    RNA editing by adenosine deamination gives an organism the ability to alter its protein-coding sequences, resulting in an overall change in protein structure and function. Adenosine deaminases acting on RNA (ADARs) convert specific adenosines (A) into inosines (I) that are interpreted as guanosines (G) by the translational machinery. Coleoid cephalopods (octopus, squid, and cuttlefish), like vertebrates, possess two catalytically active ADAR enzymes, sqADAR1 and sqADAR2, but unlike any other organism, cephalopods recode transcripts by A-to-I RNA editing to an extraordinary extent, particularly in neuronal tissue

    It\u27s Not Just Ferguson: Missouri Supreme Court Should Consolidate the Municipal Court System

    The Missouri Supreme Court\u27s unprecedented decision to take control of Ferguson\u27s Municipal Court was based primarily on issues raised during sustained protest following the killing of Mike Brown and reports published by ArchCity Defenders and the Department of Justice. These reports highlighted racial disparity in traffic stops, excessive revenue generation, and excessive warrants and arrests and confirmed the lived experiences of poor and Black people in St. Louis: there is a racially discriminatory and profit-driven approach to law enforcement made possible only by the collaborative efforts of local government, police, and courts. These condemned practices are not unique to Ferguson. Rather, many St. Louis municipalities are demonstrably worse than Ferguson with respect to the highlighted factors. And while there have been legal victories5 in the past year, new legislation,6 and the sustained efforts of the Black Lives Matter movement, the Supreme Com1 should seize the opportunity to bring meaningful and transformative change to the region by following the recommendation of the Ferguson Commission7, ArchCity Defenders, SLU Law Legal Clinics, Better Together, Missourians Organizing for Reform and Empowerment, and the Organization for Black Struggle: Order the consolidation of our 81 courts into a full-time, professional regional court system

For every injective vertex labeling f:V(G)Zf:V(G)\to\mathbb{Z}, there are two induced edge labelings, namely f+:E(G)Zf^+:E(G)\to\mathbb{Z} defined by f+(uv)=f(u)+f(v)f^+(uv)=f(u)+f(v), and f:E(G)Zf^-:E(G)\to\mathbb{Z} defined by f(uv)=f(u)f(v)f^-(uv)=|f(u)-f(v)|. The sum index and the difference index are the minimum cardinalities of the ranges of f+f^+ and ff^-, respectively. We provide upper and lower bounds on the sum index and difference index, and determine the sum index and difference index of various families of graphs.     Assessment of Mesophotic Coral Ecosystem Connectivity for Proposed Expansion of a Marine Sanctuary in the Northwest Gulf of Mexico: Larval Dynamics

    In coral reef ecosystems, mesophotic coral habitat (\u3e30 m to the end of the photic zone) are extensions of shallow reefs and contribute to the persistence of coral reef populations. In the North West Gulf of Mexico (NW GOM), the Flower Garden Banks National Marine Sanctuary (FGBNMS) is an isolated reef ecosystem comprising contiguous shallow and mesophotic reefs habitats on two central banks along the margin of the continental shelf. A future expansion of the sanctuary is proposed to include additional mesophotic banks and aims at building a network of protected areas in the NW GOM to ensure the persistence of the coral reef populations inhabiting the sanctuary. To evaluate the feasibility of this expansion and investigate the overall dynamics of coral species in the region, we studied the patterns of larval connectivity of Montastraea cavernosa, a common depth generalist coral species, using a larval dispersal modeling approach. Our results highlighted larval exports from the NW GOM banks to the northeastern and southwestern GOM, larval connectivity between all banks investigated in this study, and the potential for exporting larvae from mesophotic to shallower reefs. Our study associated with Studivan and Voss (2018; associate manuscript) demonstrates the relevance of combining modeling and genetic methods to consider both demographic and genetic timescales for the evaluation of the connectivity dynamics of marine populations. In the case of the NW GOM, both studies support the future management plan for expanding FGBNMS

