The promoter from SlREO, a highly-expressed, root-specific Solanum lycopersicum gene, directs expression to cortex of mature roots
Root-specific promoters are valuable tools for targeting transgene expression, but many of those already described have limitations to their general applicability. We present the expression characteristics of SlREO, a novel gene isolated from tomato (Solanum lycopersicum L.). This gene was highly expressed in roots but had a very low level of expression in aerial plant organs. A 2.4-kb region representing the SlREO promoter sequence was cloned upstream of the uidA GUS reporter gene and shown to direct expression in the root cortex. In mature, glasshouse-grown plants this strict root specificity was maintained. Furthermore, promoter activity was unaffected by dehydration or wounding stress but was somewhat suppressed by exposure to NaCl, salicylic acid and jasmonic acid. The predicted protein sequence of SlREO contains a domain found in enzymes of the 2-oxoglutarate and Fe(II)-dependent dioxygenase superfamily. The novel SlREO promoter has properties ideal for applications requiring strong and specific gene expression in the bulk of tomato root tissue growing in soil, and is also likely to be useful in other Solanaceous crop
Compensation of Strong Thermal Lensing in High Optical Power Cavities
In an experiment to simulate the conditions in high optical power advanced
gravitational wave detectors such as Advanced LIGO, we show that strong thermal
lenses form in accordance with predictions and that they can be compensated
using an intra-cavity compensation plate heated on its cylindrical surface. We
show that high finesse ~1400 can be achieved in cavities with internal
compensation plates, and that the cavity mode structure can be maintained by
thermal compensation. It is also shown that the measurements allow a direct
measurement of substrate optical absorption in the test mass and the

Too Embedded to Fail: the ECB and the Necessity of Calculating Europe
Calling into question the meaning of "independence" in contemporary central banking, the present article investigates the social origins and post-crisis persistence of the European Central Bank's (ECB) core macroeconomic model, despite broad acknowledgement of its failure to anticipate the financial crisis. We trace the making of the model; the process by which it became dominant in European central banking and beyond; criticism in the wake of its failure to predict the financial-cum-Eurozone crisis; and its persistence, nonetheless, in the crisis’ aftermath. We argue that the formation, meanings, and persistence of the ECB's model cannot be understood as effects of the bank's independence or the model’s intrinsic qualities. Rather, the model's trajectory is best understood in light of the ECB’s transnationally embedded social location in international finance, professional economics, and European governing institutions. The necessity of calculating Europe, irrespective of the accuracy or predictive strength of the model being used, has less to do with the ECB's independence from domestic politics and more to do with its transnational embeddedness - or, stated differently, that the ECB is, in a sense, too embedded to fail
The Interface Region Imaging Spectrograph (IRIS)
The Interface Region Imaging Spectrograph (IRIS) small explorer spacecraft
provides simultaneous spectra and images of the photosphere, chromosphere,
transition region, and corona with 0.33-0.4 arcsec spatial resolution, 2 s
temporal resolution and 1 km/s velocity resolution over a field-of-view of up
to 175 arcsec x 175 arcsec. IRIS was launched into a Sun-synchronous orbit on
27 June 2013 using a Pegasus-XL rocket and consists of a 19-cm UV telescope
that feeds a slit-based dual-bandpass imaging spectrograph. IRIS obtains
spectra in passbands from 1332-1358, 1389-1407 and 2783-2834 Angstrom including
bright spectral lines formed in the chromosphere (Mg II h 2803 Angstrom and Mg
II k 2796 Angstrom) and transition region (C II 1334/1335 Angstrom and Si IV
1394/1403 Angstrom). Slit-jaw images in four different passbands (C II 1330, Si
IV 1400, Mg II k 2796 and Mg II wing 2830 Angstrom) can be taken simultaneously
with spectral rasters that sample regions up to 130 arcsec x 175 arcsec at a
variety of spatial samplings (from 0.33 arcsec and up). IRIS is sensitive to
emission from plasma at temperatures between 5000 K and 10 MK and will advance
our understanding of the flow of mass and energy through an interface region,
formed by the chromosphere and transition region, between the photosphere and
corona. This highly structured and dynamic region not only acts as the conduit
of all mass and energy feeding into the corona and solar wind, it also requires
an order of magnitude more energy to heat than the corona and solar wind
combined. The IRIS investigation includes a strong numerical modeling component
based on advanced radiative-MHD codes to facilitate interpretation of
observations of this complex region. Approximately eight Gbytes of data (after
compression) are acquired by IRIS each day and made available for unrestricted

