    Effects of C-terminal truncations of the histone acetyltransferase p300 on the growth and gene expression patterns of human diffuse large B-cell lymphoma cell lines

    Diffuse large B-cell lymphoma (DLBCL) is the most common form of non-Hodgkin’s B- cell lymphoma, accounting for about 30% of these lymphomas in the United States. Large-scale genome analyses of DLBCL have identified mutations in the related histone acetyltransferases (HATs) p300 and CBP in approximately 15% of patient samples and patient-derived cell lines. The research presented herein characterizes two human DLBCL cell lines, RC-K8 and SUDHL2, which express C-terminally truncated HAT domain-deficient p300 proteins, p300ΔC-1087 and p300 p300ΔC-820, respectively. It is shown that p300ΔC-820 localizes to sites of active transcription in the nucleus, interacts with NF-κB transcription factor REL, weakly enhances REL-dependent transactivation, and has a half-life similar to wild-type p300. Results demonstrate that knockdown of p300ΔC-820 in SUDHL2 cells reduces cell proliferation in vitro. In RC-K8 cells, p300ΔC-1087 suppresses expression of the NF-κB target genes A20 and IκBα, both of which are cytotoxic when overexpressed in RC-K8 cells. Microarray analysis of p300ΔC1087 knockdown compared to wild-type RC-K8 cells indicated that p300ΔC-1087 suppresses an NF-κB gene expression program and activates a MYC gene expression program in RC-K8 cells. Bioinformatic analysis demonstrated that cancer cell lines— regardless of tissue type—with truncating p300 mutations have altered expression of a MYC target gene set as compared to cancer cell lines with wild-type p300/CBP. Taken together, this research indicates that p300 truncations contribute to cell growth in DLBCL by modifying the transcriptional output of two lymphoid cell-specific oncoproteins, NF- κB and MYC, to optimal levels and suggests that p300 truncating mutations similarly modify the activity of oncogenic drivers in other cancer cell types. Based on this work, p300 truncation is proposed to represent a new class of oncogenic mutation that serves to optimize the activity of context-specific oncogenic transcription factors, and it is suggested that such oncogenic mutations be termed “cancer modifying” mutations.2017-09-30T00:00:00

    The purpose of this study is to optimize the manufacturing process of the SME bread product using Taguchi method. The study is focused on the quality problems occurred as the outcomes of the process (quality of the bun produced) related to controllable factors of products design identified (machine temperature and length duration time) for the improvement required. By implementing the technique of analysis of variance (ANOVA), the composition of the controlled parameters, such as machine temperatures and duration times, is therefore determined and constructed into Orthogonal Array (OA) of L9(33) related to what the setting parameters produces the optimal output. To improve the quality of the manufactured product, the setting of parameter recommended in this study is A2B2C2 or A1B2C2 {(Low or High) ∩ 10minutes ∩ 200OC)}

    This research discussed on the results obtained for each sample that have been conducted to the solid fuel briquettes made of empty fruit bunch fiber and waste papers from view of heat released during combustion test. The results shows tremendous amount of energy can be generated from these biomass compositions. The energy can easily harness to generate heat and electricity for many productive operations. Hence, the possibility result in this research is the development of solid fuel briquette by mixing the empty fruit bunch with a waste paper can be one sources of fuel energy. From the combustion analysis shows sample briquette with ratio 60:40 has given a highest value of heat released which is 162.77 kJ, whereas sample with ratio 40:60 has good burning time

    Analysis and development of recycled materials for wood plastic composite product

    This study is to find out the best optimum ratio of the mixed waste materials of high-density polyethylene and wood’s flour by comparing the mechanical properties of recycled product (WPC). In this study, the fabrication process of wood plastic composite (WPC) contained of virgin material, post-consumer high-density polyethylene, and wood’s flour is through a twin-screw extruder and hot compression machine. The WPC product with four different ratio filler content based on weight percentage, i.e. 0 wt %, 20 wt %, 30 wt % and 40 wt % , were tested using universal tensile machine and impact tester according to ASTM D 3039, ASTM D 790, and ASTM D 6110 . The results of experiment showed that tensile and flexural properties of the composite increased with the adding of the wood’s flour material. The testing showed, however, totally opposite to the result of the impact test. In overall, the results of observations showed that recycled WPC have better mechanical properties compared to non-recycled WPC

