    Le bouturage d'Acacia senegal est possible à partir de fragments de rameaux prélevés dans le houppier d'arbres adultes (13 ans). Le matériel végétal le plus réactif est constitué par des boutures de 15 cm de long (12 à 15 nouds) et de 10 +/- 6 mm de diamètre. L'ablation des feuilles avant mise en culture est un facteur favorable à l'enracinement des boutures. Celui-ci est aussi très influencé par la date de prélèvement des boutures et il varie de 10% en saison sèche à plus de 70% en saison des pluies. La collecte de boutures peut se faire sous forme de fragments de rameaux de 50 cm de long. Il est alors possible de conserver ce matériel pendant huit jours dans une toile maintenue humide et d'y découper les boutures au moment de la mise en place en pépinière. Les boutures présentent des caractères de maturité (croissance lente, plagiotropie, fructification). Il est cependant possible de conserver les génotypes ainsi mobilisés au champ, ou dans des pots en pépinière, afin d'amplifier le processus de rajeunissement physiologique en vue de leur clonage in vitr

    Molecular Modeling Studies to Probe the Binding Hypothesis of Novel Lead Compounds against Multidrug Resistance Protein ABCB1.

    The expression of drug efflux pump ABCB1/P-glycoprotein (P-gp), a transmembrane protein belonging to the ATP-binding cassette superfamily, is a leading cause of multidrug resistance (MDR). We previously curated a dataset of structurally diverse and selective inhibitors of ABCB1 to develop a pharmacophore model that was used to identify four novel compounds, which we showed to be potent and efficacious inhibitors of ABCB1. Here, we dock the inhibitors into a model structure of the human transporter and use molecular dynamics (MD) simulations to report the conformational dynamics of human ABCB1 induced by the binding of the inhibitors. The binding hypotheses are compared to the wider curated dataset and those previously reported in the literature. Protein-ligand interactions and MD simulations are in good agreement and, combined with LipE profiling, statistical and pharmacokinetic analyses, are indicative of potent and selective inhibition of ABCB1

    Accuracy of CT Colonography for Detection of Large Adenomas and Cancers

    Background Computed tomographic (CT) colonography is a noninvasive option in screening for colorectal cancer. However, its accuracy as a screening tool in asymptomatic adults has not been well defined. Methods We recruited 2600 asymptomatic study participants, 50 years of age or older, at 15 study centers. CT colonographic images were acquired with the use of standard bowel preparation, stool and fluid tagging, mechanical insufflation, and multidetector-row CT scanners (with 16 or more rows). Radiologists trained in CT colonography reported all lesions measuring 5 mm or more in diameter. Optical colonoscopy and histologic review were performed according to established clinical protocols at each center and served as the reference standard. The primary end point was detection by CT colonography of histologically confirmed large adenomas and adenocarcinomas (10 mm in diameter or larger) that had been detected by colonoscopy; detection of smaller colorectal lesions (6 to 9 mm in diameter) was also evaluated. Results Complete data were available for 2531 participants (97%). For large adenomas and cancers, the mean (±SE) per-patient estimates of the sensitivity, specificity, positive and negative predictive values, and area under the receiver-operating-characteristic curve for CT colonography were 0.90±0.03, 0.86±0.02, 0.23±0.02, 0.99± Conclusions In this study of asymptomatic adults, CT colonographic screening identified 90% of subjects with adenomas or cancers measuring 10 mm or more in diameter. These findings augment published data on the role of CT colonography in screening patients with an average risk of colorectal cancer.     Sarcoid-Like Reaction Associated with Renal Cell Carcinoma - A Case Report.

