Linking a Pharmaceutical Chemistry Workshop to Pharmacy Practice
This paper describes the design and implementation of a workshop to enhance pharmacy students’ appreciation of the importance of chemistry for pharmacy practice. The workshop was designed to form part of the practical work of two modules taught in the second year of the MPharm degree. In this mandatory workshop, second year pharmacy students were required to spot in the dispensary drugs based on their chemical properties like chirality, their origin and chemical structure. The lecturers involved in the workshop showed examples of the application of chemistry in the day to day work of the dispensary (e.g. calculating the dose for a patient in millimoles or how small modifications from a natural product can change its ability to cross the blood-brain-barrier). Feedback from participating students was collected via two survey instruments to examine the impact of the intervention. The survey results showed a clear shift towards a more positive perception by students of the chemistry taught in the MPharm curriculum
Relative Sea-Level Change in the Northern Strait of Georgia, British Columbia
Twenty-four new radiocarbon dates from isolation basin cores, excavations and natural exposures, and an archeological site, constrain relative sea-level change since the last glaciation in the northern Strait of Georgia, British Columbia. Relative sea level fell rapidly from about 150 m elevation to 45 m elevation from 11 750 to 11 000 BP (13 750 to 13 000 cal BP), then its rate of fall slowed. The initial rapid emergence began soon after the transition from proximal to distal glaciomarine sedimentation, when the glacial front retreated from the Strait of Georgia and the Earth’s surface was unloaded. A sea-level lowstand a few metres below present-day sea level may have occurred in the early Holocene, but sea level was near its present level by 2000 BP. Sea-level change in the northern Strait of Georgia lagged the mid Strait of Georgia, 80 km to the south, by a few hundred years during initial emergence. The lowstand in the northern strait was later and probably shallower than in the mid strait. Isostatic depression inferred from the sea-level observations can be fit with two decaying exponential terms with characteristic decay times of 500 and 2600 years. The faster decay time corresponds to a shallow mantle viscosity of about 1019 Pa s, consistent with previous glacio-isostatic modelling. The present-day crustal uplift rate from the residual isostatic effects of the Cordilleran Ice Sheet is about 0.25 mm/a. Crustal uplift is not expected to significantly ameliorate projected sea-level rise in the mid and northern Strait of Georgia because present-day vertical crustal movements are inferred to be small.Vingt-quatre nouvelles dates au 14C provenant de carottes sédimentaires de bassins isolés, d’excavations, de coupes naturellement exposées et archéologiques permettent de cerner les changements du niveau marin relatif depuis la dernière glaciation dans la partie nord du détroit de Géorgie, en Colombie-Britannique. Le niveau marin relatif a rapidement passé de 150 m à 45 m d’altitude entre 11 750 et 11 000 ans BP (13 750-13 000 cal. BP), le taux d’abaissement ayant ralenti par la suite. L’émersion initiale rapide correspond à la fin de la sédimentation glaciomarine distale qui accompagnait le retrait du front glaciaire. Un bas niveau de quelques mètres sous le niveau actuel a pu s’instaurer durant l’Holocène inférieur, jusqu’en 2000 ans BP. Au début, le soulèvement isostatique a accusé un retard de quelques siècle sur celui de la zone du détroit à 80 km plus au sud. Le bas niveau de la partie nord du détroit est survenu plus tard et fut moins prononcé. Les données indiquent un enfoncement isostatique de type exponentiel avec des constantes de désintégration de 500 et de 2600 ans 14C. Le taux le plus rapide correspond à une viscosité du manteau terrestre d’environ 1019 Pa s, ce qui concorde avec les résultats de la modélisation glacio-isostatique. Le taux de soulèvement glacio-isostatique actuel résultant de l’inlandsis de la Cordillère s’établit à environ 0,25 mm/a. Ce taux ne peut contrer les effets du rehaussement du niveau marin prévu pour cette région en raison des faibles mouvements de la croûte terrestre qui y sont envisagés
PhenDisco: phenotype discovery system for the database of genotypes and phenotypes.
The database of genotypes and phenotypes (dbGaP) developed by the National Center for Biotechnology Information (NCBI) is a resource that contains information on various genome-wide association studies (GWAS) and is currently available via NCBI's dbGaP Entrez interface. The database is an important resource, providing GWAS data that can be used for new exploratory research or cross-study validation by authorized users. However, finding studies relevant to a particular phenotype of interest is challenging, as phenotype information is presented in a non-standardized way. To address this issue, we developed PhenDisco (phenotype discoverer), a new information retrieval system for dbGaP. PhenDisco consists of two main components: (1) text processing tools that standardize phenotype variables and study metadata, and (2) information retrieval tools that support queries from users and return ranked results. In a preliminary comparison involving 18 search scenarios, PhenDisco showed promising performance for both unranked and ranked search comparisons with dbGaP's search engine Entrez. 11 x 11 Domineering is Solved: The first player wins
We have developed a program called MUDoS (Maastricht University Domineering
Solver) that solves Domineering positions in a very efficient way. This enables
the solution of known positions so far (up to the 10 x 10 board) much quicker
(measured in number of investigated nodes).
