    Magnetic Flux Braiding: Force-Free Equilibria and Current Sheets

    We use a numerical nonlinear multigrid magnetic relaxation technique to investigate the generation of current sheets in three-dimensional magnetic flux braiding experiments. We are able to catalogue the relaxed nonlinear force-free equilibria resulting from the application of deformations to an initially undisturbed region of plasma containing a uniform, vertical magnetic field. The deformations are manifested by imposing motions on the bounding planes to which the magnetic field is anchored. Once imposed the new distribution of magnetic footpoints are then taken to be fixed, so that the rest of the plasma must then relax to a new equilibrium configuration. For the class of footpoint motions we have examined, we find that singular and nonsingular equilibria can be generated. By singular we mean that within the limits imposed by numerical resolution we find that there is no convergence to a well-defined equilibrium as the number of grid points in the numerical domain is increased. These singular equilibria contain current "sheets" of ever-increasing current intensity and decreasing width; they occur when the footpoint motions exceed a certain threshold, and must include both twist and shear to be effective. On the basis of these results we contend that flux braiding will indeed result in significant current generation.     Improving the engine power of a catalytic Janus-sphere micromotor by roughening its surface

    Microspheres with catalytic caps have become a popular model system for studying self-propelled colloids. Existing experimental studies involve predominantly "smooth" particle surfaces. In this study we determine the effect of irregular surface deformations on the propulsive mechanism with a particular focus on speed. The particle surfaces of polymer microspheres were deformed prior to depositing a layer of platinum which resulted in the formation of nanoscopic pillars of catalyst. Self-propulsion was induced upon exposure of the micromotors to hydrogen peroxide, whilst they were dispersed in water. The topological surface features were shown to boost speed (~2×) when the underlying deformations are small (nanoscale), whilst large deformations afforded little difference despite a substantial apparent catalytic surface area. Colloids with deformed surfaces were more likely to display a mixture of rotational and translational propulsion than their "smooth" counterparts

    poster abstractThe variance of phase angle changes over the network is a good display of total stress and angle stability. The integral square generator angle (ISGA) changes had been recommended earlier to evaluate how severe the stable and unstable transient contingencies in simulation are. This project offers its addition to bus voltage angles (ISBA) which could be measured with synchronized phasor measurement units (PMUs) over a wide-ranging area. By restructuring continuous paths that go outside the boundary be-tween positive and negative 180 degrees before calculating the ISBA, the cutoff of bus angles at positive and negative 180 degrees is recovered. The project also directs the matter of obtaining the best angle stability index as the threshold between stable and unstable classes with use of simulation da-ta. This issue becomes more difficult by the fact that large databases might include a few events for which loss of synchronism happens toward the end of the simulation sequence

    The Cytochrome P450 gene CYP6P12 confers pyrethroid resistance in kdr-free Malaysian populations of the dengue vector Aedes albopictus

    Control of Aedes albopictus, major dengue and chikungunya vector, is threatened by growing cases of insecticide resistance. The mechanisms driving this resistance remain poorly characterised. This study investigated the molecular basis of insecticide resistance in Malaysian populations of Ae. albopictus. Microarray-based transcription profiling revealed that metabolic resistance (cytochrome P450 up-regulation) and possibly a reduced penetration mechanism (consistent over-expression of cuticular protein genes) were associated with pyrethroid resistance. CYP6P12 over-expression was strongly associated with pyrethroid resistance whereas CYP6N3 was rather consistently over-expressed across carbamate and DDT resistant populations. Other detoxification genes also up-regulated in permethrin resistant mosquitoes included a glucuronosyltransferase (AAEL014279-RA) and the glutathione-S transferases GSTS1 and GSTT3. Functional analyses further supported that CYP6P12 contributes to pyrethroid resistance in Ae. albopictus as transgenic expression of CYP6P12 in Drosophila was sufficient to confer pyrethroid resistance in these flies. Furthermore, molecular docking simulations predicted CYP6P12 possessing enzymatic activity towards pyrethroids. Patterns of polymorphism suggested early sign of selection acting on CYP6P12 but not on CYP6N3. The major role played by P450 in the absence of kdr mutations suggests that addition of the synergist PBO to pyrethroids could improve the efficacy of this insecticide class and overcome resistance in field populations of Ae. albopictus

    Confident, fearful and hopeless cooks: Findings from the development of a food-skills initiative

    One of the many barriers to a healthier diet in low-income communities is a presumed lack of practical food skills. This article reports findings from exploratory qualitative research conducted with potential participants in a cooking skills intervention, in low income communities in Scotland. The research found widely varying levels of skill and confidence regarding cooking, supported the need for a community-based intervention approach, and demonstrated the importance of consumer research to inform the content of interventions. Challenges the view that low income communities lack skills, suggesting that food skills should be defined more broadly than “cooking from scratch”. Other barriers to healthy eating, such as poverty, food access and taste preferences, remain important

    Herd-level prevalence of selected endemic infectious diseases of dairy cows in Great Britain

    To implement appropriate and effective disease control programs at the national level, up-to-date and unbiased information on disease frequency is needed. The aim of this study was to estimate the prevalence of selected endemic infectious diseases in the population of dairy herds in Great Britain. Bulk milk tank (BMT) samples from 225 randomly selected dairy farms, stratified by region and herd size, were tested for antibodies against bovine viral diarrhea virus (BVDV), bovine herpesvirus type 1, Mycobacterium avium ssp. paratuberculosis, Leptospira Hardjo, Salmonella spp., Coxiella burnetii, Fasciola hepatica, Neospora caninum, and Ostertagia ostertagi. Furthermore, the presence of BVDV, C. burnetii, and Chlamydia-like organisms was determined by PCR. The apparent herd prevalence was estimated as a weighted proportion of positive herds. The true prevalence was calculated when a test was used with known test characteristics for the cut-off value used. Among unvaccinated herds, the true prevalence of BMT antibodies against BVDV was estimated at 66% [95% confidence interval (CI): 56–77%], M. avium ssp. paratuberculosis 68% (95% CI: 59–77%), bovine herpesvirus type 1 62% (95% CI: 52–73%), Leptospira Hardjo 47% (95% CI: 34–60%), and Salmonella spp. 48% (95% CI: 39–56%). The apparent prevalence of BMT antibodies against C. burnetii was 80% (95% CI: 75–85%), F. hepatica 55% (95% CI: 48–62%), N. caninum 46% (95% CI: 38–54%), and O. ostertagi 95% (95% CI: 91–98%). The BVDV, C. burnetii, and Chlamydia-like antigens were detected in 5 (95% CI: 2–9%), 29 (95% CI: 21–36%), and 31% (95% CI: 24–38%) of herds, respectively. Our results show that dairy cows across GB are frequently exposed to the studied pathogens, which are endemic at high levels with some geographical variations. These prevalence estimates provide a much-needed basis to assess whether nationwide control programs for the studied pathogens are justified by their potential economic, environmental, and public health implications. Should surveillance and control programs be initiated, the estimates presented here are a baseline against which progress can be assessed

    Improving the molecular diagnosis of Chlamydia psittaci and Chlamydia abortus infection with a species-specific duplex real-time PCR.

    Chlamydia psittaci and Chlamydia abortus are closely related intracellular bacteria exhibiting different tissue tropism that may cause severe but distinct infection in humans. C. psittaci causes psittacosis, a respiratory zoonotic infection transmitted by birds. C. abortus is an abortigenic agent in small ruminants, which can also colonize the human placenta and lead to foetal death and miscarriage. Infections caused by C. psittaci and C. abortus are underestimated mainly due to diagnosis difficulties resulting from their strict intracellular growth. We developed a duplex real-time PCR to detect and distinguish these two bacteria in clinical samples. The first PCR (PCR1) targeted a sequence of the 16S-23S rRNA operon allowing the detection of both C. psittaci and C. abortus. The second PCR (PCR2) targeted the coding DNA sequence CPSIT_0607 unique to C. psittaci. The two PCRs showed 100 % detection for ≥ 10 DNA copies per reaction (1000 copies ml- 1). Using a set of 120 samples, including bacterial reference strains, clinical specimens and infected cell culture material, we monitored 100 % sensitivity and 100 % specificity for the detection of C. psittaci and C. abortus for PCR1. When PCR1 was positive, PCR2 could discriminate C. psittaci from C. abortus with a positive predictive value of 100 % and a negative predictive value of 88 %. In conclusion, this new duplex PCR represents a low-cost and time-saving method with high-throughput potential, expected to improve the routine diagnosis of psittacosis and pregnancy complication in large-scale screening programs and also during outbreaks

    Prevalence of Chlamydia abortus in Belgian ruminants

    Chlamydia (C.) abortus enzootic abortion still remains the most common cause of reproductive failure in sheep-breeding countries all over the world. Chlamydia abortus in cattle is predominantly associated with genital tract disease and mastitis. In this study, Belgian sheep (n=958), goats (n=48) and cattle (n=1849) were examined, using the ID Screen (TM) Chlamydia abortus indirect multi-species antibody ELISA. In the sheep, the highest prevalence rate was found in Limburg (4.05%). The animals of Antwerp, Brabant and Liege tested negative. The prevalence in the remaining five regions was low (0.24% to 2.74%). Of the nine goat herds, only one herd in Luxembourg was seropositive. In cattle, the highest prevalence rate was found in Walloon Brabant (4.23%). The animals of Limburg and Namur tested negative. The prevalence rate in the remaining seven regions ranged between 0.39% and 4.02%
