Criteria are established for comparing alternate concepts for: (1) deployment of 220 klb space shuttle from the space station; (2) tether assisted launch of a 20,000 lb payload to geosynchronous orbit; (3) placement of the 20,000 lb AXAF into 320 nmi orbit via orbiter; (4) retrieval of 20,000 lb AXAF from 205 nmi circular orbit for maintenance and reboost to 320 nmi; and (5) tethered OMV rendezvous and retrieval of OTV returning from a geosynchronous mission.     The application of tether technology has the potential to increase the overall performance efficiency and capability of the integrated space operations and transportation systems through the decade of the 90s. The primary concepts for which significant economic benefits were identified are dependent on the space station as a storage device for angular momentum and as an operating base for the tether system. Concepts examined include: (1) tether deorbit of shuttle from space station; (2) tethered orbit insertion of a spacecraft from shuttle; (3) tethered platform deployed from space station; (4) tether-effected rendezvous of an OMV with a returning OTV; (5) electrodynamic tether as an auxiliary power source for space station; and (6) tether assisted launch of an OTV mission from space station

    Violence Outside to Violence Within: The Experience of Sexual Minorities in Schools and Intimate Relationships

    The current study explored the association between sexual minorities’ experiences in schools and relationships. Socio-political-psychological theory provided a framework for the exploration of how retrospective reports of sexual orientation violence in school (SOVS) and school environment predicted the experience and perpetration of sexual minority intimate partner violence (SMIPV). Because of its relation to both school and interpersonal violence, alcohol was also hypothesized to predict rates of experiencing and perpetrating SMIPV. Group differences for all scales were explored on the basis of sexual orientation, gender, race/ethnicity, and education. Chi-square and analysis of variance analyses revealed several significant differences. Logistic regressions revealed that the experience of SOVS was not found to significantly affect the risk of experiencing or perpetrating SMIPV. However, a negative school environment was found to affect the risk of experiencing and perpetrating SMIPV differentially by gender and race, respectively. Results also revealed that alcohol significantly predicted the perpetration of SMIPV

    Lean on me: Informal social networks and the prevention of intimate partner violence in sexual minority communities

    Research finds that intimate partner violence (IPV) occurs at comparable rates for heterosexuals and sexual minorities; however, few IPV prevention programs exist for sexual minority communities. Most programs are developed on heterosexuals and ignore the unique contexts and dynamics of IPV for sexual minorities. Community capacity IPV prevention programs aim to increase the skills and resources within informal social networks, and they represent a promising approach to IPV prevention for sexual minority communities. The current study explores the informal networks of sexual minorities in order to build knowledge that can inform the future development of community capacity IPV prevention programs for sexual minorities. The goal of the current study was to provide information on three major aspects of sexual minorities’ informal networks: network structure, network function, and the use of networks by sexual minorities experiencing IPV. The study used a mixed method design. The quantitative component included an online survey completed by 367 sexual minorities. The survey asked with whom sexual minorities discuss their intimate relationships, and it asked the response and helpfulness of each member. These data illustrated the structure and function of informal networks. The study also included interviews with seven sexual minority women on their experiences of seeking help for IPV from their social networks. This information addressed the third aspect of informal networks. The quantitative results revealed that sexual minorities turn to on average only three people to discuss relationship issues. Surprisingly, a substantial number were family, and almost half were heterosexual. The qualitative results illustrated that many informal networks members could benefit from receiving education on sexual minority identities and issues, IPV in sexual minority communities, and communication skills. The findings illustrated key aspects of informal networks that can be used to inform future community capacity IPV prevention programs for sexual minorities. Specifically, the quantitative data on network structure and function can be used to inform relevant targets for future programs, and the data from the interviews can inform aspects of program curricula

    Engineering HIV envelope protein to activate germline B cell receptors of broadly neutralizing anti-CD4 binding site antibodies

    Broadly neutralizing antibodies (bnAbs) against HIV are believed to be a critical component of the protective responses elicited by an effective HIV vaccine. Neutralizing antibodies against the evolutionarily conserved CD4-binding site (CD4-BS) on the HIV envelope glycoprotein (Env) are capable of inhibiting infection of diverse HIV strains, and have been isolated from HIV-infected individuals. Despite the presence of anti–CD4-BS broadly neutralizing antibody (bnAb) epitopes on recombinant Env, Env immunization has so far failed to elicit such antibodies. Here, we show that Env immunogens fail to engage the germline-reverted forms of known bnAbs that target the CD4-BS. However, we found that the elimination of a conserved glycosylation site located in Loop D and two glycosylation sites located in variable region 5 of Env allows Env-binding to, and activation of, B cells expressing the germline-reverted BCRs of two potent broadly neutralizing antibodies, VRC01 and NIH45-46. Our results offer a possible explanation as to why Env immunogens have been ineffective in stimulating the production of such bNAbs. Importantly, they provide key information as to how such immunogens can be engineered to initiate the process of antibody-affinity maturation against one of the most conserved Env regions

    Computational analysis of anti-HIV-1 antibody neutralization panel data to identify potential functional epitope residues

    Advances in single-cell antibody cloning methods have led to the identification of a variety of broadly neutralizing anti–HIV-1 antibodies. We developed a computational tool (Antibody Database) to help identify critical residues on the HIV-1 envelope protein whose natural variation affects antibody activity. Our simplifying assumption was that, for a given antibody, a significant portion of the dispersion of neutralization activity across a panel of HIV-1 strains is due to the amino acid identity or glycosylation state at a small number of specific sites, each acting independently. A model of an antibody’s neutralization IC_(50) was developed in which each site contributes a term to the logarithm of the modeled IC_(50). The analysis program attempts to determine the set of rules that minimizes the sum of the residuals between observed and modeled IC_(50) values. The predictive quality of the identified rules may be assessed in part by whether there is support for rules within individual viral clades. As a test case, we analyzed antibody 8ANC195, an anti-glycoprotein gp120 antibody of unknown specificity. The model for this antibody indicated that several glycosylation sites were critical for neutralization. We evaluated this prediction by measuring neutralization potencies of 8ANC195 against HIV-1 in vitro and in an antibody therapy experiment in humanized mice. These experiments confirmed that 8ANC195 represents a distinct class of glycan-dependent anti–HIV-1 antibody and validated the utility of computational analysis of neutralization panel data

Preliminary designs are included of pilot plant utilizing large numbers of heliostats in a collector field.     Hazard Communication: A Review of the Science Underpinning the Art of Communication for Health and Safety

    This report was commissioned by the United States Occupational Safety and Health Administration to review the state of scientific inquiry supporting our knowledge regarding key elements of chemical hazard communication programs: labeling, warnings, material safety data sheets, and worker training. This endeavor supports the international effort to harmonize laws, regulations, and consensus standards affecting the ways in which information about hazardous chemicals is communicated. The international effort can be divided into three major functions: classifying health and environmental hazards, classifying physical hazards; and communicating hazard information. This last component involves the determination of what information will be communicated to users regarding the hazards and appropriate protective measures, as well as the way in which it will be transmitted, i.e. through symbols, labels, standard phrases, and training