    Renal cell carcinoma (RCC) is a highly vascular tumor, which may spread to the lungs and other organs. It often presents with localized or systemic manifestation, including paraneoplastic syndromes. Sarcoidosis is a systemic granulomatous inflammatory disease characterized by non-caseating granulomas that typically afflicts the respiratory system. In the absence of any evidence of systemic sarcoidosis they are referred to as sarcoid-like reactions. Non-caseating epithelioid granulomas, also regarded to sarcoid-like granulomas have been described in association with certain malignancies such as carcinomas of the breast, colon, seminoma, and Hodgkin\u27s lymphoma. However, sarcoid like reaction associated with renal cell carcinoma is uncommon.     Mycobacterium tuberculosis (MTB) infection is the ninth leading cause of death worldwide, with many individuals with undiagnosed active or latent disease. The presence of parenchymal lung disease, such as interstitial lung disease (ILD), has been suggested to increase the risk of pulmonary tuberculosis (TB). In the clinical setting of ILD, the diagnosis of an underlying MTB infection may be challenging due to the interstitial process and underlying fibrosis, which may mask the infection. An atypical presentation and misleading radiological patterns may delay the diagnosis of the underlying MTB infection. Herein, we describe a unique case of ILD complicated by active MTB infection in an 84-year-old male, which presented as a diagnostic and clinical challenge.     The need for an online collection of traditional african food habits

    Amongst the difficulties facing the indigenous people of Africa today is the deleterious shift from traditional food habits to the processed and packaged food products of western-owned corporations. This nutrition transition has been implicated in the rise of non-communicable diseases (NCDs) throughout Africa. The purpose of the present investigation was to determine whether there is a current need to document traditional African food habits via an online collection in an attempt to stimulate further research in this area and potentially improve the health status of indigenous Africans threatened by the nutrition transition. A systematic  search was performed to assess possible gaps in online collections focused on traditional African food habits. A questionnaire was administered to opinion leaders in the nutritional sciences at the 18th International Congress of Nutrition (ICN) in Durban, South Africa, September 2005, to determine the level of awareness of the importance of traditional African food habits within the context of the nutrition transition, and to determine the support among this cohort for an online collection of traditional African food habits. Our systematic review resulted in nine collections being identified. None of these collections were specifically  designed to raise  awareness of traditional African food habits however. Findings from the survey revealed that 86% of our cohort agreed that Africa is currently undergoing a  nutrition transition. Nearly 80% believed that knowledge of traditional African food habits is being lost. Indigenous African interviewees noted reduced consumption of sorghum and millet and an increased   consumption of wheat and rice within their region of origin. Approximately 82% believed that there was currently a gap in online collections focused on presenting information on traditional African food habits. Ninety-two percent of the cohort indicated their preparedness to make use of a novel, online collection of data on traditional African food habits. The findings revealed a critical need to collate and present data on traditional African food habits via a novel, online collection that could be used to stimulate education and research of food habits and their health implications, to provide a well-rounded forum in which such information is presented and shared.Key words: Africa, traditional foods, wild species, dietary practices, information networks and database

    Developmental and/or epileptic encephalopathies (DEEs) are a group of devastating genetic disorders, resulting in early-onset, therapy-resistant seizures and developmental delay. Here we report on 22 individuals from 15 families presenting with a severe form of intractable epilepsy, severe developmental delay, progressive microcephaly, visual disturbance and similar minor dysmorphisms. Whole exome sequencing identified a recurrent, homozygous variant (chr2:64083454A > G) in the essential UDP-glucose pyrophosphorylase (UGP2) gene in all probands. This rare variant results in a tolerable Met12Val missense change of the longer UGP2 protein isoform but causes a disruption of the start codon of the shorter isoform, which is predominant in brain. We show that the absence of the shorter isoform leads to a reduction of functional UGP2 enzyme in neural stem cells, leading to altered glycogen metabolism, upregulated unfolded protein response and premature neuronal differentiation, as modeled during pluripotent stem cell differentiation in vitro. In contrast, the complete lack of all UGP2 isoforms leads to differentiation defects in multiple lineages in human cells. Reduced expression of Ugp2a/Ugp2b in vivo in zebrafish mimics visual disturbance and mutant animals show a behavioral phenotype. Our study identifies a recurrent start codon mutation in UGP2 as a cause of a novel autosomal recessive DEE syndrome. Importantly, it also shows that isoform-specific start-loss mutations causing expression loss of a tissue-relevant isoform of an essential protein can cause a genetic disease, even when an organism-wide protein absence is incompatible with life. We provide additional examples where a similar disease mechanism applies
