Heat Sensing Receptor TRPV1 Is a Mediator of Thermotaxis in Human Spermatozoa
The molecular bases of sperm thermotaxis, the temperature-oriented cell motility, are currently under investigation. Thermal perception relies on a subclass of the transient receptor potential [TRP] channels, whose member TRPV1 is acknowledged as the heat sensing receptor. Here we investigated the involvement of TRPV1 in human sperm thermotaxis. We obtained semen samples from 16 normozoospermic subjects attending an infertility survey programme, testis biopsies from 6 patients with testicular germ cell cancer and testis fine needle aspirates from 6 patients with obstructive azoospermia undergoing assisted reproductive technologies. Expression of TRPV1 mRNA was assessed by RT-PCR. Protein expression of TRPV1 was determined by western blot, flow cytometry and immunofluorescence. Sperm motility was assessed by Sperm Class Analyser. Acrosome reaction, apoptosis and intracellular-Ca2+ content were assessed by flow cytometry. We found that TRPV1 mRNA and protein were highly expressed in the testis, in both Sertoli cells and germ-line cells. Moreover, compared to no-gradient controls at 31°C or 37°C (Ctrl 31°C and Ctrl 37°C respectively), sperm migration towards a temperature gradient of 31-37°C (T gradient) in non-capacitated conditions selected a higher number of cells (14,9 ± 4,2×106 cells T gradient vs 5,1± 0,3×106 cells Ctrl 31°C and 5,71±0,74×106 cells Ctrl 37°C; P = 0,039). Capacitation amplified the migrating capability towards the T gradient. Sperms migrated towards the T gradient showed enriched levels of both TRPV1 protein and mRNA. In addition, sperm cells were able to migrate toward a gradient of capsaicin, a specific agonist of TRPV1, whilst capsazepine, a specific agonist of TRPV1, blocked this effect. Finally, capsazepine severely blunted migration towards T gradient without abolishing. These results suggest that TRPV1 may represent a facilitating mediator of sperm thermotaxis
Size/Age Models for Monitoring of the Pink Sea Fan Eunicella verrucosa (Cnidaria: Alcyonacea) and a Case Study Application
The pink sea fan Eunicella verