Cloning and expression of Phanerochaete chrysosporium cellobiohydrolase (cbhI.2)
The Phanerochaete chrysosporium ME446 cbhI.2 cDNA was cloned and expressed in Escherichia coli using a pET system. Pulse-labelling revealed the expression of an induced protein of approximately 50- 60 kDa that reacted with anti-P. chrysosporium ME446 CBH antibodies. The expressed protein remained undegraded in vivo for 135 min but was biologically inactive in vitro
Refolding and characterisation of a heterologous expressed Phanerochaete chrysosporium cellobiohydrolase (CBHI.2)
Cloned Phanerochaete chrysosporium ME446 cbhI.2 cDNA was successfully expressed in Escherichia coli as an insoluble, internal, biologically inactive protein. In vitro chemical refolding restored the activity of the crude CBHI.2. However, this enzyme was active against 4-methylumbelliferyl-ß-Dcellobioside(MUC) and 4-methyllumbelliferyl-ß-D-lactopyranoside (MUL) substrates only. The crude enzyme lost almost 50% of its activity at 5 min at 100oC heat treatment whereas total inactivation was achieved at 30 min
Skin Lesion Analyser: An Efficient Seven-Way Multi-Class Skin Cancer Classification Using MobileNet
Skin cancer, a major form of cancer, is a critical public health problem with
123,000 newly diagnosed melanoma cases and between 2 and 3 million non-melanoma
cases worldwide each year. The leading cause of skin cancer is high exposure of
skin cells to UV radiation, which can damage the DNA inside skin cells leading
to uncontrolled growth of skin cells. Skin cancer is primarily diagnosed
visually employing clinical screening, a biopsy, dermoscopic analysis, and
histopathological examination. It has been demonstrated that the dermoscopic
analysis in the hands of inexperienced dermatologists may cause a reduction in
diagnostic accuracy. Early detection and screening of skin cancer have the
potential to reduce mortality and morbidity. Previous studies have shown Deep
Learning ability to perform better than human experts in several visual
recognition tasks. In this paper, we propose an efficient seven-way automated
multi-class skin cancer classification system having performance comparable
with expert dermatologists. We used a pretrained MobileNet model to train over
HAM10000 dataset using transfer learning. The model classifies skin lesion
image with a categorical accuracy of 83.1 percent, top2 accuracy of 91.36
percent and top3 accuracy of 95.34 percent. The weighted average of precision,
recall, and f1-score were found to be 0.89, 0.83, and 0.83 respectively. The
model has been deployed as a web application for public use at
(https://saketchaturvedi.github.io). This fast, expansible method holds the
potential for substantial clinical impact, including broadening the scope of
primary care practice and augmenting clinical decision-making for dermatology

Health literacy, health status, and healthcare utilization of Taiwanese adults: results from a national survey
Abstract Background Low health literacy is considered a worldwide health threat. The purpose of this study is to assess the prevalence and socio-demographic covariates of low health literacy in Taiwanese adults and to investigate the relationships between health literacy and health status and health care utilization. Methods A national survey of 1493 adults was conducted in 2008. Health literacy was measured using the Mandarin Health Literacy Scale. Health status was measured based on self-rated physical and mental health. Health care utilization was measured based on self-reported outpatient clinic visits, emergency room visits, and hospitalizations. Results Approximately thirty percent of adults were found to have low (inadequate or marginal) health literacy. They tended to be older, have fewer years of schooling, lower household income, and reside in less populated areas. Inadequate health literacy was associated with poorer mental health (OR, 0.57; 95% CI, 0.35-0.91). No association was found between health literacy and health care utilization even after adjusting for other covariates. Conclusions Low (inadequate and marginal) health literacy is prevalent in Taiwan. High prevalence of low health literacy is not necessarily indicative of the need for interventions. Rethinking feasibility analysis for urban development: a multidimensional decision support tool
Large-scale urban development projects featured over the past thirty years have shown some critical issues related to the implementation phase. Con-sequently, the current practice seems oriented toward minimal and wide-spread interventions meant as urban catalyst. This planning practice might solve the problem of limited reliability of large developments’ feasibility studies, but it rises an evaluation demand related to the selection of coali-tion of projects within a multidimensional and multi-stakeholders deci-sion-making context. This study aims to propose a framework for the generation of coalitions of elementary actions in the context of urban regeneration processes and for their evaluation using a Multi Criteria Decision Analysis approach. The proposed evaluation framework supports decision makers in exploring dif-ferent combinations of actions in the context of urban interventions taking into account synergies, i.e. positive or negative effects on the overall per-formance of an alternative linked to the joint realization of specific pairs of actions. The proposed evaluation framework has been tested on a pilot case study dealing with urban regeneration processes in the city of Milan (Italy)
