Parental stress increases body mass index trajectory in pre-adolescents.
What is already known about this subjectRates of childhood obesity have increased since the mid-1970s. Research into behavioural determinants has focused on physical inactivity and unhealthy diets. Cross-sectional studies indicate an association between psychological stress experienced by parents and obesity in pre-adolescents.What this study addsWe provide evidence of a prospective association between parental psychological stress and increased weight gain in pre-adolescents. Family-level support for those experiencing chronic stress might help promote healthy diet and exercise behaviours in children.ObjectiveWe examined the impact of parental psychological stress on body mass index (BMI) in pre-adolescent children over 4 years of follow-up.MethodsWe included 4078 children aged 5-10 years (90% were between 5.5 and 7.5 years) at study entry (2002-2003) in the Children's Health Study, a prospective cohort study in southern California. A multi-level linear model simultaneously examined the effect of parental stress at study entry on the attained BMI at age 10 and the slope of change across annual measures of BMI during follow-up, controlled for the child's age and sex. BMI was calculated based on objective measurements of height and weight by trained technicians following a standardized procedure.ResultsA two standard deviation increase in parental stress at study entry was associated with an increase in predicted BMI attained by age 10 of 0.287 kg m(-2) (95% confidence interval 0.016-0.558; a 2% increase at this age for a participant of average attained BMI). The same increase in parental stress was also associated with an increased trajectory of weight gain over follow-up, with the slope of change in BMI increased by 0.054 kg m(-2) (95% confidence interval 0.007-0.100; a 7% increase in the slope of change for a participant of average BMI trajectory).ConclusionsWe prospectively demonstrated a small effect of parental stress on BMI at age 10 and weight gain earlier in life than reported previously. Interventions to address the burden of childhood obesity should address the role of parental stress in children
Traffic-related air pollution and obesity formation in children: a longitudinal, multilevel analysis.
BackgroundBiologically plausible mechanisms link traffic-related air pollution to metabolic disorders and potentially to obesity. Here we sought to determine whether traffic density and traffic-related air pollution were positively associated with growth in body mass index (BMI = kg/m2) in children aged 5-11 years.MethodsParticipants were drawn from a prospective cohort of children who lived in 13 communities across Southern California (N = 4550). Children were enrolled while attending kindergarten and first grade and followed for 4 years, with height and weight measured annually. Dispersion models were used to estimate exposure to traffic-related air pollution. Multilevel models were used to estimate and test traffic density and traffic pollution related to BMI growth. Data were collected between 2002-2010 and analyzed in 2011-12.ResultsTraffic pollution was positively associated with growth in BMI and was robust to adjustment for many confounders. The effect size in the adjusted model indicated about a 13.6% increase in annual BMI growth when comparing the lowest to the highest tenth percentile of air pollution exposure, which resulted in an increase of nearly 0.4 BMI units on attained BMI at age 10. Traffic density also had a positive association with BMI growth, but this effect was less robust in multivariate models.ConclusionsTraffic pollution was positively associated with growth in BMI in children aged 5-11 years. Traffic pollution may be controlled via emission restrictions; changes in land use that promote jobs-housing balance and use of public transit and hence reduce vehicle miles traveled; promotion of zero emissions vehicles; transit and car-sharing programs; or by limiting high pollution traffic, such as diesel trucks, from residential areas or places where children play outdoors, such as schools and parks. These measures may have beneficial effects in terms of reduced obesity formation in children

Governance Indicators Can Make Sense: Under-five Mortality Rates are an Example
Governance indicators have come under fire in recent years, especially the World Governance Indicators (WGIs). Critics present these indicators as a-theoretical and biased. Critics of the critics counter that no better alternatives exist. We suggest otherwise, arguing that more appropriate ‘governance’ indicators will (i) have theoretical grounding, (ii) focus on specific fields of engagement, (iii) emphasize outcomes, and (iv) control for key contextual differences in comparing countries. Such measures can help indicate where countries seem to have governance problems, allowing second stage analyses of what these problems are. We present under national five mortality rates adjusted for country income groups as an example of such measure, presenting data for contextually controlled outcomes in this specific field to show where governance seems better and worse. The United States is shown up as relatively weak, whereas a country like Pakistan seems to have better governance in this sector than other low income countries. The indicator allows questions about why governance of this sector might be problematic in certain contexts and easier in others
A paleoclimatic and paleoatmospheric record from peatlands accumulating during the Cretaceous-Paleogene boundary event, Western Interior Basin, Canada
