    Acute Myocardial Infarction after MaxMan Consumption with Normal Angiography: A Case Report

    Acute myocardial infarction (MI) with a normal coronary angiogram is a well-established abnormality, although it is usually related with stenosis in coronary arteries. We reported here a case of a 24-year-old man referring with acute inferior MI after MaxMan capsule consumption. The patient had not any related risk factors of coronary artery disease and a history of prior MI. The acute inferior infarction in this patient was detected in the electrocardiogram in the absence of any stenosis or obstruction in the angiogram. We do not recommend MaxMan intake as a usual drug before intercourse

    Background: Laron syndrome, is a disease that treated by IGF-1. This protein is a single chain and has three disulfide bonds. People with Laron syndrome have low rates of cancer and diabetes, although they appear to be at increased risk of casual death due to their stature. IGF-1 is synthesized by many tissues and is secreted from liver as an endocrine hormone which is transmitted to other tissues. IGF-1 is responsible for cell differentiation, transformation, suppression of apoptosis, cell cycle progression, cell proliferation and differentiation. Expression system, the igf-1 gene, codon adaptation index (CAI) and GC contents are very critical for the large scale production of this protein. Current study was aimed to Optimization and Construction of human Insulin-like Growth Factor 1 Gene in E. coli DH5α. Material and Methods: we used DNA2 and ProtParam softwares for designing the best form to produce IGF-1. First the coding sequence was verified and then synthesized. For confirmation of the pUC18-IGF-1, sequencing test was carried out using M13 reverse primer. Finally it was inserted into the cloning site of pUC18. vector. Results: After coding optimization, the CAI rate was increased from 84 to 90 and GC content from 55.07 to 56.62. The presence of the band near 225bp resulted from enzymatic digestion with two restriction enzymes demonstrates the correct cloning of the recombinant vectors in the cloning site of pUC18 cloning vector. Conclusion: According to software and experimental analysis, the designed sequence probably in the best form could be used for production of recombinant protein

    زمینه و هدف: مطالعات مختلف نشان داده اند که خانم های باردار بدون توجه به تاثیرات تراتوژن بیشتر گیاهان دارویی به استفاده از این گیاهان می پردازند. از جمله این گیاهان که دارای خواص متعدد درمانی است، چای کوهی می باشد که تا به حال اثر تراتوژن آن بررسی نشده است. لذا این مطالعه با هدف بررسی تاثیر عصاره چای کوهی در غلظت های مختلف در ایجاد ناهنجاری در موش های سوری انجام شده است. روش بررسی: در این مطالعه تجربی، 60 سر موش سوری ماده بالغ پس از جفت گیری و مشاهده پلاک واژنی، به صورت تصادفی به شش گروه (دو گروه کنترل و 4 گروه مورد) تقسیم شدند. چهار گروه مورد از روز 7 تا 12 حاملگی به مدت 6 روز عصاره چای کوهی با غلظت های 50، 100، 150 و 200 میلی گرم بر کیلوگرم را به صورت داخل صفاقی دریافت کردند. در روز 18 حاملگی جنین ها از لوله رحمی خارج شدند و ابتدا وزن و قد آنها با ترازوی حساس دیجیتالی و کولیس اندازه گیری و سپس با رنگ الیزارین رد رنگ آمیزی و از نظر ناهنجاری های اسکلتی بررسی شدند. داده ها به کمک آزمون های آماری کای دو، کروسکال والیس و آزمون تعقیبی دان تجزیه و تحلیل شد. یافته ها: عصاره چای کوهی در دوزهای مختلف باعث اختلال در رشد (قد و وزن) جنین ها شد و این اختلال با افزایش دوز عصاره افزایش نشان داد (01/0

    Expression of recombinant human insulin-like growth factor type 1 (rhiGF-1) in Escherichia Coli

