    Novel ETHE1 mutation in a carrier couple having prior offspring affected with ethylmalonic encephalopathy: Genetic analysis, clinical management and reproductive outcome.

    Ethylmalonic encephalopathy (EE) is an autosomally recessive inherited disorder with a relentlessly progressive decline in neurological function, usually fatal by the age of ten. It is characterised by generalised hypotonia, psychomotor regression, spastic tetraparesis, dystonia, seizures and, eventually, global neurological failure. Approximately 50 reports have been published worldwide describing this devastating disease, most involving patients of Mediterranean or Arab origin. The fundamental defect in EE likely involves the impairment of a mitochondrial sulphur dioxygenase coded by the ETHE1 gene responsible for the catabolism of sulphide, which subsequently accumulates to toxic levels. A diagnosis of EE should initiate careful genetic evaluation and counselling, particularly if the parents intend to have additional offspring. The present report describes the diagnosis of EE in a reproductive endocrinology context, where both members of a non-consanguineous couple were confirmed to be carriers of an identical A↷G mutation. This previously unknown mutation at nucleotide position c.494 resulted in an amino acid substitution, p.Asp165Gly. Although consideration was given to in vitro fertilisation, embryo biopsy and single gene pre-implantation genetic diagnosis, the couple decided to first utilise a less aggressive therapeutic approach with donor sperm insemination. Pregnancy with a low risk of EE was indeed achieved; however, the infant was affected with a different anomaly (hypoplastic left heart). As this case demonstrates, prior to the initiation of fertility therapy, genetic analysis may be used to provide a confirmatory diagnosis when EE is suspected

    Re-entrance of the metallic conductance in a mesoscopic proximity superconductor

    We present an experimental study of the diffusive transport in a normal metal near a superconducting interface, showing the re-entrance of the metallic conductance at very low temperature. This new mesoscopic regime comes in when the thermal coherence length of the electron pairs exceeds the sample size. This re-entrance is suppressed by a bias voltage given by the Thouless energy and can be strongly enhanced by an Aharonov Bohm flux.     Resolving the dusty circumstellar environment of the A[e] supergiant HD 62623 with the VLTI/MIDI

    B[e] stars are hot stars surrounded by circumstellar gas and dust responsible for the presence of emission lines and IR-excess in their spectra. How dust can be formed in this highly illuminated and diluted environment remains an open issue. HD 62623 is one of the very few A-type supergiants showing the B[e] phenomenon. We obtained nine calibrated visibility measurements using the VLTI/MIDI instrument in SCI-PHOT mode and PRISM spectral dispersion mode with projected baselines ranging from 13 to 71 m and with various position angles. We used geometrical models and physical modeling with a radiative transfer code to analyze these data. The dusty circumstellar environment of HD 62623 is partially resolved by the VLTI/MIDI even with the shortest baselines. The environment is flattened and can be separated into two components: a compact one whose extension grows from 17 mas at 8 microns to 30 mas at 9.6 microns and stays almost constant up to 13 microns, and a more extended one that is over-resolved even with the shortest baselines. Using the radiative transfer code MC3D, we managed to model HD 62623's circumstellar environment as a dusty disk with an inner radius of 3.85+-0.6 AU, an inclination angle of 60+-10 deg, and a mass of 2x10^-7Mo. It is the first time that the dusty disk inner rim of a supergiant star exhibiting the B[e] phenomenon is significantly constrained. The inner gaseous envelope likely contributes up to 20% to the total N band flux and acts like a reprocessing disk. Finally, the hypothesis of a stellar wind deceleration by the companion's gravitational effects remains the most probable case since the bi-stability mechanism does not seem to be efficient for this star.Comment: 13 pages, 11 figures.     Two dimensional smoothing via an optimised Whittaker smoother

    Background In many applications where moderate to large datasets are used, plotting relationships between pairs of variables can be problematic. A large number of observations will produce a scatter-plot which is difficult to investigate due to a high concentration of points on a simple graph. In this article we review the Whittaker smoother for enhancing scatter-plots and smoothing data in two dimensions. To optimise the behaviour of the smoother an algorithm is introduced, which is easy to programme and computationally efficient. Results The methods are illustrated using a simple dataset and simulations in two dimensions. Additionally, a noisy mammography is analysed. When smoothing scatterplots the Whittaker smoother is a valuable tool that produces enhanced images that are not distorted by the large number of points. The methods is also useful for sharpening patterns or removing noise in distorted images. Conclusion The Whittaker smoother can be a valuable tool in producing better visualisations of big data or filter distorted images. The suggested optimisation method is easy to programme and can be applied with low computational cost

    Toxic metal enrichment and boating intensity: sediment records of antifoulant copper in shallow lakes of eastern England

