    Chitosanase may enhance anti-fungal defense responses in transgenic tobacco

    Chitosanase is an enzyme, similar to chitinase, capable of hydrolyzing the β-1,4-linkages between N-acetyl-D-glucosamine and D-glucosamine residues in partially acetylated chitosan polymers found in fungal cell walls. When attacked by pathogenic fungi, many plants exploit this hydrolytic action as a component of a larger post-attack defense response, but these enzymes may also play a role in the initial plant-pathogen interaction via the generation of elicitors resulting from the hydrolysis of fungal cell walls. To gain insight into these mechanisms, a Paenbacillus chitosanase was cloned, sequenced, and modified for plant expression. The modified gene was delivered to tobacco (Nicotiana tabacum L. cv. Xanthine) leaf disks via Agrobacterium tumenfaciensmediated transformation. Whole plants were regenerated from the transformed cells. The putative transformants were tested for transgene integration, transcription, and translation. Confirmed transformants were then screened for enhanced responses to a Rhizoctonia solani cellwall preparation by measuring time-course production of hydrogen peroxide, phenylalanine ammonia lyase, and peroxidase. These compounds play roles at different points in a pathogenesis-related signal transduction pathway and thus allow for an initial assessment of the global defense response. Preliminary data suggest that transgenic tobacco constitutively expressing a Paenbacillus chitosanase may activate pathogenesis-related defense responses more quickly than wild type tobacco

    Fungicides are expensive, dangerous, and can be harmful to the environment, but they are often necessary for profitable farming operations. New technologies may soon allow farmers to replace these chemicals with genetically engineered plants producing antifungal enzymes that degrade fungal cell walls. To explore this option, a Paenbacillus chitosanase gene was cloned, sequenced, and modified for plant expression. The modified gene was delivered to tobacco (Nicotiana tabacum L cv.     Energy spectrum of turbulent fluctuations in boundary driven reduced magnetohydrodynamics

    The nonlinear dynamics of a bundle of magnetic flux ropes driven by stationary fluid motions at their endpoints is studied, by performing numerical simulations of the magnetohydrodynamic (MHD) equations. The development of MHD turbulence is shown, where the system reaches a state that is characterized by the ratio between the Alfven time (the time for incompressible MHD waves to travel along the field lines) and the convective time scale of the driving motions. This ratio of time scales determines the energy spectra and the relaxation toward different regimes ranging from weak to strong turbulence. A connection is made with phenomenological theories for the energy spectra in MHD turbulence.Comment: Published in Physics of Plasma

    MHD consistent cellular automata (CA) models II. Applications to solar flares

    In Isliker et al. (2000b), an extended cellular automaton (X-CA) model for solar flares was introduced. In this model, the interpretation of the model's grid-variable is specified, and the magnetic field, the current, and an approximation to the electric field are yielded, all in a way that is consistent with Maxwell's and the MHD equations. Here, we reveal which relevant plasma physical processes are implemented by the X-CA model and in what form, and what global physical set-up is assumed by this model when it is in its natural state (SOC). The basic results are: (1) On large-scales, all variables show characteristic quasi-symmetries. (2) The global magnetic topology forms either (i) closed magnetic field lines, or (ii) an arcade of field lines above the bottom plane line, if the model is slightly modified. (3) In case of the magnetic topology (ii), loading can be interpreted as if there were a plasma which flows predominantly upwards, whereas in case of the magnetic topology (i), as if there were a plasma flow expanding from the neutral line. (4) The small-scale physics in the bursting phase represent localized diffusive processes. (5) The local diffusivity usually has a value which is effectively zero, and it turns locally to an anomalous value if a threshold is exceeded, whereby diffusion dominates the quiet evolution (loading). (6) Flares (avalanches) are accompanied by the appearance of localized, intense electric fields. (7) In a variant on the X-CA model, the magnitude of the current is used directly in the instability criterion. First results indicate that the SOC state persists. (8) The current-dissipation during flares is spatially fragmented into a large number of dissipative current-surfaces of varying sizes, which show a highly dynamic temporal evolution.Comment: 13 pages, 12 figures; in press at Astronomy and Astrophysics (2001

    Game-based approaches to learning are increasingly being recognized as having the potential to stimulate intrinsic motivation amongst learners. Whilst a range of examples of effective serious games exist, creating the high-fidelity content with which to populate a serious game is resource-intensive task. To reduce this resource requirement, research is increasingly exploring means to reuse and repurpose existing games and relevant sources of content. Education has proven a popular application area for Adaptive Hypermedia, as adaptation can offer enriched learning experiences to students. Whilst content to-date has mainly been in the form of rich text, various efforts have been made to integrate Serious Games into Adaptive Hypermedia via run-time adaptation engines. However, there is little in the way of effective integrated authoring and user modeling support for these efforts. This paper explores avenues for effectively integrating serious games into adaptive hypermedia. In particular, we consider authoring and user modeling aspects in addition to integration into run-time adaptation engines, thereby enabling authors to create Adaptive Hypermedia that includes an adaptive game, thus going beyond mere selection of a suitable game and towards an approach with the capability to adapt and respond to the needs of learners and educators

Using these classical, reformed English discussions of education and of the nature of children reveals an anxiety about the status of the child. Laughing appropriately - using tile mind and not merely the body - is a key feature of being human, and as such, the child's lack of "true' laughter reveals that child's status to be never always-already human.     Reliability and efficiency at global level in power systems

    In EU project E-Price it is proposed to attribute reliability to the Transmission System Operator (TSO) and the drive for efficiency to the Balance Responsible Parties (BRP). Two main ideas are proposed. The first idea is to make BRPs themselves responsible for estimating their uncertainty in real time and hedge their imbalance risks, yielding the proposal to introduce dual-sided markets for ancillary services. The second idea is to reduce the conservatism in exploiting the inter-area transmission capacity (ATC), while still guaranteeing sufficient degrees of reliability. This paper explains, elucidates and concludes, based on qualitative arguments and supported by quantitative simulations and calculations, that both proposals are beneficial to improve the stated compromise between reliability and economy
