    Aquest treball té com a objectiu reflexionar sobre la informació i la documentació dels Jocs Olímpics des de tres perspectives. En primer lloc, el treball presenta l'entorn informatiu dels organitzadors d'events, els Comitès organitzadors dels JJ.OO (els OCOG) i presenta l'exemple de de la gestió de la informació i documentació dins del Comitè organitzador dels JJ.OO. de Sydney, el SOCOG (Sydney Organising Committee for the Olympic Games). En la segona part del treball es destaca el paper de la organització principal en Comitè Olímpic Internacional (COI), com a creador, receptor i col·leccionista de la informació olímpica. L'últim apartat del treball està dedicat a la intersecció de fonts d'informació i documentació olímpica compilades per organitzacions i institucions externes al Moviment Olímpic, com per exemple estudiosos, bibliotecaris i Centres d'Estudis Olímpics. Aquests tres sectors en conjunt, tant dins com fora del Moviment Olímpic, conformen un òrgan complet del coneixement sobre els Jocs Olímpics.Este trabajo tiene como objetivo reflexionar sobre la información y la documentación de los Juegos Olímpicos desde tres perspectivas. Estos tres sectores en conjunto, tanto dentro como fuera del Movimiento Olímpico, conforman un órgano completo del conocimiento sobre los Juegos Olímpicos.This work aims to reflect on the information and documentation of the Olympic Games from three perspectives. In the first place, the work presents the informative environment of the event organizers, the organizing committees of the Olympic Games (the OCOG) and presents the example of the management of the information and documentation within the organizing committee Sydney Olympics, the SOCOG (Sydney Organizing Committee for the Olympic Games). In the second part of the paper, the role of the main organization in the International Olympic Committee (COI), as creator, recipient and collector of Olympic information, stands out. The last section of the work is devoted to the intersection of sources of information and Olympic documentation compiled by organizations and institutions external to the Olympic Movement, such as scholars, librarians and Olympic Studies Centers. These three sectors as a whole, both inside and outside of the Olympic Movement, make up a complete body of knowledge about the Olympic Games

    Beetroot was grown in an open soilless cultivation system with nutrition supplied by organic and inorganic sources. This low emission system was tested for microgardening high quality red beets with high water use efficiency and less pollutant emission in the environs. For this purpose, a pot experiment was planned according to completely randomized design. For inorganically grown red beets, peat moss was combined with 150, 200 and 250 ppm NH4NO3, whereas for organic red beets, peat moss was amended with compost having nitrogen equivalent to the mentioned NH4NO3 concentrations. Rosette and roots were analysed for fresh and dry biomass. Nitrate content, total soluble solids, titratable acidity, ripening index, ascorbic acid, betacyanins, flavonols and antioxidant capacity were assessed as beetroot quality attributes. Combination of peat moss with NH4NO3 showed comparatively lower fresh plant biomass, fresh and dry biomasses of rosette and root, and root to rosette ratio. However, enhanced antioxidant activity and bioaccumulation of ascorbic acid, total soluble solids, betacyanins, flavonols and reduced titratable acids, resulting in higher ripening index and good quality were observed in peat moss combined NH4NO3 treated beetroots. Overall, combination of peat moss with NH4NO3 led to higher nutritional and antioxidant quality of red beet plants.La remolacha se cultivó en un sistema de cultivo abierto sin suelo con nutrición suministrada por fuentes orgánicas e inorgánicas. Este sistema de baja emisión fue probado para la microhorticultura de remolachas rojas de alta calidad con una eficiencia alta de uso de agua y menor emisión de contaminantes a los alrededores. Con esta finalidad se planificó un experimento en macetas de acuerdo con un diseño completamente al azar. Para las remolachas rojas cultivadas inorgánicamente se combinó musgo de turbera con 150, 200 y 250 ppm de NH4NO3, mientras que para las orgánicas se adecuó el musgo de turbera con compost con un contenido de nitrógeno equivalente a las concentraciones de NH4NO3 mencionadas. Se analizó el contenido de biomasa fresca y seca de la roseta y de la raíz. El contenido de nitratos, los sólidos solubles totales, la acidez titulable, el índice de maduración, el ácido ascórbico, las betacianinas, los flavonoles y la capacidad antioxidante se evaluaron como atributos de calidad de la remolacha. La combinación de musgo de turbera con NH4NO3 mostró valores comparativamente más bajos de biomasa fresca de planta, de biomasa fresca y seca de roseta y raíz, y de relación raíz‑roseta. Sin embargo, también se observaron valores más altos de actividad antioxidante y bioacumulación de ácido ascórbico, sólidos solubles totales, betacianinas, flavonoles y menos ácidos titulables, lo que resultó en un mayor índice de maduración y buena calidad.     Possible solutions for replant problems caused by soil-borne pathogens in organic strawberry production

