All over the country, dowagers and damsels alike sigh and shed a tear of pity; and the poor fellow is voted to a high place among the ranking stars of radio. As his popularity increases, his paycheck grows about in proportion to the square of his public, and life becomes a song for the crooner with the catch in his voice. What is the first thing our poor love-starved hero does upon landing a spot on a coast-to- coast network? Why, he flies back to Sac City and marries the winsome little lass with whom he has been in love all the time, of course. Oh, it\u27s an old, old story, but it can\u27t fool me anymore. I can see through it all with ease, because my problem is the counterbalance of that of the crooner

    In the last two or three years, it has been extremely difficult for the peoples of the world to find anything strong, permanent, or lasting enough to withstand the ravages of war. Dreams and cherished hopes have been consumed overnight or in the space of but an hour or two by the ever growing blaze which is threatening to engulf the entire universe. Men have come to believe only in the strength and power of the sword and the maxim that might makes right. The cries of the idealist that war can be banished from the earth are lost in the roar of cannon, the clump-clump of marching feet, and the screams of fanatical soap-box orators. And, indeed, as the forces of evil continue to succeed in their nefarious schemes, it is hard to believe that there is any force in the world at the command of man which can overcome the great god Mars and his followers. But a force is there; a force which has proven its powers over and over again where men are ready and willing to accept it as a means of combating evil. I refer to one of the most potent forces known to man, if he will only realize it - the force of education

    It is the goal of NeighborWorks America to make every place a community of opportunity. Unfortunately, some areas are being left behind more than others as our global and national economies continue to shift. Rural communities are among them. The people who have toiled in the coal mines of Kentucky and West Virginia, as well as in the paper and textile mills in Maine and western North Carolina, have not fared well in the changing economy. Likewise, the historically disenfranchised Native Americans in the Southwest, Latinos in the border colonias and the residents of the disaster-plagued Delta are struggling to survive. Our country needs to bring opportunity back to these regions and their people.Although rural America accounts for less than 20 percent of the country's overall population, 85 percent of persistent-poverty counties are outside of metro areas. Yet at the same time, there are so many examples of people and organizations doing good work; they just need support and the resources to go to scale. Special attention clearly is required, and that's why we formed the Rural Initiative

    Performance assessment of urban precinct design: a scoping study

    Executive Summary: Significant advances have been made over the past decade in the development of scientifically and industry accepted tools for the performance assessment of buildings in terms of energy, carbon, water, indoor environment quality etc. For resilient, sustainable low carbon urban development to be realised in the 21st century, however, will require several radical transitions in design performance beyond the scale of individual buildings. One of these involves the creation and application of leading edge tools (not widely available to built environment professions and practitioners) capable of being applied to an assessment of performance across all stages of development at a precinct scale (neighbourhood, community and district) in either greenfield, brownfield or greyfield settings. A core aspect here is the development of a new way of modelling precincts, referred to as Precinct Information Modelling (PIM) that provides for transparent sharing and linking of precinct object information across the development life cycle together with consistent, accurate and reliable access to reference data, including that associated with the urban context of the precinct. Neighbourhoods are the ‘building blocks’ of our cities and represent the scale at which urban design needs to make its contribution to city performance: as productive, liveable, environmentally sustainable and socially inclusive places (COAG 2009). Neighbourhood design constitutes a major area for innovation as part of an urban design protocol established by the federal government (Department of Infrastructure and Transport 2011, see Figure 1). The ability to efficiently and effectively assess urban design performance at a neighbourhood level is in its infancy.     A proteomic approach to identify endosomal cargoes controlling cancer invasiveness

    We have previously shown that Rab17 - a small GTPase associated with epithelial polarity - is specifically suppressed by ERK2 signalling to promote an invasive phenotype. However, the mechanisms through which Rab17 loss permits invasiveness, and the endosomal cargoes that are responsible for mediating this are not known. Using quantitative mass spectrometry-based proteomics, we have found that knockdown of Rab17 leads to highly selective reduction in the cellular levels of a v-SNARE (Vamp8). Moreover, proteomics and immunofluorescence indicate that Vamp-8 is associated with Rab17 at late endosomes. Reduced levels of Vamp8 promote transition between ductal carcinoma in situ (DCIS) and a more invasive phenotype. We developed an unbiased proteomic approach to elucidate the complement of receptors that redistributes between endosomes and the plasma membrane, and have pin-pointed neuropilin-2 (NRP2) as a key pro-invasive cargo of Rab17/Vamp8-regulated trafficking. Indeed, reduced Rab17 or Vamp8 levels lead to increased mobilisation of NRP2-containing late endosomes and upregulated cell surface expression of NRP2. Finally, we show that NRP2 is required for the basement membrane disruption which accompanies transition between DCIS and a more invasive phenotype

    The initiator methionine tRNA drives cell migration and invasion leading to increased metastatic potential in melanoma

    The cell's repertoire of transfer RNAs (tRNAs) has been linked to cancer. Recently, levels of the initiator methionine tRNA (tRNAiMet) in stromal fibroblasts have been shown to influence extracellular matrix (ECM) secretion to drive tumour growth and angiogenesis. Here we show that increased tRNAiMet within cancer cells does not influence tumour growth, but drives cell migration and invasion via a mechanism that is independent from ECM synthesis and dependent on α5β1 integrin and levels of the translation initiation ternary complex. In vivo and ex vivo migration (but not proliferation) of melanoblasts is significantly enhanced in transgenic mice which express additional copies of the tRNAiMet gene. We show that increased tRNAiMet in melanoma drives migratory, invasive behaviour and metastatic potential without affecting cell proliferation and primary tumour growth, and that expression of RNA polymerase III-associated genes (which drive tRNA expression) are elevated in metastases by comparison with primary tumours. Thus specific alterations to the cancer cell tRNA repertoire drive a migration/invasion programme that may lead to metastasis