    A Comparision Analysis on Mechanical Properties between between Wax/LDPE Composite Versus Versus Coconut Coir/wax/LDPE Hybrid

    Natural fibers are, currently, as one of potential's reinforcing fiber source. One of their potential, especially which is related to mechanical reinforcement, can be found in coconut coir. By adding wax and low density polyethylene as reinforced matrix to coconut coir, in fact, will enhance the mechanical properties of the resulting hybrid composites. Therefore, in order to find out the optimum properties of coconut coir, this study is focused on how the mechanical characteristic of coconut coir and wax produced by inducing LDPE. In this study, by mixing the coconut coir, wax, and LDPE into four new polymer compositions, we found that the higher value obtained of the tensile strength, and hardness is by mixing between 8wt. % coconut coir with 2 wt. % wax hybrid, while for impact test LDPE/wax composites showed significantly higher impact. With two different methods applied, we carry out the study through hot plate magnetic stirrer for wax and LDPE mixing, while to coconut coir, wax, and LDPE with hot press. By examining the results related to mechanical properties value, we found that the most suitable mixtures of an optimal composition is obtained with 90 wt. % LDPE, 4 wt. % wax, and 6 wt. % coconut coir. To fulfil the purpose, the institution or organization need to have a good work safety and health management system. OHSAS 18001: 2007 is an international standard for the implementation of work safety and health management system. One of the clauses contained in OHSAS 18001: 2007 are hazard identification, risk assessment, and determining controls and these can be documented in the HIRADC (Hazard identification, risk assessment, and determining control) document. HIRADC is divided into three parts: Hazard Identification, Risk Assessment, and Determining Control. HIRADC, which is annually reviewed and updated by the management, discovers and controls the potential hazards that exist around the working environment.     Pengaruh Penyuluhan Seksualitas dengan Metode Stratagem Terhadap Pengetahuan Bahaya Seks Pranikah pada Remaja Awal

    Salah satu upaya yang mampu menerangkan pengetahuan mengenai bahaya seks pranikah pada remaja yakni melalui penyuluhan seksualitas. Remaja seringkali kekurangan informasi dasar mengenai bahaya seks pranikah. Penelitian ini bertujuan untuk mengetahui pengaruh seksualitas dengan metode stratagem terhadap pengetahuan bahaya seks pranikah pada remaja awal. Hasil Pengolahan data dengan menggunakan Uji Wilcoxon Test padakelompok intervensi didapatkan nilai pvalue = 0.000 atau p 0.05 yang berarti tidak terdapat perbedaan bermakna tingkat pengetahuan pre-test dan post-test tanpa pemberian penyuluhan seksualitas dengan metode stratagem. Jadi dapat disimpulkan bahwa ada pengaruh penyuluhan seksualitas dengan metode stratagem terhadap pengetahuan bahaya seks pranikah. Metode ini dapat digunakan sebagai teknik pembelajaran untuk menyampaikan pendidikan kesehatan khususnya penyuluhan seksualitas

    DESIGN AND ANALYSIS OF 5 KW SAVONIUS ROTOR BLADE

    This paper presents the design and analysis of the Savonius rotor blade to generate 5 kW power output. The relevant design parameters and theories were studied in this paper and used to determine related design geometry and requirements of the Savonius rotor blade. The Savonius rotor was designed with the rotor diameter of 3.5 m and the rotor height of 7 m. The 3D model of Savonius rotor blade was created by using SolidWorks software. Computational Fluid Dynamics (CFD) analysis and structural Finite Element Analysis (FEA) are presented in this paper. CFD analysis was performed to obtain the pressure difference between concave and convex region of the blade while FEA was done to obtain the structural response of the blade due to the wind load applied in term of stresses and its displacements