Transiting extrasolar planetary candidates in the Galactic bulge
More than 200 extrasolar planets have been discovered around relatively
nearby stars, primarily through the Doppler line shifts owing to the reflex
motions of their host stars, and more recently through transits of some planets
across the face of the host stars. The detection of planets with the shortest
known periods, 1.2 to 2.5 days, has mainly resulted from transit surveys which
have generally targeted stars more massive than 0.75 M_sun. Here we report the
results from a planetary transit search performed in a rich stellar field
towards the Galactic bulge. We discovered 16 candidates with orbital periods
between 0.4 and 4.2 days, five of which orbit stars of 0.44 to 0.75 M_sun. In
two cases, radial-velocity measurements support the planetary nature of the
companions. Five candidates have orbital periods below 1.0 day, constituting a
new class of ultra-short-period planets (USPPs), which occur only around stars
of less than 0.88 M_sun. This indicates that those orbiting very close to more
luminous stars might be evaporatively destroyed, or that jovian planets around
lower-mass stars might migrate to smaller radii.Comment: To appear in October 5, 2006 issue of Natur

Blood pressure variability and cardiovascular risk in the PROspective study of pravastatin in the elderly at risk (PROSPER)
Variability in blood pressure predicts cardiovascular disease in young- and middle-aged subjects, but relevant data for older individuals are sparse. We analysed data from the PROspective Study of Pravastatin in the Elderly at Risk (PROSPER) study of 5804 participants aged 70–82 years with a history of, or risk factors for cardiovascular disease. Visit-to-visit variability in blood pressure (standard deviation) was determined using a minimum of five measurements over 1 year; an inception cohort of 4819 subjects had subsequent in-trial 3 years follow-up; longer-term follow-up (mean 7.1 years) was available for 1808 subjects. Higher systolic blood pressure variability independently predicted long-term follow-up vascular and total mortality (hazard ratio per 5 mmHg increase in standard deviation of systolic blood pressure = 1.2, 95% confidence interval 1.1–1.4; hazard ratio 1.1, 95% confidence interval 1.1–1.2, respectively). Variability in diastolic blood pressure associated with increased risk for coronary events (hazard ratio 1.5, 95% confidence interval 1.2–1.8 for each 5 mmHg increase), heart failure hospitalisation (hazard ratio 1.4, 95% confidence interval 1.1–1.8) and vascular (hazard ratio 1.4, 95% confidence interval 1.1–1.7) and total mortality (hazard ratio 1.3, 95% confidence interval 1.1–1.5), all in long-term follow-up. Pulse pressure variability was associated with increased stroke risk (hazard ratio 1.2, 95% confidence interval 1.0–1.4 for each 5 mmHg increase), vascular mortality (hazard ratio 1.2, 95% confidence interval 1.0–1.3) and total mortality (hazard ratio 1.1, 95% confidence interval 1.0–1.2), all in long-term follow-up. All associations were independent of respective mean blood pressure levels, age, gender, in-trial treatment group (pravastatin or placebo) and prior vascular disease and cardiovascular disease risk factors. Our observations suggest variability in diastolic blood pressure is more strongly associated with vascular or total mortality than is systolic pressure variability in older high-risk subjects
The development of path integration: combining estimations of distance and heading
Efficient daily navigation is underpinned by path integration, the mechanism by which we use self-movement information to update our position in space. This process is well-understood in adulthood, but there has been relatively little study of path integration in childhood, leading to an underrepresentation in accounts of navigational development. Previous research has shown that calculation of distance and heading both tend to be less accurate in children as they are in adults, although there have been no studies of the combined calculation of distance and heading that typifies naturalistic path integration. In the present study 5-year-olds and 7-year-olds took part in a triangle-completion task, where they were required to return to the startpoint of a multi-element path using only idiothetic information. Performance was compared to a sample of adult participants, who were found to be more accurate than children on measures of landing error, heading error, and distance error. 7-year-olds were significantly more accurate than 5-year-olds on measures of landing error and heading error, although the difference between groups was much smaller for distance error. All measures were reliably correlated with age, demonstrating a clear development of path integration abilities within the age range tested. Taken together, these data make a strong case for the inclusion of path integration within developmental models of spatial navigational processing
