    La finalitat principal del vídeo realitzat correspon a un dels objectius docents que proposàvem en el projecte que ens va ser concedit per l'AGAUR: que l'alumne es pugui acostar a tècniques sofisticades que per la seva complexitat o perillositat no poden realitzar-se en els laboratoris de docència. També com a segon objectiu, pretenem que l'alumne comprengui millor conceptes que s'expliquen a teoria. En la situació actual, els alumnes de primer curs de medicina realitzen pràctiques de laboratori en que apliquen la cromatografia en capa fina per la separació de molècules i l'anàlisi qualitatiu de la seva presència en mostres tissulars. Malgrat que els principis teòrics són bàsicament els mateixos, és impossible en la pràctica realitzar cromatografies d'alta resolució (HPLC) que són comunament utilitzades per la quantificació de molècules diverses, no només en la recerca bàsica sinó en el laboratori clínic. Per altra banda, els alumnes de tercer i quart curs de la llicenciatura de bioquímica coneixen les bases teòriques de l'HPLC però tampoc en els laboratoris de pràctiques pot realitzar-se cromatografies d'alta resolució degut al grau de complexitat i despesa que comporten

    Circumnuclear structure and kinematics in the active galaxy NGC 6951

    A study is presented of the central structure and kinematics of the galaxy NGC 6951, by means of broad band B'IJK images and high resolution high dispersion longslit spectroscopy, together with archival HST WFPC2 V and NICMOS2 J and H images. We find that there is evidence of two modes of star formation, in bursts and continuously. The equivalent width of the CaII triplet absorption lines show that, in the metal rich central region, the continuum is dominated by a population of red supergiants. The gaseous and stellar kinematics along three slit position angles, suggest the existence of a hierarchy of disks within disks, whose dynamics are decoupled at the two ILRs, that we find at 180 pc and at 1100 pc. This is supported by the structure seen in the high resolution HST images. The nucleus is spatially resolved within a radius of 1.5 arcsec, just inside the innermost ILR. Outside the iILR, the stellar CaT velocity profile is resolved into two components, associated with the bar and the disk. Several results indicate that this is a dynamically old system. It is thus possible that a nuclear bar has existed in NGC 6951 that drove the gas towards the nucleus, as in the bars within bars scenario, but that this bar has already dissolved by the gas accumulated within the circumnuclear region. We discuss the possibility that the kinematical component inside the iILR could be due to a nuclear outflow produced by the combined effects of SN and SN remnants, or to a nuclear disk, as in the disk within disk scenario that we propose for the fueling of the AGN in NGC 6951.Comment: 14 pages, 16 figures.     Leucine absorption after jejunoileal bypass in rats

    Jejunal and ileal absorption of L-leucine has been studied in rats subjected to jejunoileal bypass and in sham-operated rats, for five minute periods, using a perfusion technique. Aminoacid concentrations were: 1, 2.5, 5, 10 and 25 mM. In sorne experiments methionine was added to determine simple diffusion. The ratio of the active/diffusive components of absorption were calculated at the different luminal aminoacid concentrations in both groups of rats, showing that this ratio was lower in control animal

    Orexin-Corticotropin-Releasing Factor Receptor Heteromers in the Ventral Tegmental Area as Targets for Cocaine

    Release of the neuropeptides corticotropin-releasing factor (CRF) and orexin-A in the ventral tegmental area (VTA) play an important role in stress-induced cocaine-seeking behavior. We provide evidence for pharmacologically significant interactions between CRF and orexin-A that depend on oligomerization of CRF1 receptor (CRF1R) and orexin OX1 receptors (OX1R). CRF1R–OX1R heteromers are the conduits of a negative crosstalk between orexin-A and CRF as demonstrated in transfected cells and rat VTA, in which they significantly modulate dendritic dopamine release. The cocaine target σ1 receptor (σ1R) also associates with the CRF1R–OX1R heteromer. Cocaine binding to the σ1R–CRF1R–OX1R complex promotes a long-term disruption of the orexin-A–CRF negative crosstalk. Through this mechanism, cocaine sensitizes VTA cells to the excitatory effects of both CRF and orexin-A, thus providing a mechanism by which stress induces cocaine seeking

    Application of colon capsule endoscopy (CCE) to evaluate the whole gastrointestinal tract: a comparative study of single-camera and dual-camera analysis

    Journal Article;BACKGROUND AND STUDY AIMS Colon capsule endoscopy (CCE) was developed for the evaluation of colorectal pathology. In this study, our aim was to assess if a dual-camera analysis using CCE allows better evaluation of the whole gastrointestinal (GI) tract compared to a single-camera analysis. PATIENTS AND METHODS We included 21 patients (12 males, mean age 56.20 years) submitted for a CCE examination. After standard colon preparation, the colon capsule endoscope (PillCam Colon™) was swallowed after reinitiation from its "sleep" mode. Four physicians performed the analysis: two reviewed both video streams at the same time (dual-camera analysis); one analyzed images from one side of the device ("camera 1"); and the other reviewed the opposite side ("camera 2"). We compared numbers of findings from different parts of the entire GI tract and level of agreement among reviewers. RESULTS A complete evaluation of the GI tract was possible in all patients. Dual-camera analysis provided 16% and 5% more findings compared to camera 1 and camera 2 analysis, respectively. Overall agreement was 62.7% (kappa = 0.44, 95% CI: 0.373-0.510). Esophageal (kappa = 0.611) and colorectal (kappa = 0.595) findings had a good level of agreement, while small bowel (kappa = 0.405) showed moderate agreement. CONCLUSION The use of dual-camera analysis with CCE for the evaluation of the GI tract is feasible and detects more abnormalities when compared with single-camera analysis.Ye

    Newborn Screening for Primary T- and B-Cell Immune Deficiencies—A Prospective Study in Andalucía

    Background: Quantification of T-cell-receptor-excision circles (TRECs) and kappa-deleting-recombination-excision circles (KRECs) from dried blood spots (DBS) allows detection of neonates with severe T-cell and/or B-cell lymphopenia that are potentially affected by severe combined immunodeficiency (SCID), as well as X-linked agammaglobulinemia (XLA). Methods: Determination of TRECs and KRECs using a triplex RT-PCR (TRECS-KRECS-β-actin) assay from prospectively collected DBS between February 2014 and December 2016 in three hospitals in Seville, Spain. Cut-off levels were TRECs 700/punch. Internal (SCID, XLA, ataxia telangiectasia) and external controls (CDC) were included. Results: A total of 8943 DBS samples obtained from 8814 neonates were analysed. Re-punching was necessary in 124 samples (1.4%) due to insufficient β-actin values (<700 copies/punch). Preterm neonates (GA < 37 weeks) and neonates with a BW < 2500 g showed significantly lower TRECs and KRECs levels (p < 0.001). Due to repeated pathological results, ten neonates were re-sampled (0.11%), of which five neonates (0.055%) confirmed the pathological results: one case was a fatal chromosomopathy (TRECs 1/KRECs 4); two were extreme premature newborns (TRECs 0/KRECs 0 and TRECs 1/KRECs 20 copies/punch); and 2 neonates were born to mothers receiving azathioprine during pregnancy (TRECs 92/KRECs 1 and TRECs 154/KRECs 3 copies/punch). All controls were correctly identified. Conclusions: Severe T- and B-cell lymphopenias were correctly identified by the TRECS-KRECS-β-actin assay. Prematurity and low BW are associated with lower TREC and KREC levels.
