Formation of carbohydrate-functionalised polystyrene and glass slides and their analysis by MALDI-TOF MS
Glycans functionalised with hydrophobic trityl groups were synthesised and adsorbed onto polystyrene and glass slides in an array format. The adsorbed glycans could be analysed directly on these minimally conducting surfaces by MALDI-TOF mass spectrometry analysis after aluminium tape was attached to the underside of the slides. Furthermore, the trityl group appeared to act as an internal matrix and no additional matrix was necessary for the MS analysis. Thus, trityl groups can be used as simple hydrophobic, noncovalently linked anchors for ligands on surfaces and at the same time facilitate the in situ mass spectrometric analysis of such ligands
Possible earthquake trigger for 6th century mass wasting deposit at Lake Ohrid (Macedonia/Albania)
Lake Ohrid shared by the Republics of Albania
and Macedonia is formed by a tectonically active graben within the south Balkans and suggested to be the oldest lake in Europe. Several studies have shown that the lake provides a valuable record of climatic and environmental changes and a distal tephrostratigraphic record of volcanic eruptions from Italy. Fault structures identified in seismic data demonstrate that sediments have also the potential to record tectonic activity in the region. Here, we provide an example of linking seismic and sedimentological information with tectonic activity and historical documents. Historical documents indicate that a major earthquake destroyed the city of Lychnidus (today: city of Ohrid) in the early 6th century AD. Multichannel
seismic profiles, parametric sediment echosounder profiles, and a 10.08m long sediment record from the western part of the lake indicate a 2m thick mass wasting deposit, which is tentatively correlated with this earthquake. The mass wasting deposit is chronologically well constrained, as it directly overlays the AD472/AD 512 tephra. Moreover, radiocarbon dates and cross correlation with other sediment sequences
with similar geochemical characteristics of the Holocene indicate that the mass wasting event took place prior to the onset of the Medieval Warm Period, and is attributed it to one of the known earthquakes in the region in the early 6th century AD
The Social and Economic Impact of Sanctions and Time Limits on Recipients of Temporary Assistance to Needy Families
A central feature of the reforms enacted through the Personal Responsibility and Work Opportunity Reconciliation Act (welfare reform) has been the adoption of strategies to involuntarily remove Temporary Assistance to Needy Families (TANF) recipients from the welfare rolls, including increased use of sanctions and time limits on welfare receipt. Drawing on data from a three year panel study of women who had been receiving welfare in a state which adopted stringent sanctioning and time limit policies, we investigate predictors of recipients\u27 TANF status after implementation of welfare reform, and identify differences in post-reform material resources, hardships and quality of life based on TANF status. Almost half of all welfare case closures during the first time period after reforms were implemented through involuntary strategies. Relatively few baseline characteristics predicted different outcomes once welfare time limits and sanctions were implemented. Those who were timed off welfare had substantially lower incomes in the year following their removal. One third of all respondents, regardless of reason for leaving TANF reported having insufficient food, housing problems and lack of access to needed medical care
Challenging cisgenderism through trans people's narratives of domestic violence and abuse
By drawing on empirical research that explored trans people’s experiences of domestic violence and abuse (DVA), this paper problematises the ‘gender asymmetry debate' in DVA discourse. It does so by highlighting cisgenderism and a heteronormative bias which have led to the invisibility of a trans perspective. Qualitative data was collected via narrative interviews and this was examined using a voice-centred relational technique. A total of twenty four interviews were undertaken with trans people (n = 15) and domestic abuse practitioners (n = 9). In relation to the presentation and impact of DVA, and in the context of trans and cisgender people's abuse experiences, the research findings report both similarities and differences. Four narratives are presented here to illuminate both. This paper adds new insight and challenges normative and dominant discourses by promoting the need for further theorising about the gendered nature of domestic violence and abuse
Is Welfare Reform Working? A Study of the Effects of Sanctions on Families Receiving Temporary Assistance to Needy Families
