The first is the solution of a challenge question, proposed by Etienne Ghys, on the determination of all maps from rational numbers to themselves such that the difference quotient (f(x)-f(y))/(x-y) is always a square.     Biomimetic hydrogel based on HET-s amylo\"id fibers for long-term culture of primary hippocampal neurons

    Historically, amyloid fibers (AF) in research has always been linked to degenerative diseases. However, HET-s AF, by their morphology and function, have only little in common to pathogenic amyloid fibers such as {\alpha}-synuclein or a\b{eta} and they have appeared as promising candidate for biocoating since few years. Here we have shown than HET-s amyloid fibers hydrogel is an extremely polyvalent coating material for the in vitro culture of primary hippocampal neurons. First, the non-cytotoxicity was demonstrated in vitro using standardized ISO protocols. Then, it is shown that in vitro culture of primary hippocampal neurons on HET-s AF hydrogels could last more than 45 days with clear signatures of spontaneous network activity, with which is a feat that not many other coatings have achieved yet. Finally, interactions between the cells, the dendrites and the hydrogels are highlighted, showing that dendrites might be able to penetrate the hydrogels in depth, therefore allowing recordings even within micrometer-thick hydrogels. In the end, those properties combined with group functionalization using standard biochemistry techniques, makes HET-s hydrogels ideal candidates to be used for the long-term growth of neurons as well as other types of cells. This versatility and easiness to use are definitely still unheard, especially for protein material. Due to its ability to transform from dry films to hydrogel when in contact with the extracellular matrix (ECM), it could also be used for in vivo implants, solving the issue of hydrogel damaging during the implant surgery

    Multivariate analysis of seasonal variation in the composition and thermal properties of butterfat with an emphasis on authenticity assessment

    The aim of this study was to analyze the seasonal variation in the composition and thermal properties of butterfat (BF) in order to evaluate the applicability of differential scanning calorimetry (DSC) for the authenticity assessment of butter. The composition of fatty acids (FA) and triacylglycerols (TAG) and the thermal properties of genuine BF purchased in the summer and in the winter from six producers were determined. Principal component analysis (PCA) was used to recognize variation and as a result, all BF samples were classified into two groups: one composed of mixed samples from the summer and winter and the other comprising only summer BF samples. DSC and GC analysis revealed that the group of only summer BF samples was characterized by lower melting temperatures and peak heights of low- and medium melting fractions and the highest proportions of unsaturated FAs (ΣC18:1, ΣC18:2, ΣC18:3). The results indicated that most of the variation in the composition and thermal properties was affected by summer BF samples, which may result from the alternative animal feeding systems employed in the summer season, i.e., pasture vs. indoor. Therefore, seasonal variation should be taken into consideration during the elaboration of the analytical method of authenticity assessment

    Perspectives and Potential Applications of Mitochondria-Targeted Antioxidants in Cardiometabolic Diseases and Type 2 Diabetes

    There is abundant evidence to suggest that mitochondrial dysfunction is a main cause of insulin resistance and related cardiometabolic comorbidities. On the other hand, insulin resistance is one of the main characteristics of type 2 diabetes, obesity, and metabolic syndrome. Lipid and glucose metabolism require mitochondria to generate energy, and when O2 consumption is low due to inefficient nutrient oxidation, there is an increase in reactive oxygen species, which can impair different types of molecules, including DNA, lipids, proteins, and carbohydrates, thereby inducing proinflammatory processes. Factors which contribute to mitochondrial dysfunction, such as mitochondrial biogenesis and genetics, can also lead to insulin resistance in different insulin-target tissues, and its association with mitochondrial dysfunction can culminate in the development of cardiovascular diseases. In this context, therapies that improve mitochondrial function may also improve insulin resistance. This review explains mechanisms of mitochondrial function related to the pathological effects of insulin resistance in different tissues.     Analysis of 339 pregnancies in 181 women with 13 different forms of inherited thrombocytopenia

    65Pregnancy in women with inherited thrombocytopenias is a major matter of concern as both the mothers and the newborns are potentially at risk of bleeding. However, medical management of this condition cannot be based on evidence because of the lack of consistent information in the literature. To advance knowledge on this matter, we performed a multicentric, retrospective study evaluating 339 pregnancies in 181 women with 13 different forms of inherited thrombocytopenia. Neither the degree of thrombocytopenia nor the severity of bleeding tendency worsened during pregnancy and the course of pregnancy did not differ from that of healthy subjects in terms of miscarriages, fetal bleeding and pre-term births. The degree of thrombocytopenia in the babies was similar to that in the mother. Only 7 of 156 affected newborns had delivery-related bleeding, but 2 of them died of cerebral hemorrhage. The frequency of delivery-related maternal bleeding ranged from 6.8% to 14.2% depending on the definition of abnormal blood loss, suggesting that the risk of abnormal blood loss was increased with respect to the general population. However, no mother died or had to undergo hysterectomy to arrest bleeding. The search for parameters predicting delivery-related bleeding in the mother suggested that hemorrhages requiring blood transfusion were more frequent in women with history of severe bleedings before pregnancy and with platelet count at delivery below 50 × 10(9)/L.openopenPatrizia Noris; Nicole Schlegel; Catherine Klersy; Paula G.     Validation and Evaluation of Lateral Flow Tests for the Detection of Antimicrobial Residues on Poultry Feathers

    Persistence of antimicrobial drugs (AMDs) administered to poultry is longer in feathers than in edible tissues. Hence, poultry feathers are a suitable matrix to investigate historical exposure contributing to antimicrobial resistance, since current detection methods are either non‐specific or highly technical and costly. Here we present an analysis of the performance of lateral flow test (LFT) panels in the detection of five AMD classes, namely sulfonamides, tetracyclines, beta‐lactams, quinolones, and aminoglycosides, on chicken feather samples. The limit of detection (LOD) of eight AMD substances was determined between 4.7 μg/kg for enrofloxacin and 700 μg/kg for streptomycin. The performance of feather LFT was evaluated for four AMD classes against the reference method (LC–MS/MS). From 79 samples collected from the field, LFT test specificity ranged from 0.63 (quinolones) to 0.95 (tetracyclines). Test sensitivity ranged from 0.15 (beta‐lactams) to 0.78 (quinolones and tetracyclines). LFT testing had the greatest discriminatory power for tetracyclines (specificity 0.95 and sensitivity 0.78). LFT had similar test characteristics for sulfonamides and quinolones and performed poorly for beta‐lactams. Poor recovery rates (< 15%) were observed in neomycin, kanamycin, and ampicillin. These methods are suitable for preliminarily screening tetracyclines, sulfonamides, and quinolones, with recommendations for further extraction protocols
