    This study examined some dynamic factors related to doctors’ stress in government hospitals in Nigeria using Lagos State as case study. The working hours and the number of patients the professionals attended to showed that they are likely to be stressed. A model was developed to predict the dynamic influence to doctors’ stress using logistic regression model. It was discovered that illness, environmental factors, profession and workloads and hours are the variables that highly influenced Doctors’ stress

    Targeting the CXCR4 pathway using a novel anti-CXCR4 IgG1 antibody (PF-06747143) in chronic lymphocytic leukemia.

    BackgroundThe CXCR4-CXCL12 axis plays an important role in the chronic lymphocytic leukemia (CLL)-microenvironment interaction. Overexpression of CXCR4 has been reported in different hematological malignancies including CLL. Binding of the pro-survival chemokine CXCL12 with its cognate receptor CXCR4 induces cell migration. CXCL12/CXCR4 signaling axis promotes cell survival and proliferation and may contribute to the tropism of leukemia cells towards lymphoid tissues and bone marrow. Therefore, we hypothesized that targeting CXCR4 with an IgG1 antibody, PF-06747143, may constitute an effective therapeutic approach for CLL.MethodsPatient-derived primary CLL-B cells were assessed for cytotoxicity in an in vitro model of CLL microenvironment. PF-06747143 was analyzed for cell death induction and for its potential to interfere with the chemokine CXCL12-induced mechanisms, including migration and F-actin polymerization. PF-06747143 in vivo efficacy was determined in a CLL murine xenograft tumor model.ResultsPF-06747143, a novel-humanized IgG1 CXCR4 antagonist antibody, induced cell death of patient-derived primary CLL-B cells, in presence or absence of stromal cells. Moreover, cell death induction by the antibody was independent of CLL high-risk prognostic markers. The cell death mechanism was dependent on CXCR4 expression, required antibody bivalency, involved reactive oxygen species production, and did not require caspase activation, all characteristics reminiscent of programmed cell death (PCD). PF-06747143 also induced potent B-CLL cytotoxicity via Fc-driven antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity activity (CDC). PF-06747143 had significant combinatorial effect with standard of care (SOC) agents in B-CLL treatment, including rituximab, fludarabine (F-ara-A), ibrutinib, and bendamustine. In a CLL xenograft model, PF-06747143 decreased tumor burden and improved survival as a monotherapy, and in combination with bendamustine.ConclusionsWe show evidence that PF-06747143 has biological activity in CLL primary cells, supporting a rationale for evaluation of PF-06747143 for the treatment of CLL patients

    Ulocuplumab (BMS-936564 / MDX1338): a fully human anti-CXCR4 antibody induces cell death in chronic lymphocytic leukemia mediated through a reactive oxygen species-dependent pathway.

    The CXCR4 receptor (Chemokine C-X-C motif receptor 4) is highly expressed in different hematological malignancies including chronic lymphocytic leukemia (CLL). The CXCR4 ligand (CXCL12) stimulates CXCR4 promoting cell survival and proliferation, and may contribute to the tropism of leukemia cells towards lymphoid tissues. Therefore, strategies targeting CXCR4 may constitute an effective therapeutic approach for CLL. To address that question, we studied the effect of Ulocuplumab (BMS-936564), a fully human IgG4 anti-CXCR4 antibody, using a stroma--CLL cells co-culture model. We found that Ulocuplumab (BMS-936564) inhibited CXCL12 mediated CXCR4 activation-migration of CLL cells at nanomolar concentrations. This effect was comparable to AMD3100 (Plerixafor--Mozobil), a small molecule CXCR4 inhibitor. However, Ulocuplumab (BMS-936564) but not AMD3100 induced apoptosis in CLL at nanomolar concentrations in the presence or absence of stromal cell support. This pro-apoptotic effect was independent of CLL high-risk prognostic markers, was associated with production of reactive oxygen species and did not require caspase activation. Overall, these findings are evidence that Ulocuplumab (BMS-936564) has biological activity in CLL, highlight the relevance of the CXCR4-CXCL12 pathway as a therapeutic target in CLL, and provide biological rationale for ongoing clinical trials in CLL and other hematological malignancies

    Comparison of Sulfur Dioxide Removal Reactions Kinetics by Na2CO3 and Other Different Sorbents from Coal-fired Power Plants

