    124I-HuCC49deltaCH2 for TAG-72 antigen-directed positron emission tomography (PET) imaging of LS174T colon adenocarcinoma tumor implants in xenograft mice: preliminary results

    <p>Abstract</p> <p>Background</p> <p><sup>18</sup>F-fluorodeoxyglucose positron emission tomography (<sup>18</sup>F-FDG-PET) is widely used in diagnostic cancer imaging. However, the use of <sup>18</sup>F-FDG in PET-based imaging is limited by its specificity and sensitivity. In contrast, anti-TAG (tumor associated glycoprotein)-72 monoclonal antibodies are highly specific for binding to a variety of adenocarcinomas, including colorectal cancer. The aim of this preliminary study was to evaluate a complimentary determining region (CDR)-grafted humanized C<sub>H</sub>2-domain-deleted anti-TAG-72 monoclonal antibody (HuCC49deltaC<sub>H</sub>2), radiolabeled with iodine-124 (<sup>124</sup>I), as an antigen-directed and cancer-specific targeting agent for PET-based imaging.</p> <p>Methods</p> <p>HuCC49deltaC<sub>H</sub>2 was radiolabeled with <sup>124</sup>I. Subcutaneous tumor implants of LS174T colon adenocarcinoma cells, which express TAG-72 antigen, were grown on athymic Nu/Nu nude mice as the xenograft model. Intravascular (i.v.) and intraperitoneal (i.p.) administration of <sup>124</sup>I-HuCC49deltaC<sub>H</sub>2 was then evaluated in this xenograft mouse model at various time points from approximately 1 hour to 24 hours after injection using microPET imaging. This was compared to i.v. injection of <sup>18</sup>F-FDG in the same xenograft mouse model using microPET imaging at 50 minutes after injection.</p> <p>Results</p> <p>At approximately 1 hour after i.v. injection, <sup>124</sup>I-HuCC49deltaC<sub>H</sub>2 was distributed within the systemic circulation, while at approximately 1 hour after i.p. injection, <sup>124</sup>I-HuCC49deltaC<sub>H</sub>2 was distributed within the peritoneal cavity. At time points from 18 hours to 24 hours after i.v. and i.p. injection, <sup>124</sup>I-HuCC49deltaC<sub>H</sub>2 demonstrated a significantly increased level of specific localization to LS174T tumor implants (p = 0.001) when compared to the 1 hour images. In contrast, approximately 50 minutes after i.v. injection, <sup>18</sup>F-FDG failed to demonstrate any increased level of specific localization to a LS174T tumor implant, but showed the propensity toward more nonspecific uptake within the heart, Harderian glands of the bony orbits of the eyes, brown fat of the posterior neck, kidneys, and bladder.</p> <p>Conclusions</p> <p>On microPET imaging, <sup>124</sup>I-HuCC49deltaC<sub>H</sub>2 demonstrates an increased level of specific localization to tumor implants of LS174T colon adenocarcinoma cells in the xenograft mouse model on delayed imaging, while <sup>18</sup>F-FDG failed to demonstrate this. The antigen-directed and cancer-specific <sup>124</sup>I-radiolabled anti-TAG-72 monoclonal antibody conjugate, <sup>124</sup>I-HuCC49deltaC<sub>H</sub>2, holds future potential for use in human clinical trials for preoperative, intraoperative, and postoperative PET-based imaging strategies, including fused-modality PET-based imaging platforms.</p

    Evaluating the evidence for models of life course socioeconomic factors and cardiovascular outcomes: a systematic review

