Mammary molecular portraits reveal lineage-specific features and progenitor cell vulnerabilities.
The mammary epithelium depends on specific lineages and their stem and progenitor function to accommodate hormone-triggered physiological demands in the adult female. Perturbations of these lineages underpin breast cancer risk, yet our understanding of normal mammary cell composition is incomplete. Here, we build a multimodal resource for the adult gland through comprehensive profiling of primary cell epigenomes, transcriptomes, and proteomes. We define systems-level relationships between chromatin-DNA-RNA-protein states, identify lineage-specific DNA methylation of transcription factor binding sites, and pinpoint proteins underlying progesterone responsiveness. Comparative proteomics of estrogen and progesterone receptor-positive and -negative cell populations, extensive target validation, and drug testing lead to discovery of stem and progenitor cell vulnerabilities. Top epigenetic drugs exert cytostatic effects; prevent adult mammary cell expansion, clonogenicity, and mammopoiesis; and deplete stem cell frequency. Select drugs also abrogate human breast progenitor cell activity in normal and high-risk patient samples. This integrative computational and functional study provides fundamental insight into mammary lineage and stem cell biology
Resonant Lifetime of Core-Excited Organic Adsorbates from First Principles
We investigate by first-principles simulations the resonant electron-transfer
lifetime from the excited state of an organic adsorbate to a semiconductor
surface, namely isonicotinic acid on rutile TiO(110). The
molecule-substrate interaction is described using density functional theory,
while the effect of a truly semi-infinite substrate is taken into account by
Green's function techniques. Excitonic effects due to the presence of
core-excited atoms in the molecule are shown to be instrumental to understand
the electron-transfer times measured using the so-called core-hole-clock
technique. In particular, for the isonicotinic acid on TiO(110), we find
that the charge injection from the LUMO is quenched since this state lies
within the substrate band gap. We compute the resonant charge-transfer times
from LUMO+1 and LUMO+2, and systematically investigate the dependence of the
elastic lifetimes of these states on the alignment among adsorbate and

Telomeres and the natural lifespan limit in humans
An ongoing debate in demography has focused on whether the human lifespan has a maximal natural limit. Taking a mechanistic perspective, and knowing that short telomeres are associated with diminished longevity, we examined whether telomere length dynamics during adult life could set a maximal natural lifespan limit. We define leukocyte telomere length of 5 kb as the 'telomeric brink', which denotes a high risk of imminent death. We show that a subset of adults may reach the telomeric brink within the current life expectancy and more so for a 100-year life expectancy. Thus secular trends in life expectancy should confront a biological limit due to crossing the telomeric brink
The future of mammary stem cell biology: the power of in vivo transplants
The recent review by Smith and Medina [1] of in vivo transplantation models and their role in investigating mammary stem cell (MaSC) biology provides comprehensive coverage of the history and complexity of the ‘gold standard ’ MaSC assay in mice. This includes a description of the pioneering studies that showed that mammary epithelial outgrowths can be generated in cleared mammary fat pads transplanted with explants or admixtures of mammary cells [2]. However, this approach clearly does not lend itself to prospective analysis of isolated subpopulations in order to identify which cells possess in vivo regenerative activity. More recently, success in obtaining complex mammary gland structures from transplanted suspensions of single cells has now made this possible [3-7]. El valor de los metadatos para las estaciones de recuperación de recursos del agua
Los metadatos hacen referencia a información descriptiva (como ubicación del sensor, unidad de medida, rango de medida, fecha de calibración, fecha de limpieza, si ocurrió algún evento como episodio de lluvia/fallo operativo/vertido tóxico ) que es esencial para convertir los grandes volúmenes de datos que se recogen actualmente en las instalaciones de tratamiento de agua y que están sin procesar en información y recursos útiles. Con el avance de la digitalización en el sector del agua, es fundamental evitar los cementerios de datos y, por otro lado, utilizar los datos almacenados para resolver problemas actuales y futuros. Este artículo se centra en el papel crucial que tienen los metadatos para responder a desafíos futuros y posiblemente impredecibles. El objetivo de este documento es presentar el 'reto de los