Transcriptome Analysis and Ultrastructure Observation Reveal that Hawthorn Fruit Softening Is due to Cellulose/Hemicellulose Degradation
Softening, a common phenomenon in many fruits, is a well coordinated and genetically determined process. However, the process of flesh softening during ripening has rarely been described in hawthorn. In this study, we found that ‘Ruanrou Shanlihong 3 Hao’ fruits became softer during ripening, whereas ‘Qiu JinXing’ fruits remained hard. At late developmental stages, the firmness of ‘Ruanrou Shanlihong 3 Hao’ fruits rapidly declined, and that of ‘Qiu JinXing’ fruits remained essentially unchanged. According to transmission electron microscopy (TEM), the middle lamella of ‘Qiu JinXing’ and ‘Ruanrou Shanlihong 3 Hao’ fruit flesh was largely degraded as the fruits matured. Microfilaments in ‘Qiu JinXing’ flesh were arranged close together and were deep in color, whereas those in ‘Ruanrou Shanlihong 3 Hao’ fruit flesh were arranged loosely, partially degraded and light in color. RNA-Seq analysis yielded approximately 46.72 Gb of clean data and 72,837 unigenes. Galactose metabolism and pentose and glucuronate interconversions are involved in cell wall metabolism, play an important role in hawthorn texture. We identified 85 unigenes related to the cell wall between hard- and soft-fleshed hawthorn fruits. Based on data analysis and real-time PCR, we suggest that β-GAL and PE4 have important functions in early fruit softening. The genes Ffase, Gns, α-GAL, PE63, XTH and CWP, which are involved in cell wall degradation, are responsible for the different textures of hawthorn fruits. Thus, we hypothesize that the different textures of ‘Qiu JinXing’ and ‘Ruanrou Shanlihong 3 Hao’ fruits at maturity mainly result from cellulose/hemicelluloses degradation rather than from lamella degradation. Overall, we propose that different types of hydrolytic enzymes in cells interact to degrade the cell wall, resulting in ultramicroscopic Structure changes in the cell wall and, consequently, fruit softening. These results provide fundamental insight regarding the mechanisms by which hawthorn fruits acquire different textures and also lay a solid foundation for further research
Lactobacillus pentosus expressing porcine lactoferrin elevates antibacterial activity and improves the efficacy of vaccination against Aujeszky’s disease
In this study, Lactobacillus pentosus expressing porcine lactoferrin (pLF) was tested for in vitro antibacterial activity and for its ability to enhance immunity induced by an orally administered Aujeszky’s disease virus (ADV) vaccine. The cDNA encoding N-terminus of pLF was cloned into a Lactobacillus-specific plasmid to produce L. pentosus pLF expressing transformants (pPG612.1-pLFN/ L. pentosus). The antimicrobial activity of the recombinant pLF protein inhibited bacterial growth in vitro. The supernatant of pPG612.1-pLF-N/L. pentosus had an inhibitory effect on Staphylococcus aureus strain CVCC26003, Bacillus subtilis strain CVCC63501, Escherichia coli strain CVCC10141 and Salmonella enterica ssp. enterica Choleraesuis strain CVCC79102, while it did not inhibit the growth of Lactobacillus casei strain ATCC393. A mouse model was established to test the effectiveness of the orally administered probiotic L. pentosus recombinant strain in the gastrointestinal tract. Mice were immunised with an attenuated porcine Aujeszky’s disease virus (ADV) vaccine. Serum antibody levels determined using a mouse Aujeszky’s disease IgG ELISA showed that IgG levels were significantly higher in the pPG612.1-pLFN/L. pentosus group than in the PBS and Lactobacillus pentosus groups at days 7 and 21 (P < 0.01) and at day 14 (P < 0.05), indicating that this oral recombinant strain can improve the effectiveness of the vaccine and play a role in immune enhancement through humoral immunity. These results suggest that the recombinant Lactobacillus pentosus not only has the beneficial characteristics of lactic acid bacteria but also produces biologically functional lactoferrin

Measuring Star-formation Rate and Far-Infrared Color in High-redshift Galaxies Using the CO (7-6) and [NII] 205 micron Lines
To better characterize the global star formation (SF) activity in a galaxy,
