Truck-based mobile wireless sensor networks for the experimental observation of vehicle–bridge interaction
Heavy vehicles driving over a bridge create a complex dynamic phenomenon known as vehicle–bridge interaction. In recent years, interest in vehicle–bridge interaction has grown because a deeper understanding of the phenomena can lead to improvements in bridge design methods while enhancing the accuracy of structural health monitoring techniques. The mobility of wireless sensors can be leveraged to directly monitor the dynamic coupling between the moving vehicle and the bridge. In this study, a mobile wireless sensor network is proposed for installation on a heavy truck to capture the vertical acceleration, horizontal acceleration and gyroscopic pitching of the truck as it crosses a bridge. The vehicle-based wireless monitoring system is designed to interact with a static, permanent wireless monitoring system installed on the bridge. Specifically, the mobile wireless sensors time-synchronize with the bridge's wireless sensors before transferring the vehicle response data. Vertical acceleration and gyroscopic pitching measurements of the vehicle are combined with bridge accelerations to create a time-synchronized vehicle–bridge response dataset. In addition to observing the vehicle vibrations, Kalman filtering is adopted to accurately track the vehicle position using the measured horizontal acceleration of the vehicle and positioning information derived from piezoelectric strip sensors installed on the bridge deck as part of the bridge monitoring system. Using the Geumdang Bridge (Korea), extensive field testing of the proposed vehicle–bridge wireless monitoring system is conducted. Loss of the tumor suppressor, Tp53, enhances the androgen receptor-mediated oncogenic transformation and tumor development in the mouse prostate.
Recent genome analysis of human prostate cancers demonstrated that both AR gene amplification and TP53 mutation are among the most frequently observed alterations in advanced prostate cancer. However, the biological role of these dual genetic alterations in prostate tumorigenesis is largely unknown. In addition, there are no biologically relevant models that can be used to assess the molecular mechanisms for these genetic abnormalities. Here, we report a novel mouse model, in which elevated transgenic AR expression and Trp53 deletion occur simultaneously in mouse prostatic epithelium to mimic human prostate cancer cells. These compound mice developed an earlier onset of high-grade prostatic intraepithelial neoplasia and accelerated prostate tumors in comparison with mice harboring only the AR transgene. Histological analysis showed prostatic sarcomatoid and basaloid carcinomas with massive squamous differentiation in the above compound mice. RNA-sequencing analyses identified a robust enrichment of the signature genes for human prostatic basal cell carcinomas in the above prostate tumors. Master regulator analysis revealed SOX2 as a transcriptional regulator in prostatic basal cell tumors. Elevated expression of SOX2 and its downstream target genes were detected in prostatic tumors of the compound mice. Chromatin immunoprecipitation analyses implicate a coregulatory role of AR and SOX2 in the expression of prostatic basal cell signature genes. Our data demonstrate a critical role of SOX2 in prostate tumorigenesis and provide mechanistic insight into prostate tumor aggressiveness and progression mediated by aberrant AR and p53 signaling pathways
Suggestion of new possibilities in approaching individual variability in appetite through constitutional typology: a pilot study
BACKGROUND: Appetite is intricately connected to eating behaviors and shows a high individual variability. In an attempt to approach the problem of gut hormone profiles, appetite, and eating behaviors at the individual level, we have adopted a constitutional typing system widely used in traditional East-Asian medicine, the Sasang constitutional typology, in order to determine the individual variations in appetite, eating behavior, and weight change. METHODS: This pilot study was designed to investigate the variability of appetite among individuals by tracking the gut hormone patterns across different constitutional types. Pre- and post-prandial concentrations of anorectic (peptide YY (PYY), glucagon-like peptide 1 (GLP-1)) and orexigenic (active ghrelin) gut hormones were measured in healthy, normal-weight (18.5 kg/m(2) ≤BMI <23 kg/m(2)) male subjects aged 20–35 (Soyang (SY) (n = 9), Taeeum (TE) (n = 9), and Soeum (SE) (n = 10) constitutional types). RESULTS: Significant differences were found only in the PYY concentrations across the three groups (p = 0.031). The PYY concentration peaked at 30-min post-prandial in the SE group and was significantly higher compared to the other two groups (p = 0.004). The GLP-1 concentration peaked at 15-min post-prandial in the SE group (not significant). The ghrelin levels at 30-min pre-prandial were relatively lower in the TE group compared to the other groups (not significant). CONCLUSIONS: In conclusion, although with weak statistical power, meaningful gut hormone patterns specific to each constitutional type were discovered in this pilot study, which could offer a new method of approaching the problem of appetite and eating behavior from the angle of individual variability in appetite

Chromosomal instability in untreated primary prostate cancer as an indicator of metastatic potential.
