Acclimation to Elevated CO2 Increases Constitutive Glucosinolate Levels of Brassica Plants and Affects the Performance of Specialized Herbivores from Contrasting Feeding Guilds
Plants growing under elevated CO2 concentration may acclimate by modifying chemical traits. Most studies have focused on the effects of environmental change on plant growth and productivity. Potential effects on chemical traits involved in resistance, and the consequences of such effects on plant-insect interactions, have been largely neglected. Here, we evaluated the performance of two Brassica specialist herbivores from contrasting feeding guilds, the leaf-feeding Pieris brassicae and the phloem-feeding Brevicoryne brassicae, in response to potential CO2-mediated changes in primary and major secondary metabolites (glucosinolates) in Brassica oleracea. Plants were exposed to either ambient (400ppm) or elevated (800ppm) CO2 concentrations for 2, 6, or 10weeks. Elevated CO2 did not affect primary metabolites, but significantly increased glucosinolate content. The performance of both herbivores was significantly reduced under elevated CO2 suggesting that CO2-mediated increases in constitutive defense chemistry could benefit plants. However, plants with up-regulated defenses could also be subjected to intensified herbivory by some specialized herbivores, due to a chemically-mediated phagostimulatory effect, as documented here for P. brassicae larvae. Our results highlight the importance of understanding acclimation and responses of plants to the predicted increases in atmospheric CO2 concentrations and the concomitant effects of these responses on the chemically-mediated interactions between plants and specialized herbivore
Spectral and localization properties of the Dirichlet wave guide with two concentric Neumann discs
Bound states of the Hamiltonian describing a quantum particle living on three
dimensional straight strip of width are investigated. We impose the Neumann
boundary condition on the two concentric windows of the radii and
located on the opposite walls and the Dirichlet boundary condition on the
remaining part of the boundary of the strip. We prove that such a system
exhibits discrete eigenvalues below the essential spectrum for any .
When and tend to the infinity, the asymptotic of the eigenvalue is
derived. A comparative analysis with the one-window case reveals that due to
the additional possibility of the regulating energy spectrum the anticrossing
structure builds up as a function of the inner radius with its sharpness
increasing for the larger outer radius. Mathematical and physical
interpretation of the obtained results is presented; namely, it is derived that
the anticrossings are accompanied by the drastic changes of the wave function
localization. Parallels are drawn to the other structures exhibiting similar
phenomena; in particular, it is proved that, contrary to the two-dimensional
geometry, at the critical Neumann radii true bound states exist.Comment: 25 pages, 7 figure

Nonlinear Dynamics of Silicon Nanowire Resonator Considering Nonlocal Effect
In this work, nonlinear dynamics of silicon nanowire resonator considering nonlocal effect has been investigated. For the first time, dynamical parameters (e.g., resonant frequency, Duffing coefficient, and the damping ratio) that directly influence the nonlinear dynamics of the nanostructure have been derived. Subsequently, by calculating their response with the varied nonlocal coefficient, it is unveiled that the nonlocal effect makes more obvious impacts at the starting range (from zero to a small value), while the impact of nonlocal effect becomes weaker when the nonlocal term reaches to a certain threshold value. Furthermore, to characterize the role played by nonlocal effect in exerting influence on nonlinear behaviors such as bifurcation and chaos (typical phenomena in nonlinear dynamics of nanoscale devices), we have calculated the Lyapunov exponents and bifurcation diagram with and without nonlocal effect, and results shows the nonlocal effect causes the most significant effect as the device is at resonance. Horizontal DNA transfer mechanisms of bacteria as weapons of intragenomic conflict
Horizontal DNA transfer (HDT) is a pervasive mechanism of diversification in many microbial species, but its primary evolutionary role remains controversial. Much recent research has emphasised the adaptive benefit of acquiring novel DNA, but here we argue instead that intragenomic conflict provides a coherent framework for understanding the evolutionary origins of HDT. To test this hypothesis, we developed a mathematical model of a clonally descended bacterial population undergoing HDT through transmission of mobile genetic elements (MGEs) and genetic transformation. Including the known bias of transformation toward the acquisition of shorter alleles into the model suggested it could be an effective means of counteracting the spread of MGEs. Both constitutive and