    This work deals with kinetic parameters estimation of Na2CO3+SO2 reaction employing sophisticated random pore model. The temperature of experiments ranges from 100 to 250 °C, and various SO2 concentrations are within 0.13–1.12 vol.%. According to the results, the reaction rate concentration dependency follows the fractional function. The values of rate constants and product layer diffusivities are expressed at various temperatures. Finally, it was attempted to describe the significance of this sorbent for SO2 removal. Therefore, the kinetic results of Na2CO3+SO2 reaction were compared with other similar studies on SO2 reaction kinetics with CaO, CuO, and MgO sorbents. It was concluded that Na2CO3 shows advantages of higher rate constants, lower operating temperatures, and less possibility of incomplete conversion problem. The reported kinetic constants are essential for design of flue gas desulfurization reactors, especially in coalfired power plants.     HEALTH COMMUNICATION ANALYSIS OF DOCTOR-PATIENT RELATIONSHIP AND PATIENTS’ HEALTH BEHAVIOUR IN SOUTH WEST NIGERIA

    Communication in doctor-patient relationship has undergone a transition over time from doctor-dominance to patient-centred approach. Unfortunately, the patient-centred approach adopted today has not positively influenced the health behaviour of patients. For this, health communication scholars wonder if there is any difference between patient-centred and the doctor-dominance approach. The study adopted the descriptive survey research method, using two sets of questionnaires for data collection from patients and doctors. Multi-stage sampling technique was used to select 120 respondents for the study. Data collected were analyzed using frequency counts, percentages and chi-square technique. Communication in doctor-patient relationship had significant influence on patients’ follow-up appointments (χ2 = 112.867) and compliance to prescribed drugs (χ2 = 48.333).   Communication in doctor-patient   relationship had significant influence on patients’ choice of the hospital (χ2= 44.083) and  consumption of balanced or specific diet (χ2 = 61.350). Communication in doctor-patient relationship had significant influence on patients’ exercising regularly (χ2 =18.80).The study concluded that doctor-patient relationship in health communication influences patients’ health behaviour in South West Nigeria. It is therefore recommended that doctors should apply an integrated/ synergetic approach in communicating with their patients and that audience-specific social media platforms should be utilized to complement the doctor-patient communication for more effective result. &nbsp

    Monoclonal antibodies directed to fucoidan preparations from brown algae

    Cell walls of the brown algae contain a diverse range of polysaccharides with useful bioactivities. The precise structures of the sulfated fucan/fucoidan group of polysaccharides and their roles in generating cell wall architectures and cell properties are not known in detail. Four rat monoclonal antibodies, BAM1 to BAM4, directed to sulfated fucan preparations, have been generated and used to dissect the heterogeneity of brown algal cell wall polysaccharides. BAM1 and BAM4, respectively, bind to a non-sulfated epitope and a sulfated epitope present in the sulfated fucan preparations. BAM2 and BAM3 identified additional distinct epitopes present in the fucoidan preparations. All four epitopes, not yet fully characterised, occur widely within the major brown algal taxonomic groups and show divergent distribution patterns in tissues. The analysis of cell wall extractions and fluorescence imaging reveal differences in the occurrence of the BAM1 to BAM4 epitopes in various tissues of Fucus vesiculosus. In Ectocarpus subulatus, a species closely related to the brown algal model Ectocarpus siliculosus, the BAM4 sulfated epitope was modulated in relation to salinity levels. This new set of monoclonal antibodies will be useful for the dissection of the highly complex and yet poorly resolved sulfated polysaccharides in the brown algae in relation to their ecological and economic significance