This research provides a preliminary descriptive analysis of the impact of new welfare sanctions on recipients living in a southern metropolitan region. The data from this phone survey indicate that many families report considerable hardship no matter why they exited from welfare. Compared to those who left voluntarily, those who were sanctioned off welfare were significantly different in terms of having unmet medical needs, going without food, and having their utilities turned off. Given the high number of problems reported and the low income reported by these respondents, it is not surprising to find that only 10 percent of former recipients who were sanctioned off of welfare feel that they are better off now than when they received cash assistance. These problems can represent a significant disruption in the lives of children and their parents
The environmental and evolutionary history of Lake Ohrid (FYROM/Albania). Interim results from the SCOPSCO deep drilling project
This study reviews and synthesises existing information generated within the SCOPSCO (Scientific Collaboration on Past Speciation Conditions in Lake Ohrid) deep drilling project. The four main aims of the project are to infer (i) the age and origin of Lake Ohrid (Former Yugoslav Republic of Macedonia/Republic of Albania), (ii) its regional seismotectonic history, (iii) volcanic activity and climate change in the central northern Mediterranean region, and (iv) the influence of major geological events on the evolution of its endemic species. The Ohrid basin formed by transtension during the Miocene, opened during the Pliocene and Pleistocene, and the lake established de novo in the still relatively narrow valley between 1.9 and 1.3 Ma. The lake history is recorded in a 584 m long sediment sequence, which was recovered within the framework of the International Continental Scientific Drilling Program (ICDP) from the central part (DEEP site) of the lake in spring 2013. To date, 54 tephra and cryptotephra horizons have been found in the upper 460 m of this sequence. Tephrochronology and tuning biogeochemical proxy data to orbital parameters revealed that the upper 247.8 m represent the last 637 kyr. The multi-proxy data set covering these 637 kyr indicates long- term variability. Some proxies show a change from generally cooler and wetter to drier and warmer glacial and interglacial periods around 300 ka. Short-term environmental change caused, for example, by tephra deposition or the climatic impact of millennial-scale Dansgaard–Oeschger and Heinrich events are superimposed on the long-term trends. Evolutionary studies on the extant fauna indicate that Lake Ohrid was not a refugial area for regional freshwater animals. This differs from the surrounding catchment, where the mountainous setting with relatively high water availability provided a refuge for temperate and montane trees during the relatively cold and dry glacial periods. Although Lake Ohrid experienced significant environmental change over the last 637 kyr, preliminary molecular data from extant microgastropod species do not indicate significant changes in diversification rate during this period. The reasons for this constant rate remain largely unknown, but a possible lack of environ- mentally induced extinction events in Lake Ohrid and/or the high resilience of the ecosystems may have played a role
Molecular detection of thyroglobulin mRNA transcripts in peripheral blood of patients with thyroid disease by RT-PCR
The sensitive detection of circulating tumour cells in patients with differentiated thyroid cancer may precede the detection of relapse by other diagnostic studies – such as serum thyroglobulin – and thus may have important therapeutic and prognostic implications. We performed reverse transcription-polymerase chain reaction (RT-PCR) on blood samples from patients diagnosed with thyroid disease using two different RT-PCR sensitivities. Additionally, tissue specificity of TG mRNA-expression was determined using RNA extracts from 27 different human tissues. The lower limit of detection was 50–100 TG mRNA producing cells/ml blood using a ‘normal’ RT-PCR sensitivity and 10–20 cells/ml blood using a ‘high’ sensitivity. With the normal sensitivity TG mRNA was detected in 9/13 patients with thyroid cancer and metastasis, 63/137 patients with a history of thyroid cancer and no metastasis, 21/85 with non-malignant thyroid disease and 9/50 controls. With the high sensitivity TG mRNA was detected in 11/13 patients with thyroid cancer and metastasis, 111/137 patients with a history of thyroid cancer and no metastasis, 61/85 with non-malignant thyroid disease and 41/50 controls. Interestingly, using the normal RT-PCR sensitivity TG mRNA transcripts are specific for thyroid tissue and detectable in the peripheral blood of controls and patients with thyroid disease, which correlates with a diagnosis of metastasized thyroid cancer. However, with a high RT-PCR sensitivity, TG mRNA expression was found not to be specific for thyroid tissue and was not correlated with a diagnosis of thyroid cancer in patients. As a consequence, to date TG mRNA detected by RT-PCR in the peripheral blood cannot be recommended as a tumour marker superior to TG serum-level. © 2000 Cancer Research Campaig
A new type of Halimeda bioherm on the Queensland Plateau, NE Australia