    BACKGROUND: A relatively consistent body of research supports an inverse graded relationship between socioeconomic status (SES) and cardiovascular disease (CVD). More recently, researchers have proposed various life course SES hypotheses, which posit that the combination, accumulation, and/or interactions of different environments and experiences throughout life can affect adult risk of CVD. Different life course designs have been utilized to examine the impact of SES throughout the life course. This systematic review describes the four most common life course hypotheses, categorizes the studies that have examined the associations between life course SES and CVD according to their life course design, discusses the strengths and weaknesses of the different designs, and summarizes the studies' findings. METHODS: This research reviewed 49 observational studies in the biomedical literature that included socioeconomic measures at a time other than adulthood as independent variables, and assessed subclinical CHD, incident CVD morbidity and/or mortality, and/or the prevalence of traditional CVD risk factors as their outcomes. Studies were categorized into four groups based upon life course design and analytic approach. The study authors' conclusions and statistical tests were considered in summarizing study results. RESULTS: Study results suggest that low SES throughout the life course modestly impacts CVD risk factors and CVD risk. Specifically, studies reviewed provided moderate support for the role of low early-life SES and elevated levels of CVD risk factors and CVD morbidity and mortality, little support for a unique influence of social mobility on CVD, and consistent support for the detrimental impact of the accumulation of negative SES experiences/conditions across the life course on CVD risk. CONCLUSIONS: While the basic life course SES study designs have various methodologic and conceptual limitations, they provide an important approach from which to examine the influence of social factors on CVD development. Some limitations may be addressed through the analysis of study cohorts followed from childhood, the evaluation of CVD risk factors in early and middle adulthood, and the use of multiple SES measures and multiple life course analysis approaches in each life course study

Observations made about the behavior of the drug, in the study sample, are then generalized to the population from which the sample was drawn. However, the magnitude and pharmacodynamic qualities of the response to many central nervous system-active drugs are known to vary in the population.     A comprehensive overview of radioguided surgery using gamma detection probe technology

    The concept of radioguided surgery, which was first developed some 60 years ago, involves the use of a radiation detection probe system for the intraoperative detection of radionuclides. The use of gamma detection probe technology in radioguided surgery has tremendously expanded and has evolved into what is now considered an established discipline within the practice of surgery, revolutionizing the surgical management of many malignancies, including breast cancer, melanoma, and colorectal cancer, as well as the surgical management of parathyroid disease. The impact of radioguided surgery on the surgical management of cancer patients includes providing vital and real-time information to the surgeon regarding the location and extent of disease, as well as regarding the assessment of surgical resection margins. Additionally, it has allowed the surgeon to minimize the surgical invasiveness of many diagnostic and therapeutic procedures, while still maintaining maximum benefit to the cancer patient.     Role of macronutrients in cotton production

    Sound nutrition plays a key role in enhancing cotton yield. As cotton undergoes vegetative and reproductive growth at the same time, its nutritional requirements are dissimilar, compared to other field crops. Cotton is grown as an annual crop with an indeterminate growth pattern. The vegetative branching provides a potential fruiting place except under abiotic and biotic stresses. Moreover, cotton has a deep root system with low density of roots in the surface layer of soils where availability of nutrients is high. The rooting system makes cotton crop more dependent on the subsoil for nutrition. A continuous supply of nutrients is required to sustain morphogenesis. The rate of both nutrients absorption and dry matter production increases progressively during the seedling, vegetative, and fruiting periods and peaks near the end of the bloom period. Nitrogen, phosphorus, and potassium are required in large quantities and are limited in many soils. The deficiencies of macro-and micronutrients decrease plant growth and development, and consequently seed cotton yield is reduced. The deficiency of phosphorous (P), calcium (Ca), potassium (K), boron (B), magnesium (Mg), and zinc (Zn) affects fruit production in cotton than vegetative growth, while the deficiencies of nitrogen (N), sulfur (S), molybdenum (Mo), and manganese (Mn) affect equally vegetative and reproductive growth of cotton.     User engagement in online and social networks has been studied widely over the last decade. The recent rise in popularity of social networks, especially social media and social streaming services has increased the amount of academic research and literature, especially on those subjects specifically but not concerning the whole social networks field. This thesis aims to shed light to the principles companies and application designers could use to utilize the principles of persuasive technology and behavioural economics, combined with other fields presented, to increase user engagement and forming habits within social network applications. As the amount of social network applications rise increasingly, companies and application designers rely deeply on ongoing customer/user relationships and how to attract more users to be profitable. Understanding what drives user engagement and how to form user habits is paramount for success in the field of social networks. This thesis was conducted through a multidisciplinary literature review by combining previous literature and research with insights from other affecting fields and through that, this thesis is able to present principles, theoretical views and models necessary to answer the research question. The results and gathered information of this thesis give an overview of the principles and models used in the field of social networks to increase user engagement and habit-forming functionalities in social network applications. Furthermore, these findings presented in this literary review prove that the principles and models presented here can provide a framework of thought for planning and designing features to increase user engagement and form habits within users. Examples such as Instagram, the world’s fourth most used social network application, are presented how utilizing these principles in the planning process, it has had success in forming habits with users and increase in user engagement.