    Genome evolution in the allotetraploid frog Xenopus laevis

    To explore the origins and consequences of tetraploidy in the African clawed frog, we sequenced the Xenopus laevis genome and compared it to the related diploid X. tropicalis genome. We characterize the allotetraploid origin of X. laevis by partitioning its genome into two homoeologous subgenomes, marked by distinct families of ???fossil??? transposable elements. On the basis of the activity of these elements and the age of hundreds of unitary pseudogenes, we estimate that the two diploid progenitor species diverged around 34 million years ago (Ma) and combined to form an allotetraploid around 17-18 Ma. More than 56% of all genes were retained in two homoeologous copies. Protein function, gene expression, and the amount of conserved flanking sequence all correlate with retention rates. The subgenomes have evolved asymmetrically, with one chromosome set more often preserving the ancestral state and the other experiencing more gene loss, deletion, rearrangement, and reduced gene expression.ope

    Nitroxyl Hybrids with Curcumin and Stilbene Scaffolds Display Potent Antioxidant Activity, Remodel the Amyloid Beta Oligomer, and Reverse Amyloid Beta-Induced Cytotoxicity

    The disorder and heterogeneity of low-molecular-weight amyloid-beta oligomers (AβOs) underlie their participation in multiple modes of cellular dysfunction associated with the etiology of Alzheimer's disease (AD). The lack of specified conformational states in these species complicates efforts to select or design small molecules to targeting discrete pathogenic states. Furthermore, targeting AβOs alone may be therapeutically insufficient, as AD progresses as a multifactorial, self-amplifying cascade. To address these challenges, we have screened the activity of seven new candidates that serve as Paramagnetic Amyloid Ligand (PAL) candidates. PALs are bifunctional small molecules that both remodel the AβO structure and localize a potent antioxidant that mimics the activity of SOD within live cells. The candidates are built from either a stilbene or curcumin scaffold with nitroxyl moiety to serve as catalytic antioxidants. Measurements of PAL AβO binding and remolding along with assessments of bioactivity allow for the extraction of useful SAR information from screening data. One candidate (HO-4450; PMT-307), with a six-membered nitroxyl ring attached to a stilbene ring, displays the highest potency in protecting against cell-derived Aβ. A preliminary low-dose evaluation in AD model mice provides evidence of modest treatment effects by HO-4450. The results for the curcumin PALs demonstrate that the retention of the native curcumin phenolic groups is advantageous to the design of the hybrid PAL candidates. Finally, the PAL remodeling of AβO secondary structures shows a reasonable correlation between a candidate's bioactivity and its ability to reduce the fraction of antiparallel β-strand

    Assessing the potential of inversion layer solar cells based on highly charged dielectric nanolayers

    The production and performance of p-type inversion layer (IL) Si solar cells, manufactured with an ion-injection technique that produces a highly charged dielectric nanolayer, are investigated. It is demonstrated that the field-induced electron layer underneath the dielectric can reach a dark sheet resistance of 0.95 kΩ sq−1 on a 1 Ω cm n-type substrate, lower than any previously reported. In addition, it is shown that the implied open-circuit voltage of a p-type IL cell precursor with a highly charged dielectric is equivalent to that of a cell with a phosphorous emitter. In the cell precursor, light-beam-induced current measurements are performed, and the uniformity and performance of the IL is demonstrated. Finally, simulations are used to explain the physical characteristics of the interface leading to extremely low sheet resistances, and to assess the efficiency potential of IL cells. IL cells are predicted to reach an efficiency of 24.5%, and 24.8% on 5/10 Ω cm substrates, by replacing the phosphorous emitter with a simpler manufacturing process. This requires a charge density of beyond 2 × 1013 cm−2, as is demonstrated here. Moreover, IL cells perform even better at higher charge densities and when negative charge is optimized at the rear dielectric
