Ecology: a prerequisite for malaria elimination and eradication
* Existing front-line vector control measures, such as insecticide-treated nets and residual sprays, cannot break the transmission cycle of Plasmodium falciparum in the most intensely endemic parts of Africa and the Pacific
* The goal of malaria eradication will require urgent strategic investment into understanding the ecology and evolution of the mosquito vectors that transmit malaria

Heat acclimatization blunts copeptin responses to hypertonicity from dehydrating exercise in humans.
Acclimatization favors greater extracellular tonicity from lower sweat sodium, yet hyperosmolality may impair thermoregulation during heat stress. Enhanced secretion or action of vasopressin could mitigate this through increased free water retention. Aims were to determine responses of the vasopressin surrogate copeptin to dehydrating exercise and investigate its relationships with tonicity during short and long-term acclimatization. Twenty-three participants completed a structured exercise programme following arrival from a temperate to a hot climate. A Heat Tolerance Test (HTT) was conducted on Day-2, 6, 9 and 23, consisting of 60-min block-stepping at 50% VO2 peak, with no fluid intake. Resting sweat [Na+ ] was measured by iontophoresis. Changes in body mass (sweat loss), core temperature, heart rate, osmolality (serum and urine) and copeptin and aldosterone (plasma) were measured with each Test. From Day 2 to Day 23, sweat [Na+ ] decreased significantly (adjusted P < 0.05) and core temperature and heart rate fell. Over the same interval, HTT-associated excursions were increased for serum osmolality (5 [-1, 9] vs. 9 [5, 12] mosm·kg-1 ), did not differ for copeptin (9.6 [6.0, 15.0] vs. 7.9 [4.3, 14.7] pmol·L-1 ) and were reduced for aldosterone (602 [415, 946] vs. 347 [263, 537] pmol·L-1 ). Urine osmolality was unchanging and related consistently to copeptin at end-exercise, whereas the association between copeptin and serum osmolality was right-shifted (P = 0.0109) with acclimatization. Unchanging urine:serum osmolality argued against increased renal action of vasopressin. In conclusion, where exercise in the heat is performed without fluid replacement, heat acclimatization does not appear to enhance AVP-mediated free water retention in humans
Holocene relative sea-level changes in northwest Ireland: An empirical test for glacial isostatic adjustment models
The late-Quaternary relative sea-level (RSL) history of Ireland is complex, positioned at the margins of the former British-Irish Ice Sheet, and subject to the influence of ice unloading and forebulge collapse. Geophysical models of post-glacial isostatic adjustment (GIA) provide estimates of the pattern of RSL change since deglaciation which may be tested and validated with empirical data from proxy records. For the region of northwest Ireland, there is a paucity of high-quality RSL data and, therefore, equivocal evidence to support the GIA models that predict a mid to Late-Holocene RSL highstand of between +0.5 and +2 m above present. This study aims to investigate this model-data discrepancy by reconstructing RSL change from a near continuous salt-marsh sequence at Bracky Bridge, Donegal, spanning the last ca. 2500 years. We develop a transfer function model to reconstruct the vertical position of sea level using a regional diatom training set to quantify the indicative meaning and predict the palaeomarsh elevation of the core samples. A chronology is provided by a combination of 14 C and 210 Pb data, with sample specific ages derived from an age-depth model using a Bayesian framework. Our reconstruction shows ca. 2 m of relative sea-level rise in the past 2500 years. This is not compatible with some previously published sea-level index points from the region, which we re-interpret as freshwater/terrestrial limiting data. These results do not provide any evidence to support a Mid-Holocene RSL highstand above present sea level. Whilst none of the available GIA models replicate the timing and magnitude of the Late-Holocene RSL rise in our reconstruction, those which incorporate a thick and extensive British-Irish Sea Ice Sheet provide the best fit
Current tidal power technologies and their suitability for applications in coastal and marine areas
