Automatic differentiation of u- and n-serrated patterns in direct immunofluorescence images
Epidermolysis bullosa acquisita (EBA) is a subepidermal autoimmune blistering disease of the skin. Manual u- and n-serrated patterns analysis in direct immunofluorescence (DIF) images is used in medical practice to differentiate EBA from other forms of pemphigoid. The manual analysis of serration patterns in DIF images is very challenging, mainly due to noise and lack of training of the immunofluorescence (IF) microscopists. There are no automatic techniques to distinguish these two types of serration patterns. We propose an algorithm for the automatic recognition of such a disease. We first locate a region where u- and n-serrated patterns are typically found. Then, we apply a bank of B-COSFIRE filters to the identified region of interest in the DIF image in order to detect ridge contours. This is followed by the construction of a normalized histogram of orientations. Finally, we classify an image by using the nearest neighbors algorithm that compares its normalized histogram of orientations with all the images in the dataset. The best results that we achieve on the UMCG publicly available data set is 84.6% correct classification, which is comparable to the results of medical experts
Investigation of the midgut structure and ultrastructure in Cimex lectularius and Cimex pipistrelli (Hemiptera, Cimicidae)
Cimicidae are temporary ectoparasites, which means that they cannot obtain food continuously. Both Cimex species examined here, Cimex lectularius (Linnaeus 1758) and Cimex pipistrelli (Jenyns 1839), can feed on a non-natal host, C. lectularius from humans on bats, C. pipistrelli on humans, but never naturally. The midgut of C. lectularius and C. pipistrelli is composed of three distinct regions—the anterior midgut (AMG), which has a sack-like shape, the long tube-shaped middle midgut (MMG), and the posterior midgut (PMG). The different ultrastructures of the AMG, MMG, and PMG in both of the species examined suggest that these regions must fulfill different functions in the digestive system. Ultrastructural analysis showed that the AMG fulfills the role of storing food and synthesizing and secreting enzymes, while the MMG is the main organ for the synthesis of enzymes, secretion, and the storage of the reserve material. Additionally, both regions, the AMG and MMG, are involved in water absorption in the digestive system of both Cimex species. The PMG is the part of the midgut in which spherites accumulate. The results of our studies confirm the suggestion of former authors that the structure of the digestive tract of insects is not attributed solely to diet but to the basic adaptation of an ancestor

centrioles result in multipolar mitosis and genomic instability. Normally, just one
daughter centriole forms on each mature (mother) centriole; however, a mother
centriole can produce multiple daughters within a single cell cycle. The
mechanisms that prevent centriole ‘overduplication’ are poorly understood. Here we
use laser microsurgery to test the hypothesis that attachment of the daughter
centriole to the wall of the mother inhibits formation of additional daughters. We
show that physical removal of the daughter induces reduplication of the mother in Sarrested
cells. Under conditions when multiple daughters simultaneously form on a
single mother, all of these daughters must be removed to induce reduplication.
Intriguingly, the number of daughter centrioles that form during reduplication does
not always match the number of ablated daughter centrioles. We also find that
exaggeration of the pericentriolar material (PCM) via overexpression of the PCM
protein pericentrin in S-arrested CHO cells induces formation of numerous daughter
centrioles. We propose that that the size of the PCM cloud associated with the
mother centriole restricts the number of daughters that can form simultaneously.This work was supported

Gut Microbial Gene Expression in Mother-Fed and Formula-Fed Piglets
Effects of diet on the structure and function of gut microbial communities in newborn infants are poorly understood. High-resolution molecular studies are needed to definitively ascertain whether gut microbial communities are distinct in milk-fed and formula-fed infants.Pyrosequencing-based whole transcriptome shotgun sequencing (RNA-seq) was used to evaluate community wide gut microbial gene expression in 21 day old neonatal piglets fed either with sow's milk (mother fed, MF; n = 4) or with artificial formula (formula fed, FF; n = 4). Microbial DNA and RNA were harvested from cecal contents for each animal. cDNA libraries and 16S rDNA amplicons were sequenced on the Roche 454 GS-FLX Titanium system. Communities were similar at the level of phylum but were dissimilar at the level of genus; Prevotella was the dominant genus within MF samples and Bacteroides was most abundant within FF samples. Screened cDNA sequences were assigned functional annotations by the MG-RAST annotation pipeline and based upon best-BLASTX-hits to the NCBI COG database. Patterns of gene expression were very similar in MF and FF animals. All samples were enriched with transcripts encoding enzymes for carbohydrate and protein metabolism, as well as proteins involved in stress response, binding to host epithelium, and lipopolysaccharide metabolism. Carbohydrate utilization transcripts were generally similar in both groups. The abundance of enzymes involved in several pathways related to amino acid metabolism (e.g., arginine metabolism) and oxidative stress response differed in MF and FF animals.Abundant transcripts identified in this study likely contribute to a core microbial metatranscriptome in the distal intestine. Although microbial community gene expression was generally similar in the cecal contents of MF and FF neonatal piglets, several differentially abundant gene clusters were identified. Further investigations of gut microbial gene expression will contribute to a better understanding of normal and abnormal enteric microbiology in animals and humans