More importantly, it enables the solution of the 11 x 11 Domineering board, a
board up till now far out of reach of previous Domineering solvers. The
solution needed the investigation of 259,689,994,008 nodes, using almost half a
year of computation time on a single simple desktop computer. The results show
that under optimal play the first player wins the 11 x 11 Domineering game,
irrespective if Vertical or Horizontal starts the game.
In addition, several other boards hitherto unsolved were solved. Using the
convention that Vertical starts, the 8 x 15, 11 x 9, 12 x 8, 12 x 15, 14 x 8,
and 17 x 6 boards are all won by Vertical, whereas the 6 x 17, 8 x 12, 9 x 11,
and 11 x 10 boards are all won by Horizontal
Using small-angle X-ray scattering to investigate the compaction behaviour of a granulated clay
The compaction behaviour of a commercial granulated clay (magnesium aluminium smectite, gMgSm) was investigated using macroscopic pressure-density measurements, X-ray diffraction (XRD), scanning electron microscopy (SEM), X-ray microtomography (XμT) and small-angle X-ray scattering (SAXS). This material was studied as a potential compaction excipient for pharmaceutical tabletting, but also as a model system demonstrating the capabilities of SAXS for investigating compaction in other situations.
Bulk compaction measurements showed that the gMgSm was more difficult to compact than polymeric pharmaceutical excipients such as spheronised microcrystalline cellulose (sMCC), corresponding to harder granules. Moreover, in spite of using lubrication (magnesium stearate) on the tooling surfaces, rather high ejection forces were observed, which may cause problems during commercial tabletting, requiring further amelioration. Although the compacted gMgSm specimens were more porous, however, they still exhibited acceptable cohesive strengths, comparable to sMCC. Hence, there may be scope for using granular clay as one component of a tabletting formulation.
Following principles established in previous work, SAXS revealed information concerning the intragranular structure of the gMgSm and its response to compaction. The results showed that little compression of the intragranular morphology occurred below a relative density of 0 · 6, suggesting that granule rearrangements or fragmentation were the dominant mechanisms during this stage. By contrast, granule deformation became considerably more important at higher relative density, which also coincided with a significant increase in the cohesive strength of compacted specimens.
Spatially-resolved SAXS data was also used to investigate local variations in compaction behaviour within specimens of different shape. The results revealed the expected patterns of density variations within flat-faced cylindrical specimens. Significant variations in density, the magnitude of compressive strain and principal strain direction were also revealed in the vicinity of a debossed feature (a diametral notch) and within bi-convex specimens. The variations in compaction around the debossed notch, with a small region of high density below and low density along the flanks, appeared to be responsible for extensive cracking, which could also cause problems in commercial tabletting
Methionine 129 variant of human prion protein oligomerizes more rapidly than the valine 129 variant - Implications for disease susceptibility to Creutzfeldt-Jakob disease
The human PrP gene (PRNP) has two common alleles that encode either methionine or valine at codon 129. This polymorphism modulates disease susceptibility and phenotype of human transmissible spongiform encyphalopathies, but the molecular mechanism by which these effects are mediated remains unclear. Here, we compared the misfolding pathway that leads to the formation of beta-sheet-rich oligomeric isoforms of the methionine 129 variant of PrP to that of the valine 129 variant. We provide evidence for differences in the folding behavior between the two variants at the early stages of oligomer formation. We show that Met(129) has a higher propensity to form beta-sheet-rich oligomers, whereas Val(129) has a higher tendency to fold into alpha-helical-rich monomers. An equimolar mixture of both variants displayed an intermidate folding behavior. We show that the oligomers of both variants are initially a mixture of alpha- and beta-rich conformers that evolve with time to an increasingly homogeneous beta-rich form. This maturation process, which involves no further change in proteinase K resistance, occurs more rapidly in the Met(129) form than the Val(129) form. Although the involvement of such beta-rich oligomers in prion pathogenesis is speculative, the misfolding behavior could, in part, explain the higher susceptibility of individuals that are methionine homozygote to both sporadic and variant Creutzfeldt-Jakob disease.</p
The Rank four Heterotic Modular Invariant Partition Functions
In this paper, we develop several general techniques to investigate modular
invariants of conformal field theories whose algebras of the holomorphic and
anti-holomorphic sectors are different. As an application, we find all such
``heterotic'' WZNW physical invariants of (horizontal) rank four: there are
exactly seven of these, two of which seem to be new. Previously, only those of
