    Inhibitory Activity of Leaves Extracts of Citrullus colocynthis Schrad. on HT29 Human Colon Cancer Cells

    Aims: Citrullus colocynthis is a plant endemic in Asia, Africa and in the Mediterranean basin. It is used in folk medicine against infections, inflammations and cardiovascular and immune-related diseases. There are further evidences of the use of Citrullus colocynthis Schrad in the treatment of cancer in traditional practices. The present study aimed to determine the potential antiproliferative effects of different Citrullus colocynthis leaf extracts on human cancer cells. Methodology: Antiproliferative and antioxidant effects on HT-29 human colon cancer cells were detected by MTS assay and a modified protocol of the alkaline Comet assay. In vitro antioxidant activities of different leaf extracts were evaluated through DPPH, \u3b2-carotene/linoleic acid and reducing power assays. Results: The leaf chloroform extract exhibited the higher cell growth inhibitory activity without induction of DNA damage; it showed to be able to significantly decrease DNA damage induced by H2O2 (100 M). This antioxidant activity seems to be comparable to that of vitamin C (1 mM). Ethyl acetate, acetone and methanol leaf extracts showed to be the most effective in reducing the stable free DPPH radical (IC50 =113 g/ml), in transforming the Fe3+ to Fe2+ (IC50 = 134 \ub5g/ml) and in inducing linoleic acid oxidation with an inhibition of 31.9 %. Conclusion: Our results confirm the antiproliferative potential of Citrullus colocynthis Schrad. on human cancer cells

    A New Procedure for Combining UAV-Based Imagery and Machine Learning in Precision Agriculture

    Drone images from an experimental field cropped with sugar beet with a high diffusion of weeds taken from different flying altitudes were used to develop and test a machine learning method for vegetation patch identification. Georeferenced images were combined with a hue-based preprocessing analysis, digital transformation by an image embedder, and evaluation by supervised learning. Specifically, six of the most common machine learning algorithms were applied (i.e., logistic regression, k-nearest neighbors, decision tree, random forest, neural network, and support-vector machine). The proposed method was able to precisely recognize crops and weeds throughout a wide cultivation field, training from single partial images. The information has been designed to be easily integrated into autonomous weed management systems with the aim of reducing the use of water, nutrients, and herbicides for precision agriculture

    Sono state esaminate le uova non schiuse di Saperda carcharias L. in piante di pioppo testimone e trattate con metilparathion e phenthoate (allo 0,4% di sostanza attiva) per studiare l'effetto dei due insetticidi sul parassita oofago Euderus caudatus Thom. L'analisi statistica dei dati raccolti dimostra che la mortalità di E. caudatus è significativamente più elevata nelle piante trattate (37-64%) in confronto a quelle testimone ( 0-15 % )

    Prospective randomized comparison of open versus laparoscopic management of splenic artery aneurysms: a 10-year study

    Abstract BACKGROUND: The literature does not support the choice between open and laparoscopic management of splenic artery aneurysms (SAA). METHODS: We designed a prospective, randomized comparison between open and laparoscopic surgery for SAA. Primary end points were types of surgical procedures performed and clinical outcomes. Analysis was developed on an intention-to-treat basis. RESULTS: Fourteen patients were allocated to laparotomy (group A) and 15 to laparoscopy (group B). Groups displayed similar patient- and aneurysm-related characteristics. The conversion rate to open surgery was 13.3 %. The type of surgical procedure performed on the splenic artery was similar in the two groups: aneurysmectomy with splenic artery ligature or direct anastomosis was performed in 51 % and 21 % of patients in group A and in 60 % and 20 % in group B, respectively. The splenectomy rate was similar (14 % vs. 20 %). Postoperative splenic infarction was observed in one case in each group. Laparoscopy was associated with shorter procedures (p = 0.0003) and lower morbidity (25 % vs. 64 %, p = 0.045). Major morbidity requiring interventional procedures and blood transfusion was observed only in group A. Laparoscopy was associated with quicker resumption of oral diet (p < 0.001), earlier drain removal (p = 0.046), and shorter hospital stay (p < 0.01). During a mean follow-up of 50 months, two patients in group A required hospital readmission. In group B, two patients developed a late thrombosis of arterial anastomoses. CONCLUSIONS: Our study demonstrates that laparoscopy permits multiple technical options, does not increase the splenectomy rate, and reduces postoperative complications. It confirms the supposed clinical benefits of laparoscopy when ablative procedures are required but laparoscopic anastomoses show poor long-term results

    Determination of Pesticide Residues in IV Range Artichoke (Cynara cardunculus L.) and Its Industrial Wastes