one needs to know not only the star formation rate (SFR) but also the
rest-frame, far-infrared (FIR) color (e.g., the 60-to-100 m color,
] of the dust emission. The latter probes the average intensity of
the dust heating radiation field and scales statistically with the effective
SFR surface density in star-forming galaxies including (ultra-)luminous
infrared galaxies [(U)LIRGs]. To this end, we exploit here a new spectroscopic
approach involving only two emission lines: CO\,(76) at 372 m and [NII]
at 205 m. For local (U)LIRGs, the ratios of the CO (76) luminosity
() to the total infrared luminosity (; 81000
m) are fairly tightly distributed (to within 0.12 dex) and show
little dependence on . This makes a good SFR
tracer, which is less contaminated by active galactic nuclei (AGN) than and may also be much less sensitive to metallicity than . Furthermore, the logarithmic [NII] 205 m to CO (76)
luminosity ratio is fairly steeply (at a slope of ) correlated with
, with a modest scatter (0.23 dex). This makes it a useful
estimator on with an implied uncertainty of 0.15 [or
4 K in the dust temperature () in the case of a
graybody emission with K and a dust emissivity index
]. Our locally calibrated SFR and estimators are shown

Evaluating the performance of the government venture capital guiding fund using the intuitionistic fuzzy analytic hierarchy process
The performance evaluation of the government venture capital guiding fund (GVCGF) has come into focus in the field of venture capital. Most of the existing studies, such as whether the GVCGF has guided social capital to start-up enterprises and has played its due role in the process of enterprise growth and innovation, are all based on relevant work under the framework of econometric analysis. Unlike in these existing studies, we construct the performance analysis model of the GVCGF from four dimensions, including the standardization development of the guidance fund, the risk control ability, and the leverage and the support effects under the framework of a multi-attribute decision-making analysis. Taking a GVCGF project in Ningbo City, China, as an example, we comprehensively evaluate the development performance of the GVCGF using the intuitionistic fuzzy analytic hierarchy process (IFAHP). The results show that the development performance of the GVCGF is at a “relatively high” level. Compared with the traditional analytic hierarchy process (AHP), the IFAHP effectively avoids the false, enlarged influence caused by data subjectivity and evaluation uncertainty. This study provides a feasible analytical framework for the application of the IFAHP in other project performance evaluations
Asymptotic analysis on charging dynamics for stack-electrode model of supercapacitors
Supercapacitors are promising electrochemical energy storage devices due to
their prominent performance in rapid charging/discharging rates, long cycle
life, stability, etc. Experimental measurement and theoretical prediction on
charging timescale for supercapacitors often have large difference. This work
develops a matched asymptotic expansion method to derive the charging dynamics
of supercapacitors with porous electrodes, in which the supercapacitors are
described by the stack-electrode model. Coupling leading-order solutions
between every two stacks by continuity of ionic concentration and fluxes leads
to an ODE system, which is a generalized equivalent circuit model for zeta
potentials, with the potential-dependent nonlinear capacitance and resistance
determined by physical parameters of electrolytes, e.g., specific counterion
valences for asymmetric electrolytes. Linearized stability analysis on the ODE
system after projection is developed to theoretically characterize the charging
timescale. The derived asymptotic solutions are numerically verified. Further
numerical investigations on the biexponential charging timescales demonstrate
that the proposed generalized equivalent circuit model, as well as companion
linearized stability analysis, can faithfully capture the charging dynamics of
symmetric/asymmetric electrolytes in supercapacitors with porous electrodes.Comment: 26 pages, 6 figure
