    Dasein, The Early Years: Heideggerian Reflections on Childhood

    Like most philosophers, Heidegger gave little attention to childhood, but his philosophical emphasis on pre-reflective practice and understanding seems uniquely qualified to help make sense of a child’s experience and development. Moreover, it seems to me that many central Heideggerian concepts are best defended, exemplified, and articulated by bringing child development into the discussion. A Heideggerain emphasis on pre-theoretical world-involvement opens up a rich array of phenomena for studying child development, which can improve upon standard theories that have over-emphasized exclusive conditions or criteria. I begin by laying out some basic features of Heidegger’s conception of being-in-the-world as a preparation for understanding the world of the child. Then I will briefly discuss some of Heidegger’s remarks on childhood, followed by some reflections on language acquisition and the correlation of anxiety and meaning

    GFP : green fluorescent protein a versatile gene marker for entomopathogenic nematodes

    L'expression des gènes et la répartition des protéines dans les cellules peuvent être révélées à l'aide de gènes rapporteurs, tels que la bêta-galactosidase, la luciférase du vert luisant et la luciférase bactérienne. Ces systèmes rapporteurs nécessitent des substrats et des cofacteurs exogènes qui tuent les cellules ou les organismes. Le gène de la protéine fluorescente verte (gfp) de la méduse #Aequoria victoria est utilisé comme marqueur de l'expression de gènes chez #Caenorhabditis elegans. Les substrats ou cofacteurs exogènes n'étant pas nécessaires, la gfp peut servir à suivre l'expression de gènes dans les organismes vivants. Cependant l'autofluorescence naturelle des nématodes peut gêner l'utilisation de ces marqueurs. Nous avons examiné l'autofluorescence chez les nématodes entomophages pour établir l'utilité de la gfp comme marqueur de l'expression de gènes. Des espèces d'#Heterorhabditis et de #Steinernema de tous stades montraient une autofluorescence allant du vert au jaune pâle, son intensité s'accroissant avec l'âge des nématodes. La gfp utilisée a été sous le contrôle du promoteur mec-4 de #C. elegans. Le plasmide (pGFP/mec-4) a été injecté par microinjection dans la gonade de #Heterorhabditis bacteriophora. La gfp a été exprimée dans #H. bacteriophora et a pu être facilement repérée dans la région caudale du nématode. L'expression par mec-4 indique la présence de neurones récepteurs tactiles chez #H. bacteriophora au même endroit que chez #C. elegans$.     Comparative Study of the Use of Insect Meal from Spodoptera littoralis and Bactrocera zonata for Feeding Japanese Quail Chicks

    A transformation of current livestock production towards a more sustainable operation is crucial to face nutritional and environmental challenges. There is an urgent demand for more sustainable high-quality feed sources to reduce environmental costs. Insects pose a potential alternative since they can be reared sustainably on food and feed residues. Know-how in mass rearing already exists for insect species used in biological pest control, such as the African cotton leafworm Spodoptera littoralis and the peach fruit fly Bactrocera zonata. The impact of a replacement of 50% of soybean meal by S. littoralis and B. zonata meal, respectively, on seven-days-old Japanese quail chicks was investigated in feeding trials. Concomitantly, the chemical compositions of the two insect meals and soybean meal were determined and compared. It was observed that the insect meals had higher protein and fat contents, lower carbohydrate contents and contained more saturated fatty acids than soybean meal. They also had higher methionine, and S. littoralis had a higher lysine content. Feeding trials resulted in improved growth, feed performance parameters, carcass characteristics, and biochemical indices for both insect meals.     Evaluation of the Antimicrobial Activity of Endophytic Bacterial Populations From Chinese Traditional Medicinal Plant Licorice and Characterization of the Bioactive Secondary Metabolites Produced by Bacillus atrophaeus Against Verticillium dahliae

    Endophytic bacteria associated with medicinal plants possess unique strategies that enhance growth and suvival of host plants, many of which are mediated by distinctive secondary metabolites. These bacteria and their secondary metabolites are important subjects for both basic and applied research aimed at sustainable agriculture. In the present study, 114 endophytic strains isolated from the wild ethnomedicinal plant Glycyrrhiza uralensis (licorice) were screened for their in vitro antimicrobial activities against common fungal pathogens of tomato (Fusarium oxysporum f. sp., Fulvia fulva, Alternaria solani), cotton (Fusarium oxysporum f. sp. Vesinfectum, Verticillium dahliae), pomegranite (Ceratocystis fimbriata), Cymbidinium (Colletotrichum gloeosporioides), and Tsao-ko (Pestalotiopsis microspora and Fusarium graminearum) and the common bacteria Staphylococcus aureus, Bacillus cereus, Salmonella enteritidis, and Escherichia coli. Several Bacillus strains, particularly Bacillus atrophaeus and Bacillus mojavensis, had a broad spectrum of antifungal and antibacterial activity. A total of 16 strains, selected based on broad antimicrobial activity, were shown to contain at least one putative secondary metabolite-encoding gene (i.e., polyketide synthase or non-ribosomal peptide synthetase) and/or one lytic enzyme (i.e., protease, cellulase, lipase, chitinase), which may be important mediators of antagonistic activity against pathogens. Five strains, representing Bacillus atrophaeus and Bacillus mojavensis, were selected for plant growth chamber experiments based on strong in vitro antifungal activities. All five strains significantly reduced disease severity in Arabidopsis thaliana plants challenged with V. dahlia infection. Gas-chromatography/mass-spectrometry analysis of cell-free extracts of Bacillus atrophaeus strain XEGI50 showed that at least 13 compounds were produced only during co-cultivation with V. dahlia, including putative compounds known to have antimicrobial activity, such as 1,2-benzenedicarboxylic acid, bis (2-methylpropyl) ester; 9,12-octadecadienoic acid (Z,Z)-, methyl ester; 9-octadecenoic acid, methyl ester, (E)-; and decanedioic acid, bis(2-ethylhexyl) ester. To our knowledge, this study is the first to report that bacteria isolated from G. uralensis have biocontrol abilities. Our findings provide new insights into the antimicrobial activities of natural endophytes, particularly B. atrophaeus, and suggest this species may a promising candidate as a biocontrol agent to confer resistance to Verticillium wilt disease and other phytopathogens in cotton and other crops