transient competence for transformation were found to provide an effective defence against parasitic MGEs; transient competence could also be effective at permitting the selective spread of MGEs conferring a benefit on their host bacterium. The coordination of transient competence with cell-cell killing, observed in multiple species, was found to result in synergistic blocking of MGE transmission through releasing genomic DNA for homologous recombination while simultaneously reducing horizontal MGE spread by lowering the local cell density. To evaluate the feasibility of the functions suggested by the modelling analysis, we analysed genomic data from longitudinal sampling of individuals carrying Streptococcus pneumoniae. This revealed the frequent within-host coexistence of clonally descended cells that differed in their MGE infection status, a necessary condition for the proposed mechanism to operate. Additionally, we found multiple examples of MGEs inhibiting transformation through integrative disruption of genes encoding the competence machinery across many species, providing evidence of an ongoing "arms race." Reduced rates of transformation have also been observed in cells infected by MGEs that reduce the concentration of extracellular DNA through secretion of DNases. Simulations predicted that either mechanism of limiting transformation would benefit individual MGEs, but also that this tactic's effectiveness was limited by competition with other MGEs coinfecting the same cell. A further observed behaviour we hypothesised to reduce elimination by transformation was MGE activation when cells become competent. Our model predicted that this response was effective at counteracting transformation independently of competing MGEs. Therefore, this framework is able to explain both common properties of MGEs, and the seemingly paradoxical bacterial behaviours of transformation and cell-cell killing within clonally related populations, as the consequences of intragenomic conflict between self-replicating chromosomes and parasitic MGEs. The antagonistic nature of the different mechanisms of HDT over short timescales means their contribution to bacterial evolution is likely to be substantially greater than previously appreciated
Spalling uniaxial strength of Al2O3 at high strain rates
In this article research into the uniaxial tensile strength of Al2O3 monolithic ceramic is presented. The experimental procedure of the spalling of long bars is investigated from different approaches. This method is used to obtain the tensile strength at high strain rates under uniaxial conditions. Different methodologies proposed by several authors are used to obtain the tensile strength. The hypotheses needed for the experimental set-up are also checked, and the requirements of the set-up and the variables are also studied by means of numerical simulations. The research shows that the shape of the projectile is crucial to achieve successfully tests results. An experimental campaign has been carried out including high speed video and a digital image correlation system to obtain the tensile strength of alumina. Finally, a comparison of the test results provided by three different methods proposed by different authors is presented. The tensile strength obtained from the three such methods on the same specimens provides contrasting results. Mean values vary from one method to another but the trends are similar for two of the methods. The third method gives less scatter, though the mean values obtained are lower and do not follow the same trend as the other methods for the different specimens
Efficient coupler between silicon photonic and metal-insulator-silicon-metal plasmonic waveguides
We report the experimental realization of a compact, efficient coupler between silicon waveguides and vertical metal-insulator-silicon-metal (MISM) plasmonic waveguides. Devices were fabricated using complementary metal-oxide-silicon technology processes, with copper layers that support low-loss plasmonic modes in the MISM structures at a wavelength of 1550 nm. By implementing a short (0.5 μm) optimized metal-insulator-silicon-insulator structure inserted between the photonic and plasmonic waveguide sections, we demonstrate experimental coupling loss of 2.5 dB, despite the high optical confinement of the MISM mode and mismatch with the silicon waveguide mode
Preliminary evaluation of fenugreek (Trigonella foenum-graecum) seed gum as a potential prebiotic for growing rabbits in Tunisia: effects on in vivo faecal digestibility and in vitro fermentation
