Exoplanet Atmosphere Measurements from Transmission Spectroscopy and other Planet-Star Combined Light Observations
It is possible to learn a great deal about exoplanet atmospheres even when we
cannot spatially resolve the planets from their host stars. In this chapter, we
overview the basic techniques used to characterize transiting exoplanets -
transmission spectroscopy, emission and reflection spectroscopy, and full-orbit
phase curve observations. We discuss practical considerations, including
current and future observing facilities and best practices for measuring
precise spectra. We also highlight major observational results on the

Longitudinal Evaluation of the Hypothalamic-Pituitary-Testicular Function in 8 Boys with Adrenal Hypoplasia Congenita (AHC) Due to NR0B1 Mutations
BACKGROUND:Boys carrying mutations in the NR0B1 gene develop adrenal hypoplasia congenita (AHC) and impaired sexual development due to the combination of hypogonadotropic hypogonadism (HH) and primary defects in spermatogenesis. METHODS:We analysed the evolution of hypothalamic-pituitary-testicular function of 8 boys with AHC due to NR0B1 mutations. Our objective was to characterize and monitor the progressive deterioration of this function. RESULTS:The first symptoms appeared in the neonatal period (n = 5) or between 6 months and 8.7 years (n = 3). Basal plasma adrenocorticotrophic hormone (ACTH) concentrations increased in all boys, whilst cortisol levels decreased in one case. The natremia was equal or below 134 mmol/L and kaliemia was over 5 mmol/L. All had increased plasma renin. In 3 of 4 patients diagnosed in the neonatal period and evaluated during the first year, the basal plasma gonadotropins concentrations, and their response to gonadotropin releasing hormone (GnRH) test (n = 2), and those of testosterone were normal. The plasma inhibin B levels were normal in the first year of life. With the exception of two cases these concentrations decreased to below the normal for age. Anti-Müllerian hormone concentrations were normal for age in all except one case, which had low concentrations before the initiation of testosterone treatment. In 3 of the 8 cases the gene was deleted and the remaining 5 cases carried frameshift mutations that are predicted to introduce a downstream nonsense mutation resulting in a truncated protein. CONCLUSIONS:The decreases in testosterone and inhibin B levels indicated a progressive loss of testicular function in boys carrying NR0B1 mutations. These non-invasive examinations can help to estimate the age of the testicular degradation and cryopreservation of semen may be considered in these cases as investigational procedure with the aim of restoring fertility
Gene Set Enrichment Analysis Identifies LIF as a Negative Regulator of Human Th2 Cell Differentiation
In this study we show that IL-4 is crucial during reinforcement window of human Th2 differentiation for optimal Th2 development. We have also shown here that during this stage, IL-4 helps in cellular decision-making process of differentiation versus proliferation. We have combined computational and experimental methods to analyze Th2 transcription network to name novel players of the process of Th2 differentiation. Here we report that LIF through STAT3 negatively regulates Th2 differentiation. This approach can be generalized to analyze “omics” data to identify key regulatory modules
Atmospheric retrieval of exoplanets
Exoplanetary atmospheric retrieval refers to the inference of atmospheric
properties of an exoplanet given an observed spectrum. The atmospheric
properties include the chemical compositions, temperature profiles,
clouds/hazes, and energy circulation. These properties, in turn, can provide
key insights into the atmospheric physicochemical processes of exoplanets as
well as their formation mechanisms. Major advancements in atmospheric retrieval
have been made in the last decade, thanks to a combination of state-of-the-art
spectroscopic observations and advanced atmospheric modeling and statistical
inference methods. These developments have already resulted in key constraints
on the atmospheric H2O abundances, temperature profiles, and other properties
for several exoplanets. Upcoming facilities such as the JWST will further
advance this area. The present chapter is a pedagogical review of this exciting
frontier of exoplanetary science. The principles of atmospheric retrievals of
exoplanets are discussed in detail, including parametric models and statistical
inference methods, along with a review of key results in the field. Some of the
main challenges in retrievals with current observations are discussed along
with new directions and the future landscape
