watchpeopledie.tv
The thesis piece is a performance called watchpeopledie.tv performed four times at the Anderson Gallery in the Hidden Galleries. There are two rooms in the gallery. The first room is where the audience enters. There is a line on the floor marking that the audience is not to move further into the first room or into the second room. The audio consists of verbatim comments sourced from videos of violence against trans people from the website watchpeopledie.tv, a website in which users post real videos of shootings, suicides, car accidents, etc. The main text, which is my thesis writing below, also called watchpeopledie.tv, is an interpretation of these comments. It was included in the space during the performance as a booklet for the audience to read. The performance consists of me sitting naked on a stool, restricting all movement in order to remain completely still, yet trying to get up and leave the frame at the same time. When I do move my presence is disturbed by the walls separating the two rooms, the lines created by physical space/time. The performance lasts 3 hours.
The thesis writing is a performance text documenting and further enacting the thesis performance. It is a text made from performing an erasure poem on the text of the transphobic comments and then commenting on the erased words. It is accompanied by the Introduction, a trimmed transcript of an audio recording in which I describe how I came to make the thesis
Automated Species Classification Methods for Passive Acoustic Monitoring of Beaked Whales
The Littoral Acoustic Demonstration Center has collected passive acoustic monitoring data in the northern Gulf of Mexico since 2001. Recordings were made in 2007 near the Deepwater Horizon oil spill that provide a baseline for an extensive study of regional marine mammal populations in response to the disaster. Animal density estimates can be derived from detections of echolocation signals in the acoustic data. Beaked whales are of particular interest as they remain one of the least understood groups of marine mammals, and relatively few abundance estimates exist. Efficient methods for classifying detected echolocation transients are essential for mining long-term passive acoustic data. In this study, three data clustering routines using k-means, self-organizing maps, and spectral clustering were tested with various features of detected echolocation transients. Several methods effectively isolated the echolocation signals of regional beaked whales at the species level. Feedforward neural network classifiers were also evaluated, and performed with high accuracy under various noise conditions. The waveform fractal dimension was tested as a feature for marine biosonar classification and improved the accuracy of the classifiers. African Trypanosomes undermine humoral responses and vaccine development : link with inflammatory responses?
African trypanosomosis is a debilitating disease of great medical and socioeconomical importance. It is caused by strictly extracellular protozoan parasites capable of infecting all vertebrate classes including human, livestock, and game animals. To survive within their mammalian host, trypanosomes have evolved efficient immune escape mechanisms and manipulate the entire host immune response, including the humoral response. This report provides an overview of how trypanosomes initially trigger and subsequently undermine the development of an effective host antibody response. Indeed, results available to date obtained in both natural and experimental infection models show that trypanosomes impair homeostatic B-cell lymphopoiesis, B-cell maturation and survival and B-cell memory development. Data on B-cell dysfunctioning in correlation with parasite virulence and trypanosome-mediated inflammation will be discussed, as well as the impact of trypanosomosis on heterologous vaccine efficacy and diagnosis. Therefore, new strategies aiming at enhancing vaccination efficacy could benefit from a combination of (i) early parasite diagnosis, (ii) anti-trypanosome (drugs) treatment, and (iii) anti-inflammatory treatment that collectively might allow B-cell recovery and improve vaccination
Biological responses to spider silk - antibiotic fusion protein
The development of a new generation of multifunctional biomaterials is a continual goal for the
field of materials science. The in vivo functional behaviour of a new fusion protein that combines
the mechanical properties of spider silk with the antimicrobial properties of hepcidin was addressed
in this study. This new chimeric protein, termed 6mer + hepcidin, fuses spider dragline consensus
sequences (6mer) and the antimicrobial peptide hepcidin, as we have recently described, with
retention of bactericidal activity and low cytotoxicity. In the present study, mouse subcutaneous
implants were studied to access the in vivo biological response to 6mer + hepcidin, which were
compared with controls of silk alone (6mer), polylactic–glycolic acid (PLGA) films and empty defects.
