    Fatty acid 16:4(n-3) stimulates a GPR120-induced signaling cascade in splenic macrophages to promote chemotherapy resistance

    Although chemotherapy is designed to eradicate tumor cells, it also has significant effects on normal tissues. The platinum-induced fatty acid 16:4(n-3) (hexadeca-4,7,10,13-tetraenoic acid) induces systemic resistance to a broad range of DNA-damaging chemotherapeutics. We show that 16:4(n-3) exerts its effect by activating splenic F4/80+/CD11blow macrophages, which results in production of chemoprotective lysophosphatidylcholines (LPCs). Pharmacologic studies, together with analysis of expression patterns, identified GPR120 on F4/80+/CD11blow macrophages as the relevant receptor for 16:4(n-3). Studies that used splenocytes from GPR120-deficient mice have confirmed this conclusion. Activation of the 16:4(n-3)-GPR120 axis led to enhanced cPLA2 activity in these splenic macrophages and secretion of the resistance-inducing lipid mediator, lysophosphatidylcholine(24:1). These studies identify a novel and unexpected function for GPR120 and suggest that antagonists of this receptor might be effective agents to limit development of chemotherapy resistance.—Houthuijzen, J.     Effect of single intralesional treatment of surgically induced equine superficial digital flexor tendon core lesions with adipose-derived mesenchymal stromal cells: a controlled experimental trial

    Background: Adipose tissue is a promising source of mesenchymal stromal cells (MSCs) for the treatment of tendon disease. The goal of this study was to assess the effect of a single intralesional implantation of adipose tissue-derived mesenchymal stromal cells (AT-MSCs) on artificial lesions in equine superficial digital flexor tendons (SDFTs). Methods: During this randomized, controlled, blinded experimental study, either autologous cultured AT-MSCs suspended in autologous inactivated serum (AT-MSC-serum) or autologous inactivated serum (serum) were injected intralesionally 2 weeks after surgical creation of centrally located SDFT lesions in both forelimbs of nine horses. Healing was assessed clinically and with ultrasound (standard B-mode and ultrasound tissue characterization) at regular intervals over 24 weeks. After euthanasia of the horses the SDFTs were examined histologically, biochemically and by means of biomechanical testing. Results: AT-MSC implantation did not substantially influence clinical and ultrasonographic parameters. Histology, biochemical and biomechanical characteristics of the repair tissue did not differ significantly between treatment modalities after 24 weeks. Compared with macroscopically normal tendon tissue, the content of the mature collagen crosslink hydroxylysylpyridinoline did not differ after AT-MSC-serum treatment (p = 0.074) while it was significantly lower (p = 0.027) in lesions treated with serum alone. Stress at failure (p = 0.048) and the modulus of elasticity (p = 0.001) were significantly lower after AT-MSC-serum treatment than in normal tendon tissue. Conclusions: The effect of a single intralesional injection of cultured AT-MSCs suspended in autologous inactivated serum was not superior to treatment of surgically created SDFT lesions with autologous inactivated serum alone in a surgical model of tendinopathy over an observation period of 22 weeks. AT-MSC treatment might have a positive influence on collagen crosslinking of remodelling scar tissue. Controlled long-term studies including naturally occurring tendinopathies are necessary to verify the effects of AT-MSCs on tendon disease

    Recovery of extracellular vesicles from human breast milk is influenced by sample collection and vesicle isolation procedures

    Extracellular vesicles (EV) in breast milk carry immune relevant proteins and could play an important role in the instruction of the neonatal immune system. To further analyze these EV and to elucidate their function it is important that native populations of EV can be recovered from (stored) breast milk samples in a reproducible fashion. However, the impact of isolation and storage procedures on recovery of breast milk EV has remained underexposed. Here, we aimed to define parameters important for EV recovery from fresh and stored breast milk. To compare various protocols across different donors, breast milk was spiked with a well-defined murine EV population. We found that centrifugation of EV down into density gradients largely improved density-based separation and isolation of EV, compared to floatation up into gradients after high-force pelleting of EV. Using cryo-electron microscopy, we identified different subpopulations of human breast milk EV and a not previously described population of lipid tubules. Additionally, the impact of cold storage on breast milk EV was investigated. We determined that storing unprocessed breast milk at −80°C or 4°C caused death of cells present in breast milk, leading to contamination of the breast milk EV population with storage-induced EV. Here, an alternative method is proposed to store breast milk samples for EV analysis at later time points. The proposed adaptations to the breast milk storage and EV isolation procedures can be applied for EV-based biomarker profiling of breast milk and functional analysis of the role of breast milk EV in the development of the neonatal immune system

    Käesoleva töö eesmärgiks on tõestada või ümber lükata, kas madal sissetulek mõjutab parempoolsete ja konservatiivsete erakondade valimise eelistamist riikide parlamendivalimistel. Töös analüüsitakse võimalikku seost madala sissetuleku ja parempoolsuse vahel Ida- ja Lääne-Euroopa valimi riikide näitel. Töö esimeses osas defineeritakse konservatismi kontseptsioon antud töös, kirjeldatakse, kuidas töös konservatismi mõõdetakse ning võrreldakse teiste võimalike variantidega. Samuti kirjeldatakse madala sissetuleku mõõtmise meetodit antud töös ning seletatakse, milline võib madala sissetuleku mõju valimiskäitumisele olla. Töö teises osas kirjeldatakse uurimustöö meetodit ning kirjeldatakse valimit, tuues välja põhjused, miks antud valim sai uurimustöö hüpoteesi jaoks valitud. Töö kolmas osa on kvalitatiivne analüüs. Analüüs koosneb valimi riikide ning eelnevalt mainitud muutujate vahelisest suhtest. Analüüsiosas on avatud iga valimi riiki eraldi ning kirjeldatud muutujate muutust 21. sajandi lõikes. Töö tulemusena selgub, et madal sissetulek üksiku muutujana ei mõjuta antud valimi põhjal parempoolsete erakondade eelistamist.     On the pathology of focal segmental glomerulosclerosis

    Focal segmental glomerulosclerosis (FSGS) is a heterogeneous renal histopathological entity that affects the glomerulus. It is characterized by podocyte injury, proteinuria and scarring of the glomerular tuft. In this thesis we investigated various mechanisms that are implicated in the development of FSGS. Firstly, we studied biopsy samples of patients with minimal change disease, a related glomerulopathy with minimal glomerular injury and better prognosis. We showed that loss of nephrin could serve as a biomarker for renal function loss and the progression to FSGS in this patient group. We also investigated renal biopsy material of patients with FSGS to determine the role of complement activation and endothelial-podocyte interaction in the pathogenesis of FSGS. We show that complement activation via the classical pathway and endothelial injury, especially via endothelin-1 signaling, are associated with the development of FSGS in humans. This thesis also describes the study of the genomic architecture of the Munich Wistar Frömter rat model for FSGS, which led to the prioritization of TMEM63c and PTGR2 in the development of podocyte injury and proteinuria in this model. The identification of these mechanisms in the development of FSGS, increases our understanding of the pathophysiology of FSGS and can guide future studies into new, highly needed therapeutic strategies.Nierstichting Nederland; Janivo StichtingLUMC / Geneeskund