    Era revolusi industri 4.0 dan situasi pandemi COVID 19 menuntut entitas usaha untuk memaksimalkan penggunaan teknologi informasi berbasis digital secara tepat guna yakni melakukan pemasaran produk usaha. Namun saat ini tidak banyak BUMDes yang menggunakan teknologi informasi dalam pemasarannya. Hal ini juga terjadi di Desa Kesiut yang berlokasi di Kecamatan Kerambitan, Kabupaten Tabanan Bali.  BUMDes di Desa Kesiut masih menerapkan sistem pemasaran konvensional dalam memasarkan produk usahanya. Tentu hal ini jika tidak segera ditindaklanjuti akan menimbulkan tidak kompetitifnya usaha yang berdampak pada risiko kerugian dimasa depan terlebih saat ini sedang dilanda COVID 19 yang memerlukan model cashlees dan social distance dalam aktifitas transasksi. Kendala utama yang dihadapi adalah kurangya sumber daya manusia yang handal dan profesional. Solusi pemecahan masalah dilaksanakan dengan melakukan sosialisasi, edukasi, pelatihan dan pendampingan implementasi sistem pemasaran secara digital.  Dari solusi yang dicanangkan tersebut maka diharapkan mampu memberi pengetahuan sekaligus meningkatkan keterampilan masyarakat khususnya manajemen BUMDes untuk menggunakan teknologi informasi didalam menerapkan sistem pemasaran produk berbasis teknologi digital. Metode pelaksanaan dilakukan dalam beberapa tahap, yakni diawali dengan memberikan pemahaman akan pentingya penggunaan teknologi informasi, selanjutnya dilakukan juga pelatihan pembuatan portal website. Tahap akhir dilakukan pendampingan untuk membantu pemecahan masalah yang dihadapi terkait dengan penerapkan sistem digital marketing.  Harapan yang diinginkan melalui program kemitraan masyarakat yang dikejewantahkan melalui tahapan sosialisasi, edukasi, pelatihan maupun pendampingan ini adalah BUMDes dalam menjalankan kegiatan usahanya mampu menerapkan teknologi informasi berbasis digitalisi Marketing dalam menjalankan usahanya. Dengan melaksanakan kegiatan ini diharapkan dalam jangka panjang BUMDes di Desa Kesiut akan semakin berkembang dan mampu menggeliatkan perekonomian Desa sehingga akan terjadi akselerasi kesejahteraan masyarakat Desa

    Climate, health, and living condition crises in the expanding informal settlements and slums of South-West Nigeria:a case report of Ogun and Oyo states

    The rapid expansion of informal settlements and slums in southwestern Nigeria, particularly along the Lagos-Ogun-Oyo corridor, underscores the critical challenges posed by rapid urbanisation and population growth in this major economic hub of West Africa. This corridor, home to key infrastructure, industries, and economic activities, has seen a significant rise in unplanned communities due to its economic opportunities. In particular, Ogun and Oyo states, both sharing borders with Lagos, have become hotspots of these communities, characterised by inadequate housing, poor sanitation, unsafe drinking water, and heightened vulnerability to climate-related hazards. In this viewpoint, we examine the health and environmental challenges faced by residents in these informal settlements, including respiratory illnesses, waterborne diseases, and climate-related health risks exacerbated by poor air quality and extreme heat. Despite various research efforts, policy reforms, and programmatic interventions, challenges such as limited funding, inadequate enforcement, and a lack of coordination among stakeholders persist. We propose a holistic, multi-sectoral approach that could improve living standards and health outcomes through community empowerment, participatory urban planning, microfinance initiatives, and climate resilience programmes. We note that this requires collaborative efforts from government, non-governmental organisations, and local residents to create sustainable and resilient urban environments.</p

    MAGNETIC PROPERTIES OF HEMATITE NANOSTRUCTURES

    Son estudiados los procedimientos de producción nanopartículas de hematita, evaluándose preferentemente, las producidasdas por mecanosíntesís, mediante espectroscopia Mossbauer, difracción de rayos X y magnetometría. Se produjeron, luego de 2 horas de tratamiento mecánico, partículas de tamaños alrededor 15 nm. La técnica de producción por mecanosíntesis no implicó transformación estructural de la fase cristalográfica inicial. La espectroscopia Mossbauer del 57Fe correlacionada con la magnetometría, mostraron también que la transición de Morín no es observada inmediatamente después de 0,75 horas de mecanosíntesis.Nanostructured a-Fe203 (hematite) was produced usíng high-energy ball milling and analized by X-ray diffraction (XRD), 57Fe Mi.issbauer spectrometry and magnetization measurements. The results showed that after 2 h milling, a-Fe203 nanosize particles were obtained about 15 nm. The 57 Fe Mossbauer spectrometry correlated with magnetometry showed also that Morin transition was notobserved after 0,75 h milling