A considerable body of research is currently being performed to quantify available tidal energy resources and to develop efficient devices with which to harness them. This work is naturally focussed on maximising power generation from the most promising sites, and a review of the literature suggests that the potential for smaller scale, local tidal power generation from shallow near-shore sites has not yet been investigated. If such generation is feasible, it could have the potential to provide sustainable electricity for nearby coastal homes and communities as part of a distributed generation strategy, and would benefit from easier installation and maintenance, lower cabling and infrastructure requirements and reduced capital costs when compared with larger scale projects. This article reviews tidal barrages and lagoons, tidal turbines, oscillating hydrofoils and tidal kites to assess their suitability for small-scale electricity generation in shallow waters. This is achieved by discussing the power density, scalability, durability, maintainability, economic potential and environmental impacts of each concept. The performance of each technology in each criterion is scored against axial-flow turbines, allowing for them to be ranked according to their overall suitability. The review suggests that tidal kites and range devices are not suitable for small-scale shallow water applications due to depth and size requirements respectively. Cross-flow turbines appear to be the most suitable technology, as they have high power densities and a maximum size that is not constrained by water depth
Effectiveness of short term heat acclimation on intermittent sprint performance with moderately trained females controlling for menstrual cycle phase
YesIntroduction: Investigate the effectiveness of short-term heat acclimation (STHA), over
5-days (permissive dehydration), on an intermittent sprint exercise protocol (HST) with
females. Controlling for menstrual cycle phase.

participated. The HST (31.0◦C; 50%RH) was 9 × 5 min (45-min) of intermittent exercise,
based on exercise intensities of female soccer players, using a motorized treadmill and
Wattbike. Participants completed HST1 vs. HST2 as a control (C) trial. Followed by
90 min, STHA (no fluid intake), for five consecutive days in 39.5◦C; 60%RH, using
controlled-hyperthermia (∼rectal temperature [Tre] 38.5◦C). The HST3 occurred within
1 week after STHA. The HST2 vs HST3 trials were in the luteal phase, using self-reported
menstrual questionnaire and plasma 17β-estradiol.
Results: Pre (HST2) vs post (HST3) STHA there was a reduction at 45-min in Tre by
0.20◦C (95%CI −0.30 to −0.10◦C; d = 0.77); Tsk (−0.50; −0.90 to −0.10◦C; d = 0.80);
and Tb (−0.25; −0.35 to −0.15◦C; d = 0.92). Cardiac frequency reduced at 45-min
(−8; −16 to −1 b·min−1
; d = 1.11) and %PV increased (7.0; −0.4 to 14.5%: d = 1.27).
Mean power output increased across all nine maximal sprints by 56W (−26 to 139W;
d = 0.69; n = 9). There was limited difference (P > 0.05) for these measures in HST1
vs HST2 C trial.
Discussion: Short-term heat acclimation (5-days) using controlled-hyperthermia, leads
to physiological adaptation during intermittent exercise in the heat, in moderately trained
females when controlling for menstrual cycle phase
Multiple novel prostate cancer susceptibility signals identified by fine-mapping of known risk loci among Europeans
Genome-wide association studies (GWAS) have identified numerous common prostate cancer (PrCa) susceptibility loci. We have
fine-mapped 64 GWAS regions known at the conclusion of the iCOGS study using large-scale genotyping and imputation in
25 723 PrCa cases and 26 274 controls of European ancestry. We detected evidence for multiple independent signals at 16
regions, 12 of which contained additional newly identified significant associations. A single signal comprising a spectrum of
correlated variation was observed at 39 regions; 35 of which are now described by a novel more significantly associated lead SNP,
while the originally reported variant remained as the lead SNP only in 4 regions. We also confirmed two association signals in
Europeans that had been previously reported only in East-Asian GWAS. Based on statistical evidence and linkage disequilibrium
(LD) structure, we have curated and narrowed down the list of the most likely candidate causal variants for each region.