    Tributyltin (TBT), an aqueous biocide derived from antifouling paint pollution, is known to have impacted coastal marine ecosystems, and has been reported in the sediment of the Norfolk and Suffolk Broads, a network of rivers and shallow lakes in eastern England. In the marine environment, the 1987 TBT ban has resulted in expanded use of alternative biocides, raising the question of whether these products too have impacted the Broads ecosystem and freshwaters in general. Here we examine the lake sediment record in the Norfolk and Suffolk Broads for contamination by copper (Cu) (as an active biocide agent) and zinc (Zn) (as a component of booster biocides), to assess their occurrence and potential for causing environmental harm in freshwater ecosystems. We find that, after the introduction of leisure boating, there is a statistically significant difference in Cu enrichment between heavily and lightly boated sites, while no such difference exists prior to this time. At the heavily boated sites the onset of Cu enrichment coincides with a period of rapid increase in leisure boating. Such enrichment is maintained to the present day, with some evidence of continued increase. We conclude that Cu-based antifouling has measurably contaminated lakes exposed to boating, at concentrations high enough to cause ecological harm. Similar findings can be expected at other boated freshwater ecosystems elsewhere in the world

    Generic Mechanism of Emergence of Amyloid Protofilaments from Disordered Oligomeric aggregates

    The presence of oligomeric aggregates, which is often observed during the process of amyloid formation, has recently attracted much attention since it has been associated with neurodegenerative conditions such as Alzheimer's and Parkinson's diseases. We provide a description of a sequence-indepedent mechanism by which polypeptide chains aggregate by forming metastable oligomeric intermediate states prior to converting into fibrillar structures. Our results illustrate how the formation of ordered arrays of hydrogen bonds drives the formation of beta-sheets within the disordered oligomeric aggregates that form early under the effect of hydrophobic forces. Initially individual beta-sheets form with random orientations, which subsequently tend to align into protofilaments as their lengths increases. Our results suggest that amyloid aggregation represents an example of the Ostwald step rule of first order phase transitions by showing that ordered cross-beta structures emerge preferentially from disordered compact dynamical intermediate assemblies.Comment: 14 pages, 4 figure

    Ankyrin-mediated self-protection during cell invasion by the bacterial predator Bdellovibrio bacteriovorus

    Predatory Bdellovibrio bacteriovorus are natural antimicrobial organisms, killing other bacteria by whole-cell invasion. Self-protection against prey-metabolizing enzymes is important for the evolution of predation. Initial prey entry involves the predator’s peptidoglycan DD-endopeptidases, which decrosslink cell walls and prevent wasteful entry by a second predator. Here we identify and characterize a self-protection protein from B. bacteriovorus, Bd3460, which displays an ankyrin-based fold common to intracellular pathogens of eukaryotes. Co-crystal structures reveal Bd3460 complexation of dual targets, binding a conserved epitope of each of the Bd3459 and Bd0816 endopeptidases. Complexation inhibits endopeptidase activity and cell wall decrosslinking in vitro. Self-protection is vital — DBd3460 Bdellovibrio deleteriously decrosslink self-peptidoglycan upon invasion, adopt a round morpholog, and lose predatory capacity and cellular integrity. Our analysis provides the first mechanistic examination of self-protection in Bdellovibrio, documents protection-multiplicity for products of two different genomic loci, and reveals an important evolutionary adaptation to an invasive predatory bacterial lifestyle

    The peptidoglycan wall, located in the periplasm between the inner and outer membranes of the cell envelope in Gram-negative bacteria, maintains cell shape and endows osmotic robustness. Predatory Bdellovibrio bacteria invade the periplasm of other bacterial prey cells, usually crossing the peptidoglycan layer, forming transient structures called bdelloplasts within which the predators replicate. Prey peptidoglycan remains intact for several hours, but is modified and then degraded by predators escaping. Here we show predation is altered by deleting two Bdellovibrio N-acetylglucosamine (GlcNAc) deacetylases, one of which we show to have a unique two domain structure with a novel regulatory-”plug”. Deleting the deacetylases limits peptidoglycan degradation and rounded prey cell “ghosts” persist after mutant-predator exit. Mutant predators can replicate unusually in the periplasmic region between the peptidoglycan wall and the outer membrane rather than between wall and inner-membrane, yet still obtain nutrients from the prey cytoplasm. Deleting two further genes encoding DacB/PBP4 family proteins, known to decrosslink and round prey peptidoglycan, results in a quadruple mutant Bdellovibrio which leaves prey-shaped ghosts upon predation. The resultant bacterial ghosts contain cytoplasmic membrane within bacteria-shaped peptidoglycan surrounded by outer membrane material which could have promise as “bacterial skeletons” for housing artificial chromosomes
