    Identification of a novel linear B-cell epitope in the UL26 and UL26.5 proteins of Duck Enteritis Virus

    BACKGROUND: The Unique Long 26 (UL26) and UL26.5 proteins of herpes simplex virus are known to function during the assembly of the viruses. However, for duck enteritis virus (DEV), which is an unassigned member of the family Herpesviridae, little information is available about the function of the two proteins. In this study, the C-terminus of DEV UL26 protein (designated UL26c), which contains the whole of UL26.5, was expressed, and the recombinant UL26c protein was used to immunize BALB/c mice to generate monoclonal antibodies (mAb). The mAb 1C8 was generated against DEV UL26 and UL26.5 proteins and used subsequently to map the epitope in this region. Both the mAb and its defined epitope will provide potential tools for further study of DEV. RESULTS: A mAb (designated 1C8) was generated against the DEV UL26c protein, and a series of 17 partially overlapping fragments that spanned the DEV UL26c were expressed with GST tags. These peptides were subjected to enzyme-linked immunosorbent assay (ELISA) and western blotting analysis using mAb 1C8 to identify the epitope. A linear motif, (520)IYYPGE(525), which was located at the C-terminus of the DEV UL26 and UL26.5 proteins, was identified by mAb 1C8. The result of the ELISA showed that this epitope could be recognized by DEV-positive serum from mice. The (520)IYYPGE(525 )motif was the minimal requirement for reactivity, as demonstrated by analysis of the reactivity of 1C8 with several truncated peptides derived from the motif. Alignment and comparison of the 1C8-defined epitope sequence with those of other alphaherpesviruses indicated that the motif (521)YYPGE(525 )in the epitope sequence was conserved among the alphaherpesviruses. CONCLUSION: A mAb, 1C8, was generated against DEV UL26c and the epitope-defined minimal sequence obtained using mAb 1C8 was (520)IYYPGE(525). The mAb and the identified epitope may be useful for further study of the design of diagnostic reagents for DEV

    Mechanical properties of in-situ synthesis of Ti-Ti3Al metal composite prepared by selective laser melting

    Titanium composite strengthened by Ti3Al precipitations is considered to be one of the excellent materials that is widely used in engineering. In this work, we prepared a kind of Ti-Ti3Al metallic composite by in-situ synthesis technology during the SLM (selective laser melting) process, and analyzed its microstructure, wear resistance, microhardness, and compression properties. The results showed that the Ti-Ti3Al composite, prepared by in-situ synthesis technology based on SLM, had more homogeneous Ti3Al-enhanced phase dispersion strengthening structure. The grain size of the workpiece was about 1 μm, and that of the Ti3Al particle was about 200 nm. Granular Ti3Al was precipitated after the aluminum-containing workpiece formed, with a relatively uniform distribution. Regarding the mechanical properties, the hardness (539 HV) and the wear resistance were significantly improved when compared with the Cp-Ti workpiece. The compressive strength of the workpiece increased from 886.32 MPa to 1568 MPa, and the tensile strength of the workpiece increased from 531 MPa to 567 MPa after adding aluminum. In the future, the combination of in-situ synthesis technology and SLM technology can be used to flexibly adjust the properties of Ti-based materials

    Human migration activities drive the fluctuation of ARGs : Case study of landfills in Nanjing, eastern China

    Landfills are perfect sites to study the effect of human migration on fluctuation of antibiotic resistance genes (ARGs) as they are the final destination of municipal waste. For example, large-scale human migration during the holidays is often accompanied by changes in waste dumping having potential effects on ARG abundance. Three landfills were selected to examine fluctuation in the abundance of fifteen ARGs and Intl1 genes for 14 months in Nanjing, eastern China. Mass human migration, the amount of dumped waste and temperature exerted the most significant effects on bimonthly fluctuations of ARG levels in landfill sites. As a middle-sized cosmopolitan city in China, millions of college students and workers migrate during holidays, contributing to the dramatic increases in waste production and fluctuation in ARG abundances. In line with this, mass migration explained most of the variation in waste dumping. The waste dumping also affected the bioaccessibility of mixed-compound pollutants that further positively impacted the level of ARGs. The influence of various bioaccessible compounds on ARG abundance followed the order: antibiotics>nutrients>metals>organic pollutants. Concentrations of bioaccessible compounds were more strongly correlated with ARG levels compared to total compound concentrations. Improved waste classification and management strategies could thus help to decrease the amount of bioaccessible pollutants leading to more effective control for urban ARG dissemination

    Mechanical properties of AlSi10Mg alloy fabricated by laser melting deposition and improvements via heat treatment