Hybrid assembly with long and short reads improves discovery of gene family expansions
BACKGROUND: Long-read and short-read sequencing technologies offer competing advantages for eukaryotic genome sequencing projects. Combinations of both may be appropriate for surveys of within-species genomic variation. METHODS: We developed a hybrid assembly pipeline called "Alpaca" that can operate on 20X long-read coverage plus about 50X short-insert and 50X long-insert short-read coverage. To preclude collapse of tandem repeats, Alpaca relies on base-call-corrected long reads for contig formation. RESULTS: Compared to two other assembly protocols, Alpaca demonstrated the most reference agreement and repeat capture on the rice genome. On three accessions of the model legume Medicago truncatula, Alpaca generated the most agreement to a conspecific reference and predicted tandemly repeated genes absent from the other assemblies. CONCLUSION: Our results suggest Alpaca is a useful tool for investigating structural and copy number variation within de novo assemblies of sampled populations
All-sky search for periodic gravitational waves in LIGO S4 data
We report on an all-sky search with the LIGO detectors for periodic
gravitational waves in the frequency range 50-1000 Hz and with the frequency's
time derivative in the range -1.0E-8 Hz/s to zero. Data from the fourth LIGO
science run (S4) have been used in this search. Three different semi-coherent
methods of transforming and summing strain power from Short Fourier Transforms
(SFTs) of the calibrated data have been used. The first, known as "StackSlide",
averages normalized power from each SFT. A "weighted Hough" scheme is also
developed and used, and which also allows for a multi-interferometer search.
The third method, known as "PowerFlux", is a variant of the StackSlide method
in which the power is weighted before summing. In both the weighted Hough and
PowerFlux methods, the weights are chosen according to the noise and detector
antenna-pattern to maximize the signal-to-noise ratio. The respective
advantages and disadvantages of these methods are discussed. Observing no
evidence of periodic gravitational radiation, we report upper limits; we
interpret these as limits on this radiation from isolated rotating neutron
stars. The best population-based upper limit with 95% confidence on the
gravitational-wave strain amplitude, found for simulated sources distributed
isotropically across the sky and with isotropically distributed spin-axes, is
4.28E-24 (near 140 Hz). Strict upper limits are also obtained for small patches
on the sky for best-case and worst-case inclinations of the spin axes.Comment: 39 pages, 41 figures An error was found in the computation of the C

Searching for a Stochastic Background of Gravitational Waves with LIGO
The Laser Interferometer Gravitational-wave Observatory (LIGO) has performed
the fourth science run, S4, with significantly improved interferometer
sensitivities with respect to previous runs. Using data acquired during this
science run, we place a limit on the amplitude of a stochastic background of
gravitational waves. For a frequency independent spectrum, the new limit is
. This is currently the most sensitive
result in the frequency range 51-150 Hz, with a factor of 13 improvement over
the previous LIGO result. We discuss complementarity of the new result with
other constraints on a stochastic background of gravitational waves, and we
investigate implications of the new result for different models of this

Dispersing the Mists: An Experimental History of Medicine Study into the Quality of Volatile Inhalations

Revolutionary at the time for its ease of use and patient-friendliness, the inhaler is still in use for self-treatment by many all over the world. On the occasion of its 150th anniversary, this study reports an experimental historical medicine approach to identify evidence for the quality of vapor inhalers. Methods: Through accessing reviews of the device's use by the contemporary medical establishment, it was established that Dr. Nelson's Inhaler enjoyed a reputation of quality and efficacy among reputable physicians generating empirical evidence of clinical performance. There was a general absence of product performance tests during this period. Therefore, modern inhalation performance testing was applied to test the aerosol delivery performance for Friars' Balsam, and its key chemical constituent, benzoic acid (BA). Results: A respirable dose of 59.9 ± 9.0 μg of BA was aerosolized in a 10 minutes period from a dose of 3.3 mL Friars' Balsam (equivalent to 35.1 ± 0.2 mg of BA) in 375 mL of steaming water using the glass twin stage impinger at a flow rate of 60 L·min−1. The respirable dose from a standardized aqueous BA inhalation formulation increased from 115.9 ± 10.6 to 200.2 ± 19.9 μg by increasing the simulated inhalation period from 5 to 10 minutes. When tested with a simulated inhalation maneuver (500 mL tidal volume, 13 minutes−1 respiration rate, 1:2 inspiratory:expiratory ratio) a respirable dose of 112.8 ± 40.3 μg was produced. Conclusions: This work has highlighted the potential for aerosol drug delivery using steam inhalers that are popular with patients. Physicians should therefore be aware of the potential for lung dosing with irritants when patients self-medicate using the Nelson Inhaler with vaporizing formulations such as Friars' Balsam.Peer reviewedFinal Accepted Versio