BackgroundMetastatic prostate cancer (PC) is highly lethal. The ability to identify primary tumors capable of dissemination is an unmet need in the quest to understand lethal biology and improve patient outcomes. Previous studies have linked chromosomal instability (CIN), which generates aneuploidy following chromosomal missegregation during mitosis, to PC progression. Evidence of CIN includes broad copy number alterations (CNAs) spanning > 300 base pairs of DNA, which may also be measured via RNA expression signatures associated with CNA frequency. Signatures of CIN in metastatic PC, however, have not been interrogated or well defined. We examined a published 70-gene CIN signature (CIN70) in untreated and castration-resistant prostate cancer (CRPC) cohorts from The Cancer Genome Atlas (TCGA) and previously published reports. We also performed transcriptome and CNA analysis in a unique cohort of untreated primary tumors collected from diagnostic prostate needle biopsies (PNBX) of localized (M0) and metastatic (M1) cases to determine if CIN was linked to clinical stage and outcome.MethodsPNBX were collected from 99 patients treated in the VA Greater Los Angeles (GLA-VA) Healthcare System between 2000 and 2016. Total RNA was extracted from high-grade cancer areas in PNBX cores, followed by RNA sequencing and/or copy number analysis using OncoScan. Multivariate logistic regression analyses permitted calculation of odds ratios for CIN status (high versus low) in an expanded GLA-VA PNBX cohort (n = 121).ResultsThe CIN70 signature was significantly enriched in primary tumors and CRPC metastases from M1 PC cases. An intersection of gene signatures comprised of differentially expressed genes (DEGs) generated through comparison of M1 versus M0 PNBX and primary CRPC tumors versus metastases revealed a 157-gene "metastasis" signature that was further distilled to 7-genes (PC-CIN) regulating centrosomes, chromosomal segregation, and mitotic spindle assembly. High PC-CIN scores correlated with CRPC, PC-death and all-cause mortality in the expanded GLA-VA PNBX cohort. Interestingly, approximately 1/3 of M1 PNBX cases exhibited low CIN, illuminating differential pathways of lethal PC progression.ConclusionsMeasuring CIN in PNBX by transcriptome profiling is feasible, and the PC-CIN signature may identify patients with a high risk of lethal progression at the time of diagnosis
The Relationships among the Learning Transfer System, Managers’ Creative Learning Transfer, and Job Performance
To survive and thrive in the ever-changing global environment, organizations must constantly innovate and transform in the market by obtaining, applying, and creating new knowledge. In particular, given the importance of managers’ excellent leadership for organizational performance, organizations must find a way to enhance managers’ creative application of leadership knowledge to novel business situations (i.e., creative learning transfer). Over the past 110 years, research on learning transfer has proliferated because new knowledge and application of it to business must be at the heart of competitive advantages of an organization. Despite numerous empirical inquiries and advancement on learning transfer, there are still four major research gaps to be closed: (1) lack of a comprehensive instrument to measure predictors of creative learning transfer; (2) paucity of empirical research on learning transfer guided by sound theories; (3) ignorance of the importance of creative learning transfer in literature; and (4) little attention to a motivational factor as a mediator between transfer predictors and transfer outcomes. To fill the gaps, an overarching purpose of the present study was to examine the relationships among the learning transfer system, managers’ creative learning transfer, and job performance.
The targeted population of the current study was managers who worked for large companies in South Korea and completed leadership training programs in the companies. Based on a non-experimental research design, an electronic 76-item survey was used to collect quantitative and qualitative data from 16 companies that agreed to participate in the current study. After data screening, the valid sample consisted of 753 managers from the companies, which may represent 16 industries in the country. To analyze the quantitative data, a series of exploratory and confirmatory factor analyses (EFA/CFA) was conducted, followed by structural equation modeling (SEM) analyses. For the qualitative data, a thematic analysis was conducted.
Three major findings emerged from the current study. First, the Learning Transfer System Inventory (LTSI) Version 4 was successfully validated in an international context. Second, the nomological network among the learning transfer system, creative learning transfer, and job performance was confirmed. Third, seven themes of enablers and barriers for creative learning transfer were identified for use in the future research. HRD professionals may obtain critical implications from the current study to help organizational managers apply learned leadership knowledge and skills to novel business situations to create more competitive work systems, products, and/or services. The current study may serve as the bedrock on which researchers can theorize the concept of creative learning transfer, elaborating on organizational knowledge creation theory
Crisis to Opportunity: The Role of Consumer Awareness in Mergers and Acquisitions (M&As) during the COVID-19 Pandemic
In the uncertainty fueled by the COVID-19 pandemic, mergers and acquisitions (M&As) have emerged as key strategic responses by firms. This study explores the impact of M&As on acquirers' firm value, utilizing a firm-level panel dataset from SDC Platinum. Empirical evidence recognizes the potential negative impact of transaction value in M&As and the pandemic's effect on market volatility that may occasionally exacerbate the adverse influence on acquirers' firm value. The findings indicate that effective marketing strategies, such as enhancing consumer awareness through increasing advertising expenditures, can counterbalance these influences, particularly during uncertain times. This study accentuates the importance of adaptability and a responsive marketing approach in managing M&As during a global crisis. It provides valuable perspectives on consumer awareness in strategic decision-making, offering insights for both academic and business communities and focusing on actionable strategies for navigating the global market turmoil transformed by COVID-19
Relationship between core self-evaluation and innovative work behavior: mediating effect of affective organizational commitment and moderating effect of organizational learning capacity
Focusing on employees, this study examined the respective mediating and moderating effects of affective organizational commitment and organizational learning capacity in the relationship between core self-evaluation and innovation work behavior. We collected data via an online survey from 330 office workers at midsize and large companies in a metropolitan area of South Korea. The results of analyzing the data using PROCESS macro were as follows: (1) core self-evaluation was positively related to innovative work behavior; (2) the relationship was mediated by affective organizational commitment; (3) the relationship was buffered by organizational learning capacity, such that a higher level of organizational learning capacity diminished the impact of core self-evaluation on innovative wok behavior; and (4) the conditional effect of core self-evaluation on innovative work behavior existed only in the group of a low level of organizational learning capacity. Based on these findings, we suggested implications for theory building, research, and practice