Functional annotation using data from ENCODE filtered for PrCa cell lines and eQTL analysis demonstrated significant
enrichment for overlap with bio-features within this set. By incorporating the novel risk variants identified here alongside the
refined data for existing association signals, we estimate that these loci now explain ∼38.9% of the familial relative risk of PrCa,
an 8.9% improvement over the previously reported GWAS tag SNPs. This suggests that a significant fraction of the heritability of
PrCa may have been hidden during the discovery phase of GWAS, in particular due to the presence of multiple independent
signals within the same regio
Linking Scottish vital event records using family groups
Funding: This work was supported by ESRC Grants ES/K00574X/2 “Digitising Scotland” and ES/L007487/1 “Administrative Data Research Centre – Scotland.”The reconstitution of populations through linkage of historical records is a powerful approach to generate longitudinal historical microdata resources of interest to researchers in various fields. Here we consider automated linking of the vital events recorded in the civil registers of birth, death and marriage compiled in Scotland, to bring together the various records associated with the demographic events in the life course of each individual in the population. From the histories, the genealogical structure of the population can then be built up. Rather than apply standard linkage techniques to link the individuals on the available certificates, we explore an alternative approach, inspired by the family reconstitution techniques adopted by historical demographers, in which the births of siblings are first linked to form family groups, after which intergenerational links between families can be established. We report a small-scale evaluation of this approach, using two district-level data sets from Scotland in the late nineteenth century, for which sibling links have already been created by demographers. We show that quality measures of up to 83% can be achieved on these data sets (using F-Measure, a combination of precision and recall). In the future, we intend to compare the results with a standard linkage approach and to investigate how these various methods may be used in a project which aims to link the entire Scottish population from 1856 to 1973.PostprintPeer reviewe
Stability analysis of electrical powered wheelchair-mounted robotic-assisted transfer device
Infection of the malaria mosquito, Anopheles gambiae, with two species of entomopathogenic fungi: effects of concentration, co-formulation, exposure time and persistence
<p>Abstract</p> <p>Background</p> <p>Entomopathogenic fungi <it>Metarhizium anisopliae </it>and <it>Beauveria bassiana </it>isolates have been shown to infect and reduce the survival of mosquito vectors.</p> <p>Methods</p> <p>Here four different bioassays were conducted to study the effect of conidia concentration, co-formulation, exposure time and persistence of the isolates <it>M. anisopliae </it>ICIPE-30 and <it>B. bassiana </it>I93-925 on infection and survival rates of female <it>Anopheles gambiae sensu stricto</it>. Test concentrations and exposure times ranged between 1 × 10<sup>7 </sup>- 4 × 10<sup>10 </sup>conidia m<sup>-2 </sup>and 15 min - 6 h. In co-formulations, 2 × 10<sup>10 </sup>conidia m<sup>-2 </sup>of both fungus isolates were mixed at ratios of 4:1, 2:1, 1:1,1:0, 0:1, 1:2 and 1:4. To determine persistence, mosquitoes were exposed to surfaces treated 1, 14 or 28 d previously, with conidia concentrations of 2 × 10<sup>9</sup>, 2 × 10<sup>10 </sup>or 4 × 10<sup>10</sup>.</p> <p>Results</p> <p>Mosquito survival varied with conidia concentration; 2 × 10<sup>10 </sup>conidia m<sup>-2 </sup>was the concentration above which no further reductions in survival were detectable for both isolates of fungus. The survival of mosquitoes exposed to single and co-formulated treatments was similar and no synergistic or additive effects were observed. Mosquitoes were infected within 30 min and longer exposure times did not result in a more rapid killing effect. Fifteen min exposure still achieved considerable mortality rates (100% mortality by 14 d) of mosquitoes, but at lower speed than with 30 min exposure (100% mortality by 9 d). Conidia remained infective up to 28 d post-application but higher concentrations did not increase persistence.</p> <p>Conclusion</p> <p>Both fungus isolates are effective and persistent at low concentrations and short exposure times.</p