    Background: Human insulin-like growth factor type 1 (hIGF-1) is a protein consisting of 70 amino acids (MW=7.6 kDa) and mainly synthesized by liver. Mecasermin (Trade name INCRELEX) is the synthetic form of the protein which is used as an effective treatment for particular disorders such as short stature, type 1 and 2 diabetes, and wound healing. Current study was aimed to investigate the expression of human insulin-like growth factor type1 in Escherichia coli (E. coli) BL21 (DE3) expression system in order to produce an active recombinant form of the protein. Methods: For the purpose of the study, firstly codon optimization was done for hIGF-1 gene, using bioinformatics databases. Then, the gene was synthesized and inserted in pET-24a vector by a cutting strategy included NdeI and BamHI-HF enzymes. In the next step, gene was run in agarose gel and purified. The constructed expression cassette was transformed into E. coli BL21 (DE3) cells through CaCl2 heat shock method. Identification and confirmation of the transformed colonies were performed using screening PCR method. Synthesis of hIGF-1 was induced by IPTG. The expression in induced strains was analyzed by SDS-PAGE and western blotting techniques. Confirmation of cloning and IGF-1 expression cassette was carried out through genetic engineering procedures. Results: Analysis of transformed E. coli strain with SDS-PAGE and western blotting techniques confirmed that gene was expressed in host cells. Molecular weight of the expressed protein was estimated to be 7.6 kDa. Conclusion: hIGF-1 expression cassette for cloning and expression in E. coli was designed and the protein of interest was successfully induced and identified. In addition, E. coli BL21 (DE3) can be used as a suitable host for production of recombinant hIGF-1 and this technology has a potential to be localized. © 2015, Avicenna Journal of Medical Biotechnology.     Effect of Instructing Care Program Through Group Discussion on the Quality of Life of the Parents of the Children Afflicted With Leukemia

    Purpose: Children long-term involvement with cancer may have a negative impact on the quality of life their parents. Design and implementation of training programs for parents whose children have been diagnosed with leukemia, as the primary caregivers of children, will have a special significance and can contribute to better taking care of such children. The main purpose of the present study was to examine the impact of conducting group discussion, as care program training, on the quality of life parents whose children were suffering from leukemia. Methods: This quasi-experimental before-after intervention study encompassed two groups of parents (in total 41) of leukemia children. To collect data, a demographic questionnaire and the shortened version of SF-36 questionnaire were used to determine the quality of life of parents. Both groups completed the quality of life questionnaires before and two months after the intervention. Results: Comparison of the parents’ quality of life mean scores, obtained before and two months after training, showed that promotion in 6 domains of bodily pain, general health, emotional health, role limitation due to emotional problems, social functioning, and vitality were occurred. (P <0.05) Conclusions: Considering the important role of parents in taking care of children suffering from leukemia, introduction of care program training can be a positive step to help these parents and empower them to manage their children’s problems more systematically and will ultimately lead to improved quality of life of parents

    Abortificient effects of Stachys lavandulifolia Vahl in mice

    زمینه و هدف: مطالعات مختلف نشان داده اند که خانم ها بدون توجه به تاثیرات سقط آور و ناهنجاری زایی بیشتر گیاهان دارویی به استفاده از این گیاهان در دوران حاملگی می پردازند. از جمله این گیاهان که دارای خواص متعدد درمانی مثل اضطراب چای کوهی (Stachys lavandulifolia) است که تا به حال اثر سقط زایی آن بررسی نشده است. لذا این مطالعه با هدف بررسی تاثیر عصاره چای کوهی در غلظت های مختلف در ایجاد سقط در موش های سوری انجام شد. روش بررسی: در این مطالعه، 36 سر موش سوری بالغ ماده پس از جفت گیری و مشاهده پلاک واژنی، به صورت تصادفی به شش گروه تقسیم شدند. دو گروه به عنوان کنترل و چهار گروه به عنوان مورد که از روز 7 تا 12 حاملگی به مدت 6 روز عصاره چای کوهی با غلظت های 50، 100، 150 و 200 میلی گرم بر کیلوگرم به صورت داخل صفاقی دریافت کردند. در روز 16 حاملگی عمل سزارین انجام و تعداد جنین های جذب شده شمارش و به عنوان سقط در نظر گرفته شد. تجزیه و تحلیل داده ها با تست آماری کروسکال والیس و آزمون تعقیبی دان صورت گرفت. یافته ها: میانگین نسبت جنین های سقط شده در غلظت های 50، 100، 150 و 200 mg/kg به ترتیب 108/0±136/0، 151/0±263/0، 391/0±583/0 و 172/0±353/0 بود و در دو گروه کنترل جنین سقط شده مشاهده نشد. بر اساس آزمون کروسکال والیس اثرات سقط زایی گیاه وابسته به دوز است (05/0>P). نتیجه گیری: عصاره گیاه چای کوهی دارای خواص سقط آور می باشد و لازم است مصرف این گیاه در دوران بارداری با احتیاط صورت گیرد