    Soil-borne pathogens, above all Verticillium sp. cause plant losses and yield decreases in many Austrian strawberry fields, also in organic farming systems; in a research project possibilities to resolve this problem were examined. In a field trial, which was started 1998, organically managed and differently fertilized plots (green compost, manure, hornchips) were compared to an unfertilised control plot and to two mineral fertilized and conventionally treated plots; the differences in field characteristics of strawberries in 2004 and 2005 were low, but differences in storage and biochemical characteristics could be seen in 2005. Some new cultivars tested as alternative to the very sensitive 'Elsanta' planted in 2005 on several farms showed a higher tolerance to diseases and good yield and fruit quality characteristics. Brassica sp. planted as an intercrop before strawberries reduced the amount of microsclerotia of Verticillium dahliae in highly infested soils, nevertheless the system has to be optimised and adapted to our climatic and husbandry conditions in further trials

    Hop proanthocyanidins for the fining of beer

    Fining agents are used in the clarification of beers; they help to reduce the time required to sediment suspended yeast cells and ensure the clarity and colloidal stability of beer. Following an adventitious observation during dry-hopping experiments, we identified a fining activity associated with Saaz hops. Extracts of hop cones were subsequently shown to have the capacity to flocculate yeast and result in their sedimentation. This activity has since been identified in extracts of many different hop varieties and, significantly in spent hops, the co-product resulting from commercial extraction of hops with either CO2 or ethanol. Here we illustrate the activity of the novel finings extracted from spent hops following CO2 extraction of Galena hops. The sediments formed on fining were compact, relative to those obtained when commercial isinglass was used to fine the same beers. The hop extracts were also effective in reducing 90° haze in beers under conditions designed to mimic both cask ale (12 °C) and lager (4 °C) type applications. The compounds responsible for the fining activity appear to be large (30–100 kDa, or more) polyphenols. Analysis of the polyphenols using colourimetric tests indicated the presence of proanthocyanidins. On acidic hydrolysis these generated cyanidin, which would be derived from a polymer composed of catechin and epicatechin subunits. The presence of these materials in spent hops offers the possibility to develop commercial products, with desirable fining properties, from an existing co-product stream. Furthermore, the finings are derived from a traditional ingredient of the brewing process

    Transgenic apple plants overexpressing the - gene of show increased levels of 3-hydroxyphloridzin and reduced susceptibility to apple scab and fire blight

    Main conclusion Overexpression of chalcone-3-hydroxylase provokes increased accumulation of 3-hydroxyphloridzin inMalus. Decreased flavonoid concentrations but unchanged flavonoid class composition were observed. The increased 3-hydroxyphlorizin contents correlate well with reduced susceptibility to fire blight and scab. The involvement of dihydrochalcones in the apple defence mechanism against pathogens is discussed but unknown biosynthetic steps in their formation hamper studies on their physiological relevance. The formation of 3-hydroxyphloretin is one of the gaps in the pathway. Polyphenol oxidases and cytochrome P450 dependent enzymes could be involved. Hydroxylation of phloretin in position 3 has high similarity to the B-ring hydroxylation of flavonoids catalysed by the well-known flavonoid 3′-hydroxylase (F3′H). Using recombinant F3′H and chalcone 3-hydroxylase (CH3H) from Cosmos sulphureus we show that F3′H and CH3H accept phloretin to some extent but higher conversion rates are obtained with CH3H. To test whether CH3H catalyzes the hydroxylation of dihydrochalcones in planta and if this could be of physiological relevance, we created transgenic apple trees harbouring CH3H from C. sulphureus. The three transgenic lines obtained showed lower polyphenol concentrations but no shift between the main polyphenol classes dihydrochalcones, flavonols, hydroxycinnamic acids and flavan 3-ols. Increase of 3-hydroxyphloridzin within the dihydrochalcones and of epicatechin/catechin within soluble flavan 3-ols were observed. Decreased activity of dihydroflavonol 4-reductase and chalcone synthase/chalcone isomerase could partially explain the lower polyphenol concentrations. In comparison to the parent line, the transgenic CH3H-lines showed a lower disease susceptibility to fire blight and apple scab that correlated with the increased 3-hydroxyphlorizin contents