    Fourth-range products are those types of fresh fruit and vegetables that are ready for raw consumption or after cooking, and belong to organic or integrated cultivations. These products are subject to mild post-harvesting processing procedures (selection, sorting, husking, cutting, and washing), and are afterwards packaged in packets or closed food plates, with an average shelf life of 5–10 days. Artichokes are stripped of the leaves, stems and outer bracts, and the remaining heads are washed with acidifying solutions. The A LC-MS/MS analytical method was developed and validated following SANTE guidelines for the detection of 220 pesticides. This work evaluated the distribution of pesticide residues among the fraction of artichokes obtained during the industrial processing, and the residues of their wastes left on the field were also investigated. The results showed quantifiable residues of one herbicide (pendimethalin) and four fungicides (azoxystrobin, propyzamide, tebuconazole, and pyraclostrobin). Pendimethalin was found in all samples, with the higher values in leaves 0.046 ± 8.2 mg/kg and in field waste 0.30 ± 6.7 mg/kg. Azoxystrobin was the most concentrated in the outer bracts (0.18 ± 2.9 mg/kg). The outer bracts showed the highest number of residues.     Interleukin 2 (IL-2) is considered a key player in exacerbating multiple sclerosis (MS). Therapies targeting its receptor have been developed; however, a resolution of the disease and side effects are still an issue of concern. The involvement of other factors, such as Mycobacterium avium subspecies paratuberculosis (MAP) and envelope protein derived from human endogenous retrovirus type W (HERV-Wenv), in MS pathogenesis has been recently suggested. Here, we investigated the levels of antibodies (Abs) directed against IL-2 and HERV-Wenv in 108 MS patients, 34 patients affected by neuromyelitis optica spectrum disorder (NMOSD), and 137 healthy controls (HCs). Our results show increased levels of Abs specific to IL-2 and HERV-Wenv-su antigens in MS vs. HCs (p &lt; 0.0001 for IL-2, p = 0.0004 for HERV-Wenv) and significantly decreased levels in NMOSD vs. MS. The assessment of different 12-month-long therapies on Abs against IL-2, HERV-Wenv, and MAP lipoarabinomannan (LAM) demonstrated the strongest effect on anti-LAM Abs (p = 0.018), a slight reduction of anti-IL-2 Abs, and small variations for anti-HERV-Wenv Abs. These results highlight the conclusion that the impact of therapy is more correlated with selected epitopes than with the therapeutic agent. Screening for anti-IL-2 and anti-HERV-Wenv Abs has a potential as additional future practice to distinguish between symptomatically similar MS and NMOSD

    The association between human endogenous retroviruses and multiple sclerosis: a systematic review and meta-analysis

    Background: The interaction between genetic and environmental factors is crucial to multiple sclerosis (MS) pathogenesis. Human Endogenous Retroviruses (HERVs) are endogenous viral elements of the human genome whose expression is associated with MS. Objective: To perform a systematic review and meta-analysis and to assess qualitative and quantitative evidence on the expression of HERV families in MS patients. Methods: Medline, Embase and the Cochrane Library were searched for published studies on the association of HERVs and MS. Meta-analysis was performed on the HERV-W family. Odds Ratio (OR) and 95% confidence interval (CI) were calculated for association. Results: 43 reports were extracted (25 related to HERV-W, 13 to HERV-H, 9 to HERV-K, 5 to HRES-1 and 1 to HER-15 family). The analysis showed an association between expression of all HERV families and MS. For HERV-W, adequate data was available for meta-analysis. Results from meta-analyses of HERV-W were OR = 22.66 (95%CI 6.32 to 81.20) from 4 studies investigating MSRV/HERV-W(MS-associated retrovirus) envelope mRNA in peripheral blood mononuclear cells, OR = 44.11 (95%CI 12.95 to 150.30) from 6 studies of MSRV/ HERV-W polymerase mRNA in serum/plasma and OR = 6.00 (95%CI 3.35 to 10.74) from 4 studies of MSRV/HERV-W polymerase mRNA in CSF

    Efficacy of a Native Microbial Starter in Promoting Table Olive Fermentation: An Industrial-Scale Trial at Controlled and Ambient Temperature

    This study evaluated a multi-strain starter culture's impact on the industrial-scale fermentation of "Tonda di Cagliari" table olives, comparing processes at ambient versus controlled (23-25 degrees C) temperatures. Controlled fermentation accelerated acidification, yielding lower pH levels, higher lactic acid bacteria (LAB) counts, and better control over Enterobacteriaceae. Starter inoculation ensured the attainment of safe pH levels (&lt;4.2) even at ambient temperature, while uninoculated samples did not reach safe pH levels under those conditions (&gt;4.5 in non-inoculated samples). Regardless of processing temperature, starter-inoculated olives consistently yielded higher final concentrations of hydroxytyrosol (719.2 and 762.9 mg/kg inoculated, 480.7 and 326 mg/kg non-inoculated). Total phenolic content in olives remained higher throughout the fermentation process at the controlled temperature (3138 and 2112 mg/kg ambient temperature, 3458 and 3622 mg/kg controlled temperature). Olives maintained at controlled (higher) temperatures exhibited lower final moisture content and significantly reduced lipid content. While texture profiles were primarily affected by temperature, sensory acceptability was significantly influenced by both the starter inoculation and the fermentation temperature. These findings indicate that using microbial starters can potentially lower energy costs associated with heating processing rooms, particularly during colder seasons, while still ensuring food safety and enhancing nutraceutical value. Although the faster fermentation rate at controlled temperature did not substantially shorten overall marketing time, the starter eliminates the necessity for heating facilities to achieve a food-safe pH within a reasonable timeframe