Petrological and δ13C analyses were undertaken on contiguous specimens of coal and intercalated minor organic-rich clastic sediments collected from coal seams spanning the Cretaceous-Paleogene boundary in the Albertaand Saskatchewan portions of the Western Interior Basin. The generally high smectite content of the coal suggests that the original mires were largely small, disconnected, and rheotrophic, readily receiving abundant waterborne detrital clastic material of largely volcanic origin. Nevertheless, using the distinctive claystone that marks the Cretaceous-Paleogene boundary as a regional datum, it is possible to correlate cycles in the vitrinite and inertinite composition of the coals over >500 km. Estimates of peat accumulation rates suggest that the cycles in vitrinite and inertinite composition represent regional, cyclic fluctuations in wildfire and oxidation of the peatlands and overlying canopy at a frequency of hundreds to thousands of years. The likely causes of these fluctuations were cyclic, regional-scale changes in temperature. The Cretaceous- Paleogene boundary event occurred early during a phase of gradually increasing temperature and/or decreasing rainfall, but peak wildfire and desiccation of peat occurred up to 14,000 yr later than the Cretaceous- Paleogene boundary, and the mires did not experience significant water stress in the immediate aftermath of the extinction event. A persistent, 1.5‰-3.0‰ negative δ13C excursion occurs across the Cretaceous-Paleogene boundary, but it cannot be readily separated from four, further negative excursions later in the earliest Danian. The negative carbon isotope excursion linked to the Cretaceous- Paleogene boundary began a few hundred years before the event itself, and recovery occurred within 21 k.y., and possibly in as little as just a few thousand years, consistent with recently calibrated shallow-marine δ13C records. Hence, the atmospheric and surface ocean carbon pools were coupled at this time. The absence of evidence for catastrophic change in the climatic regime at the time of the Cretaceous-Paleogene extinction in these mires supports the notion that the negative shift in atmospheric δ13C was brought about by changes in the δ13C composition of the surface ocean. This is consistent with the greater magnitude of extinction experienced by marine fauna relative to the terrestrial realm.</p

Air Pollution Exposure Assessment for Epidemiologic Studies of Pregnant Women and Children: Lessons Learned from the Centers for Children’s Environmental Health and Disease Prevention Research
The National Children’s Study is considering a wide spectrum of airborne pollutants that are hypothesized to potentially influence pregnancy outcomes, neurodevelopment, asthma, atopy, immune development, obesity, and pubertal development. In this article we summarize six applicable exposure assessment lessons learned from the Centers for Children’s Environmental Health and Disease Prevention Research that may enhance the National Children’s Study: a) Selecting individual study subjects with a wide range of pollution exposure profiles maximizes spatial-scale exposure contrasts for key pollutants of study interest. b) In studies with large sample sizes, long duration, and diverse outcomes and exposures, exposure assessment efforts should rely on modeling to provide estimates for the entire cohort, supported by subject-derived questionnaire data. c) Assessment of some exposures of interest requires individual measurements of exposures using snapshots of personal and microenvironmental exposures over short periods and/or in selected microenvironments. d) Understanding issues of spatial–temporal correlations of air pollutants, the surrogacy of specific pollutants for components of the complex mixture, and the exposure misclassification inherent in exposure estimates is critical in analysis and interpretation. e) “Usual” temporal, spatial, and physical patterns of activity can be used as modifiers of the exposure/outcome relationships. f) Biomarkers of exposure are useful for evaluation of specific exposures that have multiple routes of exposure. If these lessons are applied, the National Children’s Study offers a unique opportunity to assess the adverse effects of air pollution on interrelated health outcomes during the critical early life period

Mortality burdens in California due to air pollution attributable to local and nonlocal emissions.
Limited research has been conducted on the contributions of local and nonlocal emission sources to ambient fine particulate matter (PM2.5) and ozone (O3) and their associated mortality. In this study, we estimated the total mortality resulting from long-term PM2.5 and O3 exposures in California in 2012 using multiple concentration response functions (CRFs) and attributed the estimated mortality to different emission groups. The point estimates of PM2.5-associated mortality in California ranged from 12,700 to 26,700, of which 53% were attributable to in-state anthropogenic emissions. Based on new epidemiological evidence, we estimated that O3 could be associated with up to 13,700 deaths from diseases of both the respiratory and cardiovascular systems in California. In addition, 75% of the ambient O3 in California was due to distant emissions outside the western United States, leading to 92% of the O3-associated mortality. Overall, distant emissions lead to greater mortality burdens of air pollution in California than local anthropogenic emissions
Sedimentology and microfacies of a mud-rich slope succession: in the Carboniferous Bowland Basin, NW England (UK)
A paucity of studies on mud-rich basin slope successions has resulted in a significant gap in our sedimentological
understanding in these settings. Here, macro- and micro-scale analysis of mudstone composition, texture and organic matter
was undertaken on a continuous core through a mud-dominated slope succession from the Marl Hill area in the Carboniferous
Bowland Basin. Six lithofacies, all dominated by turbidites and debrites, combine into three basin slope facies associations:
sediment-starved slope, slope dominated by low-density turbidites and slope dominated by debrites. Variation in slope
sedimentation was a function of relative sea-level change, with the sediment-starved slope occurring during maximum flooding
of the contemporaneous shelf, and the transition towards a slope dominated by turbidites and then debrites occurring during
normal or forced shoreline progradation towards the shelf margin. The sediment-starved slope succession is dominated by Type
II kerogen, whereas the slope dominated by low-density turbidites is dominated by Type III kerogen. This study suggests that
mud-dominated lower slope settings are largely active depositional sites, with consistent evidence for sediment traction.
