A portable magnetic field of >3 T generated by the flux jump assisted, pulsed field magnetization of bulk superconductors
A trapped magnetic field of greater than 3 T has been achieved in a single grain GdBaCuO (GdBaCuO) bulk superconductor of diameter 30 mm by employing pulsed field magnetization. The magnet system is portable and operates at temperatures between 50 K and 60 K. Flux jump behaviour was observed consistently during magnetization when the applied pulsed field, , exceeded a critical value (e.g., 3.78 T at 60 K). A sharp d/d is essential to this phenomenon. This flux jump behaviour enables the magnetic flux to penetrate fully to the centre of the bulk superconductor, resulting in full magnetization of the sample without requiring an applied field as large as that predicted by the Bean model. We show that this flux jump behaviour can occur over a wide range of fields and temperatures, and that it can be exploited in a practical quasi-permanent magnet system.This work was supported by the Boeing Company and by the Engineering and Physical Sciences Research Council (Grant No. EP/P00962X/1

Childbirth and consequent atopic disease: emerging evidence on epigenetic effects based on the hygiene and EPIIC hypotheses
Background: In most high and middle income countries across the world, at least 1:4 women give birth by cesarean
section. Rates of labour induction and augmentation are rising steeply; and in some countries up to 50 % of laboring
women and newborns are given antibiotics. Governments and international agencies are increasingly concerned about
the clinical, economic and psychosocial effects of these interventions.
Discussion: There is emerging evidence that certain intrapartum and early neonatal interventions might affect the
neonatal immune response in the longer term, and perhaps trans-generationally. Two theories lead the debate in this
area. Those aligned with the hygiene (or ‘Old Friends’) hypothesis have examined the effect of gut microbiome colonization
secondary to mode of birth and intrapartum/neonatal pharmacological interventions on immune response and epigenetic
phenomena. Those working with the EPIIC (Epigenetic Impact of Childbirth) hypothesis are concerned with the effects of
eustress and dys-stress on the epigenome, secondary to mode of birth and labour interventions.
Summary: This paper examines the current and emerging findings relating to childbirth and atopic/autoimmune
disease from the perspective of both theories, and proposes an alliance of research effort. This is likely to accelerate
the discovery of important findings arising from both approaches, and to maximize the timely understanding of the
longer-term consequences of childbirth practices