Morphology, internal structure, and in situ facies distribution of mesophotic Halimeda bioherms from the Queensland Plateau (NE Australia) are presented based on hydroacoustic and oceanographic data, seafloor observations, and discrete sediment sampling carried out during RV SONNE cruise SO292 in 2022. Halimeda buildups consist of cone-like mounds up to 500 m in diameter and 3–10 m high, with gentle slopes (2°–5° on the top of Tregrosse Bank). Bioherms occur in water depths of 10–70 m, with most bioherm between 50 and 65 m. Their internal structure consists of aggrading low-amplitude reflections at the core of the bioherm interfingering with high-amplitude reflections to the flanks. Surface facies distribution displays one to four facies belts, from distal to proximal: Halimeda rudstone, Halimeda rudstone with living plants, Halimeda rudstone with coralgal debris, and coralgal boundstone (when present, occupied the top of the bioherms). It is proposed that the alternation of two key processes contributes to the formation of these bioherms: (1) in situ accumulation of Halimeda debris and (2) episodic dismantling of the mesophotic coralgal boundstone at the centre of the bioherm by severe storms. These storms may dismantle the mesophotic reef and export coralgal rubble to the flanks. Flanks may be recolonized by Halimeda during fair-weather periods. Due to their different geomorphic expressions, complex internal structure, and surficial facies distribution, we suggest that the buildups of the Queensland Plateau represent a new Halimeda bioherm morphotype, distinct from previously described bioherms on the adjacent Great Barrier Reef and elsewhere globally.03G0292A—ICECARB from the Bundesministerium für Bildung und

Postoperative Complications of True Dropless Cataract Surgery versus Standard Topical Drops
Purpose Compare postoperative outcomes in cataract surgery between eyes with standard drop regimen versus dropless protocol by residents.
Design Retrospective cohort study between April 1, 2018 and March 31, 2020.
Methods The study was performed at Lyndon B. Johnson General Hospital in Houston, Harris County, Texas. A total of 547 eyes (234 dropless vs. 313 standard) with phacoemulsification cataract surgery and minimum of 1-month follow-up with best-corrected visual acuity (BCVA) were included. Dropless received 40 mg sub-Tenon\u27s triamcinolone and intracameral moxifloxacin. Patients were followed at postoperative day 1 (POD1), week 1 (POW1), and month 1 (POM1). Postoperative rate of BCVA better than 20/40 (Good vision) and rate of complications were compared between groups.
Results Good vision on POM1 in dropless (77.8%) was noninferior to standard (75.1%, p = 0.80). Complication rate in dropless (28.6%) was noninferior to standard (24.0%, p = 0.13). Intraocular pressure (IOP) elevation on POD1 ( p = 0.041) and anterior chamber (AC) cells on POW1 and POM1 ( p \u3c 0.001) were more frequent in dropless. Mean spherical equivalent at POM1 was better in dropless (−0.37 D [±0.81 D]) compared with standard (−0.61D [±0.77 D], p = 0.001). Early posterior capsular opacification (early PCO) was more frequent in dropless ( p = 0.042).
Conclusions Postoperative rate of BCVA better than 20/40 and rate of postoperative complications were noninferior, although dropless had higher rates of AC inflammation, IOP elevation, and early PCO
Dismantling of an isolated tropical carbonate platform through flank collapse and canyon erosion, Coral Sea, Northeast Australia
The steep slopes of carbonate platforms frequently display large-scale sediment destabilization features like rockfalls, mass transport complexes, and slope erosion. The processes and factors triggering such instabilities and how they interact are a matter of ongoing discussion.
We use hydroacoustic, sedimentological, and seafloor imaging data to map and characterize slope instabilities and potential controlling factors at the flank of the isolated Tregrosse carbonate bank in the Coral Sea, northeast Australia.
Erosion of gullies and submarine valleys is concentrated in slope segments with the platform rim at several 10s of meters of water depth, i.e. where there is potential for sediment transfer from the bank interior to the slope. Gravity core data indicate that most sediment export from the platform occurs during sea-level fall.
The toe of slopes neighboring segments with a shallower platform rim are mostly characterized by mass-transport complexes of platform rim and upper slope rocks forming extended block fields. Distal slope areas are dismantled through submarine landslides resulting in scalloped head scarps.
The basal detachment surface of these submarine landslides appears to be rooted in several 100 s of meters in the subsurface at a lithological heterogeneity, which is documented by a gamma-ray peak in the downhole logging data from Ocean Drilling Program Site 817.
Our findings show that (1) canyon erosion, (2) platform rim and upper slope destabilization as well as (3) lower slope dismantling, largely act independently of each other to destabilize the flanks of the carbonate bank. The complexity of the carbonate platform dismantling processes and the corresponding controlling factors shown in this study should also be considered when interpreting seismic morphological data