    Process optimization and heat treatment of AlSi10Mg aluminum alloy parts fabricated by laser melting deposition (LMD) based on coaxial powder feeding are conducted in this paper to improve manufacturing quality. The microstructures and mechanical properties of the LMD-built AlSi10Mg alloy parts are systematically investigated. Experimental results show the relative density of the block samples increase to 99.2% without larger pores and cracks after process optimization. The sample microstructures are found to display directional rapid solidification characteristics, with the Al-Si eutectic microstructure containing three microstructures being cellular, columnar dendrites and divergent dendrites. With solution and artificial aging heat treatments, Si atoms are rejected from the supersaturated Al matrix to form small Si granular particles. The heat treated samples display a uniform microstructure without heterogeneities and the microhardness remains stable at 118 HV. When the as-built sample is heat treated for solution time 2h, tensile strength increases from 292 MPa to 342 MPa due to the formation of strengthening phase Mg2Si

We shall determine two factors that have obvious influences on revenue through a gradual regression and then do some relevant tests. Finally, we shall study and analyze its economic significance, so as to put forward some interrelated suggestions

    Reverse Auction Based Green Offloading Scheme for Small Cell Heterogeneous Networks

    The small cell is treated as a promising proposition to provide hot spot capacity and higher data rates. However, even with dense small cell deployment scenario, the heavy traffic load pressure and low energy efficiency in the small cell heterogeneous network (HetNet) still exist. Therefore, how to make the best use of densely deployed small cells under HetNet environments becomes the focus of researches. Offloading provides a feasible solution to promote cooperation between macrocells and small cells for user traffic supporting. In this paper, we propose the reverse auction based Green Offloading (GO) scheme for energy efficiency improvements. The proposed GO scheme employs the reverse auction theory to handle the offloading decision process, aiming at maximizing the system energy efficiency under the constraints of user Quality of Service (QoS) requirements, bandwidth, and transmission power limitations. Moreover, the reverse auction model gives the facility of multicell coordination transmissions with multiple winners situation. The energy efficiency optimization problem with constraints is solved by Dynamic Programming method with Karush-Kuhn-Tucker (KKT) conditions. The Individual Rationality and Truthfulness of the reverse auction model are also proved. By comparing the energy efficiency performances of the proposed GO scheme with current works within the Long Term Evolution-Advanced (LTE-A) system downlink scenario, simulation results show the improvements of the proposed GO scheme.</jats:p

    Age similarity enhances older adults’ learning from co-viewing video lectures

    Due to the popularity of video lectures, an increasing number of older adults are engaging with this form of learning. Furthermore, co-viewing has become a common type of social interaction. Despite the popularity of video lectures among older adults, there is limited understanding of how to effectively support their learning through this medium. The present study tested the interaction effects of co-viewer similarity (young vs. older co-viewer) and the type of learning content (declarative vs. procedural knowledge) in video lectures on older adults’ learning performance, motivation, cognitive load, and PFC activation as measured by fNIRS. Our results found that older adults showed better learning performance, greater motivation, and reduced cognitive load when co-viewing with an older viewer compared to a young one, regardless of the type of learning content. Interestingly, for declarative knowledge (i.e., Traditional Chinese Medicine, older adults exhibited greater activation in the FEFs, S1, and SMC regions when co-viewing with an older viewer compared with a young viewer. For procedural knowledge (i.e., digital technology), they exhibited less activation in the dlPFC and SMC regions when co-viewing with an older viewer compared with a young viewer. Our findings suggest that educators should consider the age of co-viewers when designing video lectures for older adults, irrespective of the type of knowledge being taught.<br/

    Mask-ControlNet: Higher-Quality Image Generation with An Additional Mask Prompt

    Text-to-image generation has witnessed great progress, especially with the recent advancements in diffusion models. Since texts cannot provide detailed conditions like object appearance, reference images are usually leveraged for the control of objects in the generated images. However, existing methods still suffer limited accuracy when the relationship between the foreground and background is complicated. To address this issue, we develop a framework termed Mask-ControlNet by introducing an additional mask prompt. Specifically, we first employ large vision models to obtain masks to segment the objects of interest in the reference image. Then, the object images are employed as additional prompts to facilitate the diffusion model to better understand the relationship between foreground and background regions during image generation. Experiments show that the mask prompts enhance the controllability of the diffusion model to maintain higher fidelity to the reference image while achieving better image quality. Comparison with previous text-to-image generation methods demonstrates our method's superior quantitative and qualitative performance on the benchmark datasets