metadatos' y destacar la necesidad de tener en cuenta los metadatos cuando se recoge información como parte de las buenas prácticas de digitalización. Metadata refers to descriptive information (such as sensor location, measurement unit, measurement range, calibration date, cleaning date, if any event occurred such as rain event/operating failure/toxic spill, ) essential to convert large volumes of raw data that are currently collected at water treatment facilities into useful information and resources. With the advance of digitalization in the water sector, it is fundamental to avoid data graveyards and, on the other hand, using collected data to address current and future problems. This paper focuses on the crucial role that metadata has in responding to future and possibly unpredictable challenges. The aim of this document is to present the 'metadata challenge' and to highlight the need to consider metadata when collecting information as part of good digitalization practices
Caecal metastasis from breast cancer presenting as intestinal obstruction
<p>Abstract</p> <p>Background</p> <p>Gastrointestinal metastsasis from the breast cancer are rare. We report a patient who presented with intestinal obstruction due to solitary caecal metastasis from infiltrating ductal carcinoma of breast. We also review the available literature briefly.</p> <p>Case presentation</p> <p>A 72 year old lady with past history of breast cancer presented with intestinal obstruction due to a caecal mass. She underwent an emergency right hemicolectomy. The histological examination of the right hemicolectomy specimen revealed an adenocarcinoma in caecum staining positive for Cytokeratin 7 and Carcinoembryonic antigen and negative for Cytokeratin 20, CDX2 and Estrogen receptor. Eight out of 11 mesenteric nodes showed tumour deposits. A histological diagnosis of metastatic breast carcinoma was given.</p> <p>Conclusion</p> <p>To the best of our knowledge, this is the first case report of solitary metastasis to caecum from infiltrating ductal carcinoma of breast. Awareness of this possibility will aid in appropriate management of such patients.</p
SCA-1 Labels a Subset of Estrogen-Responsive Bipotential Repopulating Cells within the CD24(+) CD49f(hi) Mammary Stem Cell-Enriched Compartment
Estrogen stimulates breast development during puberty and mammary tumors in adulthood through estrogen receptor-α (ERα). These effects are proposed to occur via ERα+ luminal cells and not the mammary stem cells (MaSCs) that are ERαneg. Since ERα+ luminal cells express stem cell antigen-1 (SCA-1), we sought to determine if SCA-1 could define an ERα+ subset of EpCAM+/CD24+/CD49fhi MaSCs. We show that the MaSC population has a distinct SCA-1+ population that is abundant in pre-pubertal mammary glands. The SCA-1+ MaSCs have less stem cell markers and less in vivo repopulating activity than their SCA-1neg counterparts. However, they express ERα and specifically enter the cell cycle at puberty. Using estrogen-deficient aromatase knockouts (ArKO), we showed that the SCA-1+ MaSC could be directly modulated by estrogen supplementation. Thus, SCA-1 enriches for an ERα+, estrogen-sensitive subpopulation within the CD24+/CD49fhi MaSC population that may be responsible for the hormonal sensitivity of the developing mammary gland
Nationwide genetic analysis of more than 600 families with inherited eye diseases in Argentina
This study corresponds to the first large-scale genetic analysis of inherited eye diseases (IED) in Argentina and describes the comprehensive genetic profile of a large cohort of patients. Medical records of 22 ophthalmology and genetics services throughout 13 Argentinian provinces were analyzed retrospectively. Patients with a clinical diagnosis of an ophthalmic genetic disease and a history of genetic testing were included. Medical, ophthalmological and family history was collected. A total of 773 patients from 637 families were included, with 98% having inherited retinal disease. The most common phenotype was retinitis pigmentosa (RP, 62%). Causative variants were detected in 379 (59%) patients. USH2A, RPGR, and ABCA4 were the most common disease-associated genes. USH2A was the most frequent gene associated with RP, RDH12 early-onset severe retinal dystrophy, ABCA4 Stargardt disease, PROM1 cone-rod dystrophy, and BEST1 macular dystrophy. The most frequent variants were RPGR c.1345 C > T, p.(Arg449*) and USH2A c.15089 C > A, p.(Ser5030*). The study revealed 156/448 (35%) previously unreported pathogenic/likely pathogenic variants and 8 possible founder mutations. We present the genetic landscape of IED in Argentina and the largest cohort in South America. This data will serve as a reference for future genetic studies, aid diagnosis, inform counseling, and assist in addressing the largely unmet need for clinical trials to be conducted in the region