Transduction of Human T Cells with a Novel T-Cell Receptor Confers Anti-HCV Reactivity
Hepatitis C Virus (HCV) is a major public health concern, with no effective vaccines currently available and 3% of the world's population being infected. Despite the existence of both B- and T-cell immunity in HCV-infected patients, chronic viral infection and HCV-related malignancies progress. Here we report the identification of a novel HCV TCR from an HLA-A2-restricted, HCV NS3:1073–1081-reactive CTL clone isolated from a patient with chronic HCV infection. We characterized this HCV TCR by expressing it in human T cells and analyzed the function of the resulting HCV TCR-transduced cells. Our results indicate that both the HCV TCR-transduced CD4+ and CD8+ T cells recognized the HCV NS3:1073–1081 peptide-loaded targets and HCV+ hepatocellular carcinoma cells (HCC) in a polyfunctional manner with cytokine (IFN-γ, IL-2, and TNF-α) production as well as cytotoxicity. Tumor cell recognition by HCV TCR transduced CD8− Jurkat cells and CD4+ PBL-derived T cells indicated this TCR was CD8-independent, a property consistent with other high affinity TCRs. HCV TCR-transduced T cells may be promising for the treatment of patients with chronic HCV infections
Development of a multi-dimensional measure of resilience in adolescents: the Adolescent Resilience Questionnaire
Background: The concept of resilience has captured the imagination of researchers and policy makers over the past two decades. However, despite the ever growing body of resilience research, there is a paucity of relevant, comprehensive measurement tools. In this article, the development of a theoretically based, comprehensive multidimensional measure of resilience in adolescents is described.Methods: Extensive literature review and focus groups with young people living with chronic illness informed the conceptual development of scales and items. Two sequential rounds of factor and scale analyses were undertaken to revise the conceptually developed scales using data collected from young people living with a chronic illness and a general population sample.Results: The revised Adolescent Resilience Questionnaire comprises 93 items and 12 scales measuring resilience factors in the domains of self, family, peer, school and community. All scales have acceptable alpha coefficients. Revised scales closely reflect conceptually developed scales.Conclusions: It is proposed that, with further psychometric testing, this new measure of resilience will provide researchers and clinicians with a comprehensive and developmentally appropriate instrument to measure a young person’s capacity to achieve positive outcomes despite life stressors.<br /

The transiting exoplanet community early release science program for JWST
The James Webb Space Telescope (JWST) presents the opportunity to transform
our understanding of planets and the origins of life by revealing the
atmospheric compositions, structures, and dynamics of transiting exoplanets in
unprecedented detail. However, the high-precision, time-series observations
required for such investigations have unique technical challenges, and prior
experience with other facilities indicates that there will be a steep learning
curve when JWST becomes operational. In this paper we describe the science
objectives and detailed plans of the Transiting Exoplanet Community Early
Release Science (ERS) Program, which is a recently approved program for JWST
observations early in Cycle 1. The goal of this project, for which the obtained
data will have no exclusive access period, is to accelerate the acquisition and
diffusion of technical expertise for transiting exoplanet observations with
JWST, while also providing a compelling set of representative datasets that
will enable immediate scientific breakthroughs. The Transiting Exoplanet
Community ERS Program will exercise the time-series modes of all four JWST
instruments that have been identified as the consensus highest priorities,
observe the full suite of transiting planet characterization geometries
(transits, eclipses, and phase curves), and target planets with host stars that
span an illustrative range of brightnesses. The observations in this program
were defined through an inclusive and transparent process that had
participation from JWST instrument experts and international leaders in
transiting exoplanet studies. Community engagement in the project will be
centered on a two-phase Data Challenge that culminates with the delivery of
planetary spectra, time-series instrument performance reports, and open-source
data analysis toolkits in time to inform the agenda for Cycle 2 of the JWST
mission
