Noise Pollution? What\u27s the solution?: Understanding Traffic Noise Pollution in Gettysburg, Pennsylvania
Noise pollution can be damaging to a community by impacting its atmosphere as well as the health of its residents, local quality of life, and local economy. Our study sought to gain insight into the level of noise pollution in Gettysburg, both in-town and on the Gettysburg College campus, and how noise pollution may be affecting the local residents and students. We selected 9 sampling sites, 6 in-town and 3 on the college campus, and measured the noise pollution in dBA with portable sound meters three days a week and three times a day over a three week period. Our data showed no major trends in terms of time of day or day of the week, but there were clear differences in noise pollution levels between the different sites in that town sites were generally louder than on campus sites. Noise pollution in town was often louder than 70 dBA, the noise threshold that indicates possible hearing damage overtime set by the World Health Organization. Gettysburg has recently enacted a noise ordinance; our study suggests the Borough possibly could do more to mitigate traffic noise by repairing roads and manipulating road design
Physics holo.lab learning experience: Using Smartglasses for Augmented Reality labwork to foster the concepts of heat conduction
Fundamental concepts of thermodynamics rely on abstract physical quantities
such as energy, heat and entropy, which play an important role in the process
of interpreting thermal phenomena and statistical mechanics. However, these
quantities are not covered by human (visual) perception and thus, an intuitive
understanding often is lacking. Today immersive technologies like head-mounted
displays of the newest generation, especially HoloLens, allow for high quality
augmented reality learning experiences, which can overcome this perception gap
and simultaneously avoid a split attention effect. In a mixed reality (MR)
scenario as presented in this paper---which we call a holo.lab---human
perception can be extended to the thermal regime by presenting false-color
representations of the temperature of objects as a virtual augmentation
directly on the real object itself in real-time. Direct feedback to
experimental actions of the users in form of different representations allows
for immediate comparison to theoretical principles and predictions and
therefore is supposed to intensify the theory-experiment interactions and to
increase the conceptual understanding. We tested this technology for an
experiment on thermal conduction of metals in the framework of undergraduate
laboratories. A pilot study with treatment and control groups (N = 59) showed a
small positive effect of MR on students' performance measured with a
standardized concept test for thermodynamics, indicating an improvement of the
understanding of the underlying physical concepts
Variations in pre-analytical FFPE sample processing and bioinformatics: challenges for next generation molecular diagnostic testing in clinical pathology
Advances in cellular pathology techniques will improve diagnostic medicine. However, such improvements have to overcome many challenges including variations in pre-analytical sample processing, bioinformatics data analysis and clinical interpretation of data. In order to resolve such challenges, bioinformatics needs to become more tightly coupled to the experimental methodology development

A novel NGS library preparation method to characterize native termini of fragmented DNA.
Biological and chemical DNA fragmentation generates DNA molecules with a variety of termini, including blunt ends and single-stranded overhangs. We have developed a Next Generation Sequencing (NGS) assay, XACTLY, to interrogate the termini of fragmented DNA, information traditionally lost in standard NGS library preparation methods. Here we describe the XACTLY method, showcase its sensitivity and specificity, and demonstrate its utility in in vitro experiments. The XACTLY assay is able to report relative abundances of all lengths and types (5' and 3') of single-stranded overhangs, if present, on each DNA fragment with an overall accuracy between 80-90%. In addition, XACTLY retains the sequence of each native DNA molecule after fragmentation and can capture the genomic landscape of cleavage events at single nucleotide resolution. The XACTLY assay can be applied as a novel research and discovery tool for fragmentation analyses and in cell-free DNA
Regulatory T Cells Are Dispensable for Tolerance to RBC Antigens
Autoimmune hemolytic anemia (AIHA) occurs when pathogenic autoantibodies against red blood cell (RBC) antigens are generated. Whilst the basic disease pathology of AIHA is well studied, the underlying mechanism(s) behind the failure in tolerance to RBC autoantigens are poorly understood. Thus, to investigate the tolerance mechanisms required for the establishment and maintenance of tolerance to RBC antigens, we developed a novel murine model. With this model, we evaluated the role of regulatory T cells (Tregs) in tolerance to RBC-specific antigens. Herein, we show that neither sustained depletion of Tregs nor immunization with RBC-specific proteins in conjunction with Treg depletion led to RBC-specific autoantibody generation. Thus, these studies demonstrate that Tregs are not required to prevent autoantibodies to RBCs and suggest that other tolerance mechanisms are likely involved
Major Miocene exhumation by fault-propagation folding within a metamorphosed, early Paleozoic thrust belt: Northwestern Argentina
The central Andean retroarc thrust belt is characterized by a southward transition at ∼22°S in structural style (thin-skinned in Bolivia, thick-skinned in Argentina) and apparent magnitude of Cenozoic shortening (>100 km more in the north). With the aim of evaluating the abruptness and cause of this transition, we conducted a geological and geo-thermochronological study of the Cachi Range (∼24–25°S), which is a prominent topographic feature at this latitude. Our U-Pb detrital zircon results from the oldest exposed rocks (Puncoviscana Formation) constrain deposition to mainly Cambrian time, followed by major, Cambro-Ordovician shortening and ∼484 Ma magmatism. Later, Cretaceous rift faults were locally inverted during Cenozoic shortening. Coupled with previous work, our new (U-Th)/He zircon results require 8–10 km of Miocene exhumation that was likely associated with fault-propagation folding within the Cachi Range. After Miocene shortening, displacement on sinistral strike-slip faults demonstrates a change in stress state to a non-vertically orientedσ3. This change in stress state may result from an increase in gravitational potential energy in response to significant crustal thickening and/or lithospheric root removal. Our finding of localized Cenozoic shortening in the Cachi Range increases the estimate of the local magnitude of shortening, but still suggests that significantly less shortening was accommodated south of the thin-skinned Bolivian fold-thrust belt. Our results also underscore the importance of the pre-existing stratigraphic and structural architecture in orogens in influencing the style of subsequent deformation.Fil: Pearson, D. M.. Qualités psychométriques du Consumer satisfaction questionnaire (CSQ-8) et du Helping alliance questionnaire (HAQ) [Psychometric properties of the Consumer Satisfaction Questionnaire (CSQ-8) and the Helping Alliance Questionnaire (HAQ)].
BACKGROUND: This study was based on data from a quality of care assessment survey conducted in 2011 in outpatient polyclinics of the Vaud Canton in Switzerland, comprising questionnaires completed by 568 children over the age often and 672 parents of children of all ages. The objective of this study was to evaluate the psychometric qualities of the eight-item French versions for children of the Helping Alliance Questionnaire (HAQ) and the Consumer Satisfaction Questionnaire (CSQ-8) to allow formal validation and clinical application of these tools in the context of French-speaking child psychiatry.
METHODOLOGY: Responses from children over the age often to the HAQ and CSQ-8 questionnaires were submitted to confirmatory factorial analysis (CFA) for ordinal data to verify their good fit with the original long versions. Construct validity (correspondence between scores on the scales and other external criteria considered to evaluate similar concepts) of the child questionnaires was tested by Spearman's correlation with the parents' responses and their feeling of being reassured or in agreement with respect to the first visit, and with the perception of the help provided by individual and family interviews.
RESULTS: CFA showed an acceptable fit with the one-dimensional model of the original scales, both for the HAQ and the CSQ-8. Significant positive correlations of the scales with the parents' responses and with other convergent external criteria confirmed the good construct validity.
CONCLUSIONS: These psychometric analyses provide a basis for the validation and clinical application of the abridged French versions of the HAQ and CSQ-8 in quality of care assessment in child psychiatry
