    A multi-purpose modular system for high-resolution microscopy at high hydrostatic pressure

    We have developed a modular system for high-resolution microscopy at high hydrostatic pressure. The system consists of a pressurised cell of volume ~100 microlitres, a temperature controlled holder, a ram and a piston. We have made each of these components in several versions which can be interchanged to allow a wide range of applications. Here, we report two pressure cells with pressure ranges 0.1-700MPa and 0.1-100MPa, which can be combined with hollow or solid rams and pistons. Our system is designed to work with fluorescent samples (using a confocal or epifluorescence microscope), but also allows for transmitted light microscopy via the hollow ram and piston. The system allows precise control of pressure and temperature [-20-70C], as well as rapid pressure quenching. We demonstrate its performance and versatility with two applications: time-resolved imaging of colloidal phase transitions caused by pressure changes between 0.1MPa and 101MPa, and imaging the growth of Escherichia coli bacteria at 50MPa. We also show that the isotropic-nematic phase transition of pentyl-cyanobiphenyl (5CB) liquid crystal provides a simple, convenient and accurate method for calibrating pressure in the range 0.1-200MPa

    BL Lac X-ray Spectra: simpler than we thought

    We report results from {\it XMM-Newton} observations of thirteen X-ray bright BL Lacertae objects, selected from the {\it Einstein} Slew Survey sample. The spectra are generally well fit by power-law models, with four objects having hard (α<1;Fννα\alpha<1; F_\nu \propto \nu^{-\alpha}) spectra that indicates synchrotron peaks at >5>5 keV. None of our spectra show line features, indicating that soft X-ray absorption ``notches'' must be rare amongst BL Lacs, rather than common or ubiquitous as had previously been asserted. We find significant curvature in most of the spectra. This curvature is almost certainly intrinsic, as it appears nearly constant from 0.5 to 6 keV, an observation which is inconsistent with the small columns seen in these sources.Comment: 4 pages, 1 figure; to be published in proceedings of the Cozumel meeting on "Multiwavelength Surveys for AGN", Cozumel 200

    This paper attempts to examine the current view of the informal sector, particularly as stated in the I.L.O. report, It suggests that the I.L.O. view is optimistic in thinking that the informal sector can be made more productive simply through organisational changes in its relationship to both government and industry. It tries to examine the constraints that act on the informal sector and concludes that it is limited in its ability to transcend barriers to entry into the formal sector through lack of access to capital resources, and is unable to gererate this capital from its own production surplus. Two case studies are given, one of an informal motor repair workshop and the other of a successful manufacturer of beds. The paper concludes that these constraints disqualify the informal sector from serious consideration as a dynamic growth point in the economy of Kenya

    BACKGROUND: Atrial fibrillation (AF) occurs in approximately one in three patients after cardiac surgery, and is associated with increased short-term and long-term mortality, intensive care unit (ICU) and hospital stay, and increased cost of care. In an attempt to reduce AF incidence in these patients, serum potassium (K+) levels are commonly maintained at the high end of normal (4.5-5.5 mEq/L). However, such potassium supplementation is without proven benefit, and is not without negative consequences. It carries clinical risk, negatively impacts patient experience and is both time-consuming and costly. This protocol describes a randomised controlled pilot trial to assess the feasibility of a proposed randomised non-inferiority trial to investigate the impact of maintaining serum potassium ≥ 3.6 mEq/L vs ≥ 4.5 mEq/L on the incidence of new-onset atrial fibrillation in the first 120 hours after isolated elective coronary artery bypass grafting. METHODS: Design: this is a randomized feasibility trial as a pilot for a randomized non-inferiority trial. PARTICIPANTS: are 160 patients undergoing isolated coronary artery bypass grafting at two centres. Allocation: patients will be randomized (1:1) to protocols aiming to maintain serum potassium at either ≥ 3.6 mEq/L ("relaxed control") or ≥ 4.5 mEq/L ("tight control"). Primary analytic aim: was to assess the feasibility and acceptability of planning and delivering the intervention and trial methods to inform a full-scale non-inferiority trial. OUTCOME: the primary indicative efficacy outcome measures being field-tested are feasibility of participant recruitment and randomization, maintaining a protocol violation rate < 10%, and retaining 90% patient follow up 28 days after surgery. The primary clinical outcome measure of the future full "Tight K Study" will be incidence of AF after cardiac surgery. DISCUSSION: The Tight K Pilot will assess the feasibility of conducting the full trial, which is intended to confirm or refute the efficacy of current potassium management in preventing AF after cardiac surgery.     This article seeks to shift the frame of analysis within which discussions of Indian indentured migration take place. It argues that colonial discourses and practices of indenture are best understood not with regard to the common historiographical framework of whether it was 'a new system of slavery', but in the context of colonial innovations in incarceration and confinement. The article shows how Indian experiences of and knowledge about transportation overseas to penal settlements informed in important ways both their own understandings and representations of migration and the colonial practices associated with the recruitment of indentured labour. In detailing the connections between two supposedly different labour regimes, it thus brings a further layer of complexity to debates around their supposed distinctions