    Transformation pathways of 2,2-dimethyloxirane on aluminosilicates: the effects of catalyst structure and reaction conditions

    The ring transformation reactions of 2,2-dimethyloxirane were studied over various aluminosilicates (HZSM-5, CuZSM-5, HY, AIMCM-41 or NaNa-loaded A1MCM-41) in a pulse reactor at 423 K. The main reactions identified were isomerisation and cyclic dimerisation. The product formation selectivities could be tuned by varying the pore size of the molecular sieves and/or altering the reaction parameters. Large pore size and mild conditions favoured isomerisation, while an optimally constrained environment and overloading the catalyst with the epoxide (increased pulse size) was advantageous for cyclic dimerisation

    Anterior Clinoid Process Mucocele: A Case Presenting with Sudden Vision Loss

    Background: Mucoceles are benign encapsulated lesions filled with mucus located in parasinus cavities. The occurrence rate of the anterior clinoid process is varying significantly in different studies. Visual disturbances, diplopia, and retrobulbar pain are among the common manifestations of anterior clinoid process mucocele. To our knowledge, 17&nbsp;cases of Anterior clinoid process mucocele with visual involvements have previously been reported. Aim: We aim to report a rare case of a middle-aged man with a large sphenoid mucocele admitted due to the loss of vision in his left eye. Case presentation: A 46-year-old man with a sudden vision loss in his left eye was admitted to Loghman Hakim Hospital. In physical examination positive relative afferent pupillary defect&nbsp;(RAPD) was evident. The visual acuity of his right eye was normal, while his left eye had no light perception. Moreover, his visual field was normal on the right side. He had no other medical complaints. Conclusion: Clinoid mucoceles causing cranial neuropathy are extremely rare. The proper diagnosis and prompt treatment are necessary to avoid permanent complications. With proper surgical intervention, recurrence of the mucoceles can be avoided

    Iron Oxide Nanoparticles Modified with Carbon Quantum Nanodots for the Stabilization of Palladium Nanoparticles: An Efficient Catalyst for the Suzuki Reaction in Aqueous Media under Mild Conditions

    Magnetic Fe3O4 nanoparticles (NPs) functionalized with carbon dots (C-dots) that contain carboxylic acid and hydroxyl groups were synthesized successfully and used for the reduction of PdII and the formation of Pd NPs. The new material was characterized by SEM, TEM, energy-dispersive spectroscopy, solid UV spectroscopy, vibrating sample magnetometry, XRD, and X-ray photoelectron spectroscopy and was used as a very efficient catalyst in the Suzuki–Miyaura cross-coupling reaction of aryl bromides and chlorides with arylboronic acids in aqueous media. Design of experiments indicates that the use of 0.22 mol % of Pd, K2CO3 as the base, and aqueous ethanol are the best reaction conditions. The reactions of aryl bromides take place at room temperature, and aryl chlorides react at 80 °C. The easily synthesized and air-stable catalyst Pd@C-dots@Fe3O4 NPs could be separated from the reaction mixture by using an external magnet and reused in eight consecutive runs with no significant loss of catalytic activity.The authors are grateful to the Institute for Advanced Studies in Basic Sciences (IASBS) and the Iran National Science Foundation (INSF-Grant number of 94010666) for support of this work.