Along with visual observations, flow cytometry and histology analyses were used to determine the
number and type of inflammatory cells at the implantation site. The results show a mild to low
inflammatory reaction to the implanted materials and no apparent differences between the 6mer +
hepcidin films and the other experimental controls, demonstrating that the new fusion protein has

Acute biphenotypic leukaemia: immunophenotypic and cytogenetic analysis
The incidence of acute biphenotypic leukaemia has ranged from less than 1% to almost 50% in various reports in the literature. This wide variability may be attributed to a number of reasons including lack of consistent diagnostic criteria, use of various panels of antibodies, and the failure to recognize the lack of lineage specificity of some of the antibodies used. The morphology, cytochemistry, immunophenotype and cytogenetics of acute biphenotypic leukaemias from our institution were studied. The diagnostic criteria took into consideration the morphology of the analysed cells, light scatter characteristics, and evaluation of antibody fluorescence histograms in determining whether the aberrant marker expression was arising from leukaemic blasts or differentiated bone marrow elements. Fifty-two of 746 cases (7%) fulfilled our criteria for acute biphenotypic leukaemias. These included 30 cases of acute lymphoblastic leukaemia (ALL) expressing myeloid antigens, 21 cases of acute myelogenous leukaemia (AML) expressing lymphoid markers, and one case of ALL expressing both B- and T-cell associated antigens. The acute biphenotypic leukaemia cases consisted of four major immunophenotypic subgroups: CD2± AML (11), CD19± AML (8), CD13 and/or CD33± ALL (24), CD11b± ALL (5) and others (4). Chromosomal analysis was carried out in 42/52 of the acute biphenotypic leukaemia cases; a clonal abnormality was found in 31 of these 42 cases. This study highlights the problems encountered in the diagnosis of acute biphenotypic leukaemia, some of which may be reponsible for the wide variation in the reported incidence of this leukaemia. We suggest that the use of strict, uniform diagnostic criteria may help in establishing a more consistent approach towards diagnosis of this leukaemic entity. B Lymphocyte intestinal homing in inflammatory bowel disease
<p>Abstract</p> <p>Background</p> <p>Inflammatory bowel disease (IBD) is thought to be due to an abnormal interaction between the host immune system and commensal microflora. Within the intestinal immune system, B cells produce physiologically natural antibodies but pathologically atypical anti-neutrophil antibodies (xANCAs) are frequently observed in patients with IBD. The objective is to investigate the localisation of immunoglobulin-producing cells (IPCs) in samples of inflamed intestinal tissue taken from patients with IBD, and their possible relationship with clinical features.</p> <p>Methods</p> <p>The IPCs in small intestinal, colonic and rectal biopsy specimens of patients with IBD were analysed by means of immunofluorescence using polyclonal rabbit anti-human Ig and goat anti-human IgM. The B cell phenotype of the IPC-positive samples was assessed using monoclonal antibodies specific for CD79, CD20, CD23, CD21, CD5, λ and κ chains. Statistical correlations were sought between the histological findings and clinical expression.</p> <p>Results</p> <p>The study involved 96 patients (64 with ulcerative colitis and 32 with Crohn's disease). Two different patterns of B lymphocyte infiltrates were found in the intestinal tissue: one was characterised by a strong to moderate stromal localisation of small IgM<sup>+</sup>/CD79<sup>+</sup>/CD20<sup>-</sup>/CD21<sup>-</sup>/CD23<sup>-</sup>/CD5<sup>± </sup>IPCs (42.7% of cases); in the other (57.3%) no such small IPCs were detected in stromal or epithelial tissues. <it>IPCs </it>were significantly less frequent in the patients with Crohn's disease than in those with ulcerative colitis (p = 0.004).</p> <p>Conclusion</p> <p>Our findings suggest that different immunopathogenetic pathways underlie chronic intestinal inflammation with different clinical expressions. The presence of small B lymphocytes resembling B-1 cells also seemed to be negatively associated with Crohn's disease. It can therefore be inferred that the gut contains an alternative population of B cells that have a regulatory function.</p
Regulatory B Cells and Allergic Diseases
B cells are generally considered to positively regulate immune responses by producing antigen-specific antibodies. B cells are classified into classical CD5- conventional B cells and CD5+ B1 cells. The latter produce multi-specific autoantibodies and are thought to be involved in autoimmune diseases. However, evidence supporting a B cell negative regulatory function has accumulated over the past 30 years. Multiple reports have suggested that absence, or loss, of regulatory B cells exacerbates symptoms of both allergic (including contact hypersensitivity and anaphylaxis) and autoimmune (such as experimental autoimmune encephalomyelitis, chronic colitis, and collagen-induced arthritis) diseases, and in lupus-like models of autoimmunity. Regulatory B cells are characterized by production of the negative regulatory cytokines, IL-10 and TGF-β. IL-10-producing B cells were the first regulatory B cells to be recognized and were termed 'B10' cells. IL-10-producing regulatory B cells are of the CD19+CD5+IgMhiIgDloCD1dhi type. Recently, a TGF-β-producing regulatory B cell subset, Br3, has been shown to be related to immune tolerance in food allergies. Moreover, forkhead box P3 (Foxp3)-expressing B cells have also been identified in humans and may act as regulatory B cells (Bregs). The functional image of regulatory B cells is similar to that of regulatory T cells. Because of the proliferative and apoptotic responses of Br1 and Br3 cells in immune tolerance in non-IgE-mediated food allergy, reciprocal roles and counter-regulatory mechanisms of Br1 and Br3 responses are also suspected. Additionally, different roles for regulatory B and T cells at different time points during initiation and progression of autoimmune disease are described