    Localisation of RNAs into the germ plasm of vitellogenic xenopus oocytes

    We have studied the localisation of mRNAs in full-grown Xenopus laevis oocytes by injecting fluorescent RNAs, followed by confocal microscopy of the oocyte cortex. Concentrating on RNA encoding the Xenopus Nanos homologue, nanos1 (formerly Xcat2), we find that it consistently localised into aggregated germ plasm ribonucleoprotein (RNP) particles, independently of cytoskeletal integrity. This implies that a diffusion/entrapment-mediated mechanism is active, as previously reported for previtellogenic oocytes. Sometimes this was accompanied by localisation into scattered particles of the “late”, Vg1/VegT pathway; occasionally only late pathway localisation was seen. The Xpat RNA behaved in an identical fashion and for neither RNA was the localisation changed by any culture conditions tested. The identity of the labelled RNP aggregates as definitive germ plasm was confirmed by their inclusion of abundant mitochondria and co-localisation with the germ plasm protein Hermes. Further, the nanos1/Hermes RNP particles are interspersed with those containing the germ plasm protein Xpat. These aggregates may be followed into the germ plasm of unfertilized eggs, but with a notable reduction in its quantity, both in terms of injected molecules and endogenous structures. Our results conflict with previous reports that there is no RNA localisation in large oocytes, and that during mid-oogenesis even germ plasm RNAs localise exclusively by the late pathway. We find that in mid oogenesis nanos1 RNA also localises to germ plasm but also by the late pathway. Late pathway RNAs, Vg1 and VegT, also may localise into germ plasm. Our results support the view that mechanistically the two modes of localisation are extremely similar, and that in an injection experiment RNAs might utilise either pathway, the distinction in fates being very subtle and subject to variation. We discuss these results in relation to their biological significance and the results of others

    At the Fifth Space Congress, a method was [ presented for predicting the inviscid gas properties surrounding blunt nosed aerospace j vehicles in supersonic or hypersonic flight, at zero angle of attack {Reference l)o Two computer programs used in combination were utilized.) :At that time it was indicated (References 1 and 2) that this capability was to be extended, using j a new unique system of eleven different computer j programs, in order to provide for the calculation; of gaseous properties and aerodynamic characteristics for vehicles flying at angles of attack. At angles of attack, the problem of predicting the gas properties and aerodynamic characteristics is an order of magnitude more difficult due to the fact that the flow field is threedimensional. That is, the flow properties vary in all three directions: as a function of body axial station, location around the body (at a given station), and distance away from the body. ;The newer set of programs also provides the capability of handling vehicles with sharp as well as blunt noses, and includes the capability of predicting the viscous flow boundary layer properties on the vehicle surface. Most of the programs were originally developed under U.S. Air Force and Army Contracts by General Applied Science Laboratories. The programs were converted to operate on the Control Data Model CDC 6^-00 digital computer used at Martin Marietta I Corporation\u27s Orlando Division. Over the past year, many modifications have been made to the system of computer programs to increase the efficiency of utilization (by simplifying the j inputs required, etc.), reduce the computer run I time (cost of operation), and to increase its : capability (calculate aerodynamic characteristics etc.)

    In the aftermath of the 2010 and 2011 earthquakes in Christchurch, New Zealand, community-led temporary and adaptive urbanism filled a gap between the emergency response and recovery. In the space between response and recovery, the citizens of Christchurch showed their commitment to rethinking how they wanted to rebuild and then regenerate their city, leading to the embrace of collaborative processes, temporary and adaptive urbanism principles and a range of placemaking responses. In this chapter, the role of placemaking as a tool for post-disaster regeneration and resilience is considered by assessing three case study placemaking projects: the Re:START Mall, the Festival of Transitional Architecture (FESTA) and the placemaking programme at the Commons. Their development along with their success is considered within the context of the recovery of Christchurch and, in particular, how they align to the The Resilient Greater Christchurch Framework as it is set out in the Resilient Greater Christchurch Plan, in order to determine their role in building the resilience of Christchurch
