while type 26
or AI was found only in
A. cohaerens
.
Conclusions/Significance:
Both the MST and MLVA genotypes of the present
work are closely related to previously described genotypes found primarily in cattle
samples from different parts of the globe. This finding is congruent with the source
hosts of the analyzed Ethiopian ticks, as these were also collected from cattle. The
present study provides genotype information of
C. burnetii
from this seldom studied
East-African region as well as further evidence for the presumed host-specific
adaptation of this agent
Tropheryma whipplei, the Whipple's disease bacillus, induces macrophage apoptosis through the extrinsic pathway
Tropheryma whipplei, the etiological agent of Whipple's disease, is an intracellular bacterium that infects macrophages. We previously showed that infection of macrophages results in M2 polarization associated with induction of apoptosis and interleukin (IL)-16 secretion. In patients with Whipple's disease, circulating levels of apoptotic markers and IL-16 are increased and correlate with the activity of the disease. To gain insight into the understanding of the pathophysiology of this rare disease, we examined the molecular pathways involved in T. whipplei-induced apoptosis of human macrophages. Our data showed that apoptosis induction depended on bacterial viability and inhibition of bacterial protein synthesis reduced the apoptotic program elicited by T. whipplei. Induction of apoptosis was also associated with a massive degradation of both pro- and anti-apoptotic mediators. Caspase-specific inhibition experiments revealed that initiator caspases 8 and 10 were required for apoptosis, in contrast to caspases 2 and 9, in spite of cytochrome-c release from mitochondria. Finally, the effector caspases 3 and 6 were mandatory for apoptosis induction. Collectively, these data suggest that T. whipplei induces apoptosis through the extrinsic pathway and that, beside M2 polarization of macrophages, apoptosis induction contributes to bacterial replication and represents a virulence trait of this intracellular pathogen
Accelerated Partial Breast Irradiation (APBI): A review of available techniques
Breast conservation therapy (BCT) is the procedure of choice for the management of the early stage breast cancer. However, its utilization has not been maximized because of logistics issues associated with the protracted treatment involved with the radiation treatment. Accelerated Partial Breast Irradiation (APBI) is an approach that treats only the lumpectomy bed plus a 1-2 cm margin, rather than the whole breast. Hence because of the small volume of irradiation a higher dose can be delivered in a shorter period of time. There has been growing interest for APBI and various approaches have been developed under phase I-III clinical studies; these include multicatheter interstitial brachytherapy, balloon catheter brachytherapy, conformal external beam radiation therapy and intra-operative radiation therapy (IORT). Balloon-based brachytherapy approaches include Mammosite, Axxent electronic brachytherapy and Contura, Hybrid brachytherapy devices include SAVI and ClearPath. This paper reviews the different techniques, identifying the weaknesses and strength of each approach and proposes a direction for future research and development. It is evident that APBI will play a role in the management of a selected group of early breast cancer. However, the relative role of the different techniques is yet to be clearly identified
Intimate partner violence, trauma, and mental health need among female community college students
Objective: The impact of interpersonal violence on college students has received considerable attention, yet no studies have been conducted among community college students, who comprise 40% of all American college students, and have unique risk factors and needs. Community College students are more likely to be women, people of color, working, parenting, and first generation college students. Participants: Data were collected from a simple random sample from four community colleges (n=435). Methods: A cross-sectional quantitative survey was used to assess the extent of intimate partner violence, trauma exposure, sexual violence, and associated mental health consequences among female students. Results: Over 27% of participants reported IPV in the past year, while 25% reported sexual assault and 34% reported other uncomfortable sexual experiences in their lifetime. Nearly 20% of participants were currently reporting PTSD symptoms. Conclusions: Community Colleges should work with service providers to build their capacity to respond to students’ needs. Using deformable particles for single-particle measurements of velocity gradient tensors
Functional genomic analysis of an uncultured δ-proteobacterium in the sponge Cymbastela concentrica
Marine sponges are ancient, sessile, filter-feeding metazoans, which represent a significant component of the benthic communities throughout the world. Sponges harbor a remarkable diversity of bacteria, however, little is known about the functional properties of such bacterial symbionts. In this study, we present the genomic and functional characterization of an uncultured δ-proteobacterium associated with the sponge Cymbastela concentrica. We show that this organism represents a novel phylogenetic clade and propose that it lives in association with a cyanobacterium. We also provide an overview of the predicted functional and ecological properties of this δ-proteobacterium, and discuss its complex interactions with surrounding cells and milieu, including traits of cell attachment, nutrient transport and protein–protein interactions