[EN] This study aims to determine the effect of dietary inclusion of fenugreek seed gum (FSG), rich in galactomannans, on nutrient apparent digestibility and caecal environment, as well as on in vitro caecal fermentation of Tunisian growing rabbits. Three experimental diets were formulated, including 0, 0.25 and 0.5% of FSG (FSG0, FSG0.25 and FSG0.5, respectively) for the in vivo trial and 0, 0.125, 0.25, 0.5 and 100% of FSG (FSG0, FSG0125, FSG0.25, FSG0.5 and FSG100, respectively) for the in vitro trial. In the in vivo trial, 45 weaned rabbits 31 d old (15 per treatment) were housed in individual cages until 94 d of age. Apparent digestibility coefficients were determined at two ages, from 38 to 41 and from 56 to 59 d old, and caecal traits were recorded after slaughtering. In the in vitro trial, the five experimental diets were incubated with a rabbit caecal inoculum. Gas production was measured and modelled until 72 h and the fermentation traits were measured. Apparent faecal digestibility coefficients of main nutrients and main caecal environment traits were not significantly affected by the dietary inclusion of FSG (P>0.05). However, animals fed with FSG showed lower caecal pH (–0.15; P<0.05) values. Regarding the in vitro fermentation, FSG100 increased asymptotic gas production (+11.25, P<0.001), sharpness of the switching characteristic of the profile (+1.98, P<0.001) and the maximum substrate degradation rate (RM) (+0.188, P<0.001), but decreasing the time after incubation at which half of the asymptotic amount of gas has been formed (–5.86, P<0.001) and at which RM occurs (–4.53, P<0.01). Likewise, FSG100 significantly decreased caecal pH (–1.035, P<0.001), lactic acid (–9.51, P<0.069) and N-NH3 concentrations (–12.81, P<0.001). Meanwhile, it increased the total volatile fatty acids (VFA) production (+43.15, P<0.001). Gradual dietary inclusion of FSG from 0 to 0.5% only significantly increased total VFA production in the caecum (+100 mmol/L per percentage point of FSG inclusion; P<0.05). In conclusion, FSG is highly and rapidly in vitro fermented by rabbit caecal bacteria. Mesenchymal stem cell-conditioned medium reduces disease severity and immune responses in inflammatory arthritis
We evaluated the therapeutic potential of mesenchymal stem cell-conditioned medium (CM-MSC) as an alternative to cell therapy in an antigen-induced model of arthritis (AIA). Disease severity and cartilage loss were evaluated by histopathological analysis of arthritic knee joints and immunostaining of aggrecan neoepitopes. Cell proliferation was assessed for activated and naïve CD4+ T cells from healthy mice following culture with CM-MSC or co-culture with MSCs. T cell polarization was analysed in CD4+ T cells isolated from spleens and lymph nodes of arthritic mice treated with CM-MSC or MSCs. CM-MSC treatment significantly reduced knee-joint swelling, histopathological signs of AIA, cartilage loss and suppressed TNFα induction. Proliferation of CD4+ cells from spleens of healthy mice was not affected by CM-MSC but reduced when cells were co-cultured with MSCs. In the presence of CM-MSC or MSCs, increases in IL-10 concentration were observed in culture medium. Finally, CD4+ T cells from arthritic mice treated with CM-MSC showed increases in FOXP3 and IL-4 expression and positively affected the Treg:Th17 balance in the tissue. CM-MSC treatment reduces cartilage damage and suppresses immune responses by reducing aggrecan cleavage, enhancing Treg function and adjusting the Treg:Th17 ratio. A non-APOE polygenic risk score for Alzheimer's disease is associated with CSF neurofilament light in a representative sample of cognitively unimpaired 70-year-olds
The effect of Alzheimer's disease (AD) polygenic risk scores (PRSs) on amyloid and tau pathophysiology and neurodegeneration in cognitively unimpaired older adults is not known in detail. This study aims to investigate non-APOE AD-PRS and APOE ε4 in relation to AD pathophysiology evaluated by cerebrospinal fluid (CSF) biomarkers in a population-based sample of 70-year-olds. A total of 303 dementia-free individuals from the Gothenburg H70 Birth Cohort Studies were included. Genotyping was performed using the NeuroChip, and AD-PRSs were calculated. CSF levels of amyloid-β (Aβ42), total tau (t-tau), phosphorylated tau (p-tau), neurogranin (Ng), and neurofilament light (NfL) were measured with ELISA. Associations were found between non-APOE PRS and both NfL (p=0.001) and Aβ42 (p=0.02), and between APOE ε4 and Aβ42 (p=1e-10), t-tau (p=5e-4), and p-tau (p=0.002). Similar results were observed when only including individuals with CDR=0, except for no evidence of an association between non-APOE PRS and Aβ42. There was an interaction between non-APOE PRS and Aβ42 pathology status in relation to NfL (p=0.005); association was only present in individuals without Aβ42 pathology (p=0.0003). In relation to Aβ42, there was a borderline interaction (p=0.06) between non-APOE PRS and APOE ε4; association was present in ε4 carriers only (p=0.03). Similar results were observed in individuals with CDR=0 (n=246). In conclusion, among cognitively healthy 70-year-olds from the general population genetic risk of AD beyond the APOE locus was associated with NfL in individuals without Aβ42 pathology, and with Aβ42 in APOE ε4 carriers, suggesting these associations are driven by different mechanisms