    First genome edited poinsettias: targeted mutagenesis of flavonoid 3′-hydroxylase using CRISPR/Cas9 results in a colour shift

    The CRISPR/Cas9 system is a remarkably promising tool for targeted gene mutagenesis, and becoming ever more popular for modification of ornamental plants. In this study we performed the knockout of flavonoid 3′-hydroxylase (F3′H) with application of CRISPR/Cas9 in the red flowering poinsettia (Euphorbia pulcherrima) cultivar ‘Christmas Eve’, in order to obtain plants with orange bract colour, which accumulate prevalently pelargonidin. F3′H is an enzyme that is necessary for formation of cyanidin type anthocyanins, which are responsible for the red colour of poinsettia bracts. Even though F3′H was not completely inactivated, the bract colour of transgenic plants changed from vivid red (RHS 45B) to vivid reddish orange (RHS 33A), and cyanidin levels decreased significantly compared with the wild type. In the genetically modified plants, an increased ratio of pelargonidin to cyanidin was observed. By cloning and expression of mutated proteins, the lack of F3′H activity was confirmed. This confirms that a loss of function mutation in the poinsettia F3′H gene is sufficient for obtaining poinsettia with orange bract colour. This is the first report of successful use of CRISPR/Cas9 for genome editing in poinsettia

    Hybrid de novo transcriptome assembly of poinsettia (Euphorbia pulcherrima Willd. Ex Klotsch) bracts

    Background Poinsettia is a popular and important ornamental crop, mostly during the Christmas season. Its bract coloration ranges from pink/red to creamy/white shades. Despite its ornamental value, there is a lack of knowledge about the genetics and molecular biology of poinsettia, especially on the mechanisms of color formation. We performed an RNA-Seq analysis in order to shed light on the transcriptome of poinsettia bracts. Moreover, we analyzed the transcriptome differences of red- and white-bracted poinsettia varieties during bract development and coloration. For the assembly of a bract transcriptome, two paired-end cDNA libraries from a red and white poinsettia pair were sequenced with the Illumina technology, and one library from a red-bracted variety was used for PacBio sequencing. Both short and long reads were assembled using a hybrid de novo strategy. Samples of red- and white-bracted poinsettias were sequenced and comparatively analyzed in three color developmental stages in order to understand the mechanisms of color formation and accumulation in the species. Results The final transcriptome contains 288,524 contigs, with 33% showing confident protein annotation against the TAIR10 database. The BUSCO pipeline, which is based on near-universal orthologous gene groups, was applied to assess the transcriptome completeness. From a total of 1440 BUSCO groups searched, 77% were categorized as complete (41% as single-copy and 36% as duplicated), 10% as fragmented and 13% as missing BUSCOs. The gene expression comparison between red and white varieties of poinsettia showed a differential regulation of the flavonoid biosynthesis pathway only at particular stages of bract development. An initial impairment of the flavonoid pathway early in the color accumulation process for the white poinsettia variety was observed, but these differences were no longer present in the subsequent stages of bract development. Nonetheless, GSTF11 and UGT79B10 showed a lower expression in the last stage of bract development for the white variety and, therefore, are potential candidates for further studies on poinsettia coloration. Conclusions In summary, this transcriptome analysis provides a valuable foundation for further studies on poinsettia, such as plant breeding and genetics, and highlights crucial information on the molecular mechanism of color formation