Essential Domains of Anaplasma phagocytophilum Invasins Utilized to Infect Mammalian Host Cells
Anaplasma phagocytophilum causes granulocytic anaplasmosis, an emerging disease of humans and domestic animals. The obligate intracellular bacterium uses its invasins OmpA, Asp14, and AipA to infect myeloid and non-phagocytic cells. Identifying the domains of these proteins that mediate binding and entry, and determining the molecular basis of their interactions with host cell receptors would significantly advance understanding of A. phagocytophilum infection. Here, we identified the OmpA binding domain as residues 59 to 74. Polyclonal antibody generated against a peptide spanning OmpA residues 59 to 74 inhibited A. phagocytophilum infection of host cells and binding to its receptor, sialyl Lewis x (sLex-capped P-selectin glycoprotein ligand 1. Molecular docking analyses predicted that OmpA residues G61 and K64 interact with the two sLex sugars that are important for infection, α2,3-sialic acid and α1,3-fucose. Amino acid substitution analyses demonstrated that K64 was necessary, and G61 was contributory, for recombinant OmpA to bind to host cells and competitively inhibit A. phagocytophilum infection. Adherence of OmpA to RF/6A endothelial cells, which express little to no sLex but express the structurally similar glycan, 6-sulfo-sLex, required α2,3-sialic acid and α1,3-fucose and was antagonized by 6-sulfo-sLex antibody. Binding and uptake of OmpA-coated latex beads by myeloid cells was sensitive to sialidase, fucosidase, and sLex antibody. The Asp14 binding domain was also defined, as antibody specific for residues 113 to 124 inhibited infection. Because OmpA, Asp14, and AipA each contribute to the infection process, it was rationalized that the most effective blocking approach would target all three. An antibody cocktail targeting the OmpA, Asp14, and AipA binding domains neutralized A. phagocytophilumbinding and infection of host cells. This study dissects OmpA-receptor interactions and demonstrates the effectiveness of binding domain-specific antibodies for blocking A. phagocytophilum infection
Detection of equine herpesvirus-4 and physiological stress patterns in young Thoroughbreds consigned to a South African auction sale
BACKGROUND : The prevalence of equine herpesvirus types-1 and -4 (EHV-1 and -4) in South African Thoroughbreds
at auction sales is currently undefined. Commingling of young Thoroughbreds from various populations together
with physiological stress related to their transport and confinement at a sales complex, may be associated with
shedding and transmission of EHV-1 and -4. This prospective cohort study sampled 90 young Thoroughbreds
consigned from eight farms, originating from three provinces representative of the South African Thoroughbred
breeding demographic to a sales complex. Nasal swabs for quantitative real-time polymerase chain reaction
(qPCR) assay to detect EHV-1 and -4 nucleic acid and blood samples for enzyme-linked immunosorbent assay
for EHV-1 and -4 antibodies were collected from all horses on arrival and departure. Additional nasal swabs for
qPCR were obtained serially from those displaying pyrexia and, or nasal discharge. Daily faecal samples were
used for determination of faecal glucocorticoid metabolite (FGM) concentrations as a measurement of physiological
stress and these values were modelled to determine the factors best explaining FGM variability.
RESULTS : EHV-4 nucleic acid was detected in 14.4 % and EHV-1 from none of the animals in the study population.
Most (93.3 %) and very few (1.1 %) of this population showed antibodies indicating prior exposure to EHV-4 and
EHV-1 respectively. Pyrexia and nasal discharge were poor predictors for detecting EHV-4 nucleic acid. The horses’
FGM concentrations increased following arrival before decreasing for most of the remaining study period including the
auction process. Model averaging showed that variation in FGM concentrations was best explained by days post-arrival
and transport duration.
CONCLUSIONS : In this study population, sales consignment was associated with limited detection of EHV-4 nucleic
acid in nasal secretions, with most showing prior exposure to EHV-4 and very few to EHV-1. The physiological stress
response shown by most reflected the combination of stressors associated with transport and arrival and these are
key areas for future investigation into management practices to enhance health and welfare of young Thoroughbreds

Male urine signals social rank in the Mozambique tilapia (Oreochromis mossambicus)
<p>Abstract</p> <p>Background</p> <p>The urine of freshwater fish species investigated so far acts as a vehicle for reproductive pheromones affecting the behaviour and physiology of the opposite sex. However, the role of urinary pheromones in intra-sexual competition has received less attention. This is particularly relevant in lek-breeding species, such as the Mozambique tilapia (<it>Oreochromis mossambicus</it>), where males establish dominance hierarchies and there is the possibility for chemical communication in the modulation of aggression among males. To investigate whether males use urine during aggressive interactions, we measured urination frequency of dye-injected males during paired interactions between size-matched males. Furthermore, we assessed urinary volume stored in the bladder of males in a stable social hierarchy and the olfactory potency of their urine by recording of the electro-olfactogram.</p> <p>Results</p> <p>Males released urine in pulses of short duration (about one second) and markedly increased urination frequency during aggressive behaviour, but did not release urine whilst submissive. In the stable hierarchy, subordinate males stored less urine than males of higher social rank; the olfactory potency of the urine was positively correlated with the rank of the male donor.</p> <p>Conclusion</p> <p>Dominant males store urine and use it as a vehicle for odorants actively released during aggressive disputes. The olfactory potency of the urine is positively correlated with the social status of the male. We suggest that males actively advertise their dominant status through urinary odorants which may act as a 'dominance' pheromone to modulate aggression in rivals, thereby contributing to social stability within the lek.</p
