Cure of murine leishmaniasis with anti-interleukin 4 monoclonal antibody. Evidence for a T cell-dependent, interferon gamma-independent mechanism.
BALB/c mice infected with Leishmania major develop fatal, progressive disease, despite an immune response characterized by expansion of CD4+ T cells in the draining lymph nodes. The immune response has been further characterized by a lack of IFN-gamma mRNA, but increased IL-4 mRNA in lymphoid tissues, and striking elevation of serum IgE. Treatment of infected BALB/c mice with rIFN-gamma at doses shown to be beneficial in other protozoan infections was insufficient to ameliorate L. major infection. In contrast, neutralization of IL-4 by six weekly injections of mAb 11B11 led to attenuation of disease in 100% of animals, and complete cure in 85%. Resolution of disease required the presence of T cells, and recovered mice remained resistant to reinfection at 12 wk. This immunity was adoptively transferable and was dependent on both CD4+ and CD8+ cells. Although administration of anti-IL-4 was associated with fourfold increase in IFN-gamma mRNA in lymph node cells draining the lesion, the coadministration of neutralizing R4 6A2 anti-IFN-gamma mAb had no effect on resistance to disease. This was in marked contrast to resolution of disease in both resistant C57BL/6- and GK1.5-pretreated BALB/c mice that was abrogated by in vivo treatment with anti-IFN-gamma. These data suggest a novel mechanism of cellular immunity established by interference with the development of Th2 cells during infection
Reciprocal expression of interferon gamma or interleukin 4 during the resolution or progression of murine leishmaniasis. Evidence for expansion of distinct helper T cell subsets.
We purified poly(A)+ mRNA from the spleen and lymph nodes at designated times after infection with Leishmania major in genetically susceptible BALB/c and resistant C57BL/6 mice. The steady-state levels of IL-2, IFN-gamma, IL-4, and IL-1 beta mRNA were determined using Northern hybridizations. IL-2 mRNA levels in the infected organs of BALB/c and C57BL/6 mice were comparable after infection, but IFN-gamma and IL-4 mRNA levels were reciprocally expressed. Levels of IFN-gamma mRNA in C57BL/6 draining nodes and spleen were significantly greater than in BALB/c mice except at 4 and 6 wk of infection, when splenic IFN-gamma mRNA levels were transiently comparable. In contrast, IL-4 mRNA was apparent only in BALB/c and not in C57BL/6 nodes and spleen. Tissue levels of IL-1 beta mRNA were 10-20-fold greater in BALB/c mice. BALB/c mice were pretreated with GK1.5 mAb, a manipulation that promotes healing of subsequent infection by transiently depleting L3T4+ cells. At 8 wk of infection, by which time lymphoid organs were repopulated with L3T4+ cells, GK1.5-pretreated BALB/c mice produced IFN-gamma, but not IL-4 message. Serum levels of IgE were markedly elevated in infected BALB/c, but not in infected C57BL/6 or GK1.5-pretreated BALB/c mice, consistent with in vivo biologic activity of IL-4 in nonhealing mice. Treatment of infected BALB/c mice with neutralizing anti-IL-4 antibody abolished the elevation of serum IgE and significantly attenuated the progression of disease as assessed by size and ulceration of the lesion, and by reduction in the number of tissue parasites. Both protective and deleterious responses to Leishmania infection have previously been shown to be L3T4+ cell dependent. Our findings are consistent with the differential expansion of protective, IFN-gamma-producing Th1 cells in healing mice, and the expansion of deleterious, IL-4-producing Th2 cells in nonhealing mice. The inverse relationship of IFN-gamma and IL-4 gene expression during leishmaniasis may underlie the divergence of cellular and humoral immunity that occurs during chronic infection with Leishmania and possibly other intracellular parasites
Phenotypic landscape inference reveals multiple evolutionary paths to C photosynthesis
C photosynthesis has independently evolved from the ancestral C
pathway in at least 60 plant lineages, but, as with other complex traits, how
it evolved is unclear. Here we show that the polyphyletic appearance of C
photosynthesis is associated with diverse and flexible evolutionary paths that
group into four major trajectories. We conducted a meta-analysis of 18 lineages
containing species that use C, C, or intermediate C-C forms of
photosynthesis to parameterise a 16-dimensional phenotypic landscape. We then
developed and experimentally verified a novel Bayesian approach based on a
hidden Markov model that predicts how the C phenotype evolved. The
alternative evolutionary histories underlying the appearance of C
photosynthesis were determined by ancestral lineage and initial phenotypic
