A discontinuous finite element baroclinic marine model on unstructured prismatic meshes: I. Space discretization
We describe the space discretization of a three-dimensional baroclinic finite element model, based upon a discontinuous Galerkin method, while the companion paper (Comblen et al. 2010a) describes the discretization in time. We solve the hydrostatic Boussinesq equations governing marine flows on a mesh made up of triangles extruded from the surface toward the seabed to obtain prismatic three-dimensional elements. Diffusion is implemented using the symmetric interior penalty method. The tracer equation is consistent with the continuity equation. A Lax–Friedrichs flux is used to take into account internal wave propagation. By way of illustration, a flow exhibiting internal waves in the lee of an isolated seamount on the sphere is simulated. This enables us to show the advantages of using an unstructured mesh, where the resolution is higher in areas where the flow varies rapidly in space, the mesh being coarser far from the region of interest. The solution exhibits the expected wave structure. Linear and quadratic shape functions are used, and the extension to higher-order discretization is straightforward

Piecewise semi-ellipsoidal control invariant sets
Computing control invariant sets is paramount in many applications. The
families of sets commonly used for computations are ellipsoids and polyhedra.
However, searching for a control invariant set over the family of ellipsoids is
conservative for systems more complex than unconstrained linear time invariant
systems. Moreover, even if the control invariant set may be approximated
arbitrarily closely by polyhedra, the complexity of the polyhedra may grow
rapidly in certain directions. An attractive generalization of these two
families are piecewise semi-ellipsoids. We provide in this paper a convex

Heterogeneity assessment of functional T cell avidity.
The potency of cellular immune responses strongly depends on T cell avidity to antigen. Yet, functional avidity measurements are rarely performed in patients, mainly due to the technical challenges of characterizing heterogeneous T cells. The mean functional T cell avidity can be determined by the IFN-γ Elispot assay, with titrated amounts of peptide. Using this assay, we developed a method revealing the heterogeneity of functional avidity, represented by the steepness/hillslope of the peptide titration curve, documented by proof of principle experiments and mathematical modeling. Our data show that not only natural polyclonal CD8 T cell populations from cancer patients, but also monoclonal T cells differ strongly in their heterogeneity of functional avidity. Interestingly, clones and polyclonal cells displayed comparable ranges of heterogeneity. We conclude that besides the mean functional avidity, it is feasible and useful to determine its heterogeneity (hillslope) for characterizing T cell responses in basic research and patient investigation

Formation of Chromate Conversion Coatings on Aluminum and Its Alloys: An in Situ Xanes Study.
We used in situ X-ray adsorption near-edge structure (XANES) to investigate the formation of chromate conversion coatings on pure Al, commercial Al alloys (AA 1100, AA2024, and AA7075), and a series of binary Al-Cu alloys. The method employed a new electrochemical cell that can determine the ratio of hexavalent chromium (Cr(VI)) to total chromium (Cr(total)) speciation in conversion coatings as a function of exposure time to a chromate solution. The spectra showed that the initial Cr(VI)/Cr(total) ratios are greater than later ones for pure Al and AA1100, but not for AA2024 and AA7075. Measurements with Al-Cu alloys demonstrated that the difference observed in AA2024 and AA7075 may not be due to Cu alloying. The proportion of Cr(VI) in the coatings becomes approximately constant after 180 s of exposure for all the specimens examined even though the coatings continue to grow

Imaging with a twist: three-dimensional insights of the chiral nematic phase of cellulose nanocrystals via SHG microscopy
Cellulose nanocrystals (CNCs) are bio-based nanoparticles that, under the right conditions, self-align into chiral nematic liquid crystals with a helical pitch. In this work, we exploit the inherent confocal effect of second-harmonic generation (SHG) microscopy to acquire highly resolved three-dimensional (3D) images of the chiral nematic phase of CNCs in a label-free manner. An in-depth analysis revealed a direct link between the observed variations in SHG intensity and the pitch. The highly contrasted 3D images provided unprecedented detail into liquid crystal’s native structure. Local alignment, morphology, as well as the presence of defects are readily revealed, and a provisional framework relating the SHG response to the orientational distribution of CNC nanorods within the liquid crystal structure is presented. This paper illustrates the numerous benefits of using SHG microscopy for visualizing CNC chiral nematic systems directly in the suspension-liquid phase and paves the road for using SHG microscopy to characterize other types of aligned CNC structures, in wet and dry states.