Additionally, the composition and texture of basin slope mudstones, as well as organic content, vary predictably as a function of
shelf processes linked to relative sea-level change
Effect of exposure to natural environment on health inequalities: an observational population study
<b>Background:</b> Studies have shown that exposure to the natural environment, or so-called green space, has an independent effect on health and health-related behaviours. We postulated that income-related inequality in health would be less pronounced in populations with greater exposure to green space, since access to such areas can modify pathways through which low socio-economic position can lead to disease.<p></p>
<b>Methods:</b> We classified the population of England at younger than retirement age (n=40 813 236) into groups on the basis of income deprivation and exposure to green space. We obtained individual mortality records (n=366 348) to establish whether the association between income deprivation, all-cause mortality, and cause-specific mortality (circulatory disease, lung cancer, and intentional self-harm) in 2001—05, varied by exposure to green space measured in 2001, with control for potential confounding factors. We used stratified models to identify the nature of this variation.<p></p>
<b>Findings:</b> The association between income deprivation and mortality differed significantly across the groups of exposure to green space for mortality from all causes (p<0·0001) and circulatory disease (p=0·0212), but not from lung cancer or intentional self-harm. Health inequalities related to income deprivation in all-cause mortality and mortality from circulatory diseases were lower in populations living in the greenest areas. The incidence rate ratio (IRR) for all-cause mortality for the most income deprived quartile compared with the least deprived was 1·93 (95% CI 1·86—2·01) in the least green areas, whereas it was 1·43 (1·34—1·53) in the most green. For circulatory diseases, the IRR was 2·19 (2·04—2·34) in the least green areas and 1·54 (1·38—1·73) in the most green. There was no effect for causes of death unlikely to be affected by green space, such as lung cancer and intentional self-harm.<p></p>
<b>Interpretation:</b> Populations that are exposed to the greenest environments also have lowest levels of health inequality related to income deprivation. Physical environments that promote good health might be important to reduce socio-economic health inequalities.<p></p>
Potent and Potentially Non-Cardiotoxic Anthracyclines: Their Synthesis, Biological Evaluation, and Comparison of Hydrozone-Mediated Reductions
After nearly half a century since medical approval, the anthracycline doxorubicin (DOX) remains one of the most potent and clinically useful anticancer agents. In spite of its long history, however, the cytotoxic mechanisms of DOX have been debated and remain controversial. Several well-supported mechanisms will be discussed, such as the potential to intercalate DNA and induce apoptosis through topoisomerase poisoning, free radical formation, and DNA cross-linking. While DOX has substantial medical importance, it is plagued by a life-threatening dose-dependent cardiotoxic side effect associated with several structural groups. A number of modifications to DOX have been accomplished to attempt to remove treatment-induced cardiotoxicity, including the reduction of the C-13 carbonyl to a methylene and conversion of the quinone moiety to a less reactive iminoquinone. This modified form, termed 5-imino-13-deoxydoxorubicin (DIDOX), has displayed no cardiotoxic side effects in any clinical setting. However, these structural changes have reduced the potency of the drug more than four-fold.
In this work, six different analogs of DIDOX were synthesized and evaluated for in vitro cytotoxicity against an array of cancer cell lines. The six analogs were designed to incorporate reactive moieties attached to the 3\u27-amine of the daunasamine sugar. Of the six synthesized, four were at least as cytotoxic as DOX, and several were up to 100-fold more potent. Preliminary results also suggest that modifications to the 3\u27-amine attenuate the multidrug resistance observed during anthracycline treatment. In addition to the synthesis and preliminary evaluation of new anthracycline analogs, this work explored methods for improving the synthesis of DIDOX. Specifically, the reduction of DOX’s carbonyl to produce C-13-deoxydoxorubicin (DeoxyDOX) was explored using six different sulfonylhydrazones, and the relative production of the reduced form was compared. Four of the new hydrazones exhibited the potential to improve the overall amount of DeoxyDOX generated, compared to the currently used methods. In summary, the work described presents both the findings for the synthesis of potent and non-cardiotoxic derivatives of doxorubicin that show promise for overcoming multi-drug resistance and potential improvements toward enhancing the overall yield of DIDOX and related analogs