alterations unrelated to photosynthesis. We conclude that the order of C
trait acquisition is flexible and driven by non-photosynthetic drivers. This
flexibility will have facilitated the convergent evolution of this complex
trait
Learning histology – dental and medical students' study strategies
PurposeHistology, the science of cells and tissues at the microscopic level, is an integral component of most dental and medical curricula and is often taught using both traditional and novel computer‐based didactic approaches. The purpose of this study was to analyse the strategies used by dental and medical students when studying this very visual and challenging subject.MethodsData were collected from 75 dental and 143 medical students, who had almost identical histology learning resources at their disposal.ResultsWhen compared with their medical counterparts, dental students view histology as a more difficult subject and as less relevant for their future career. Whereas dental students, who are required to attend class unlike medical students, made more use of in‐classroom learning opportunities, they did not take as much advantage of out‐of‐classroom resources. In addition, dental students reported a significantly higher tendency than medical students to work together, rather than to study alone.DiscussionSmall differences in the dental versus the medical learning environment associate with several observed differences in learning strategies that are adopted by dental and medical students.ConclusionsThese differences should be considered when teaching the subject of histology to dental or to medical students.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/111121/1/eje12104.pd
THE LEADERSHIP DEVELOPMENT OF FRATERNITY AND SORORITY LIFE MEMBERS
This study examines the relationship of perceived leadership development opportunities in fraternities and sororities. The study\u27s main goal was to determine if there was a discrepancy in perceived leadership development opportunities in general members, versus executive board members, whether there was a difference in findings for fraternity members versus sorority members, and suggestions on how to improve opportunities on a university level for students in Fraternity and Sorority Life going forward. The study was distributed to all social Fraternity and Sorority members at Rowan University that were in their respective organization for at least one year prior to the survey. The findings suggest that perceived opportunities are on average high and both fraternity and sorority members reported close to or exceeded the mean. Opportunities for executive board members were also perceived to be slightly higher for sorority members versus their general membership, while fraternity members reported more of equal opportunity
Carbon partitioning and export in transgenic Arabidopsis thaliana with altered capacity for sucrose synthesis grown at low temperature: a role for metabolite transporters
We investigated the role of metabolite transporters in cold acclimation by comparing the responses of wild-type (WT) Arabidopsis thaliana (Heynh.) with that of transgenic plants over-expressing sucrose-phosphate synthase (SPSox) or with that of antisense repression of cytosolic fructose-1,6-bisphosphatase (FBPas). Plants were grown at 23 degrees C and then shifted to 5 degrees C. We compared the leaves shifted to 5 degrees C for 3 and 10 d with new leaves that developed at 5 degrees C with control leaves on plants at 23 degrees C. At 23 degrees C, ectopic expression of SPS resulted in 30% more carbon being fixed per day and an increase in sucrose export from source leaves. This increase in fixation and export was supported by increased expression of the plastidic triose-phosphate transporter AtTPT and, to a lesser extent, the high-affinity Suc transporter AtSUC1. The improved photosynthetic performance of the SPSox plants was maintained after they were shifted to 5 degrees C and this was associated with further increases in AtSUC1 expression but with a strong repression of AtTPT mRNA abundance. Similar responses were shown by WT plants during acclimation to low temperature and this response was attenuated in the low sucrose producing FBPas plants. These data suggest that a key element in recovering flux through carbohydrate metabolism in the cold is to control the partitioning of metabolites between the chloroplast and the cytosol, and Arabidopsis modulates the expression of AtTPT to maintain balanced carbon flow. Arabidopsis also up-regulates the expression of AtSUC1, and to lesser extent AtSUC2, as down-stream components facilitate sucrose transport in leaves that develop at low temperatures.info:eu-repo/semantics/publishedVersio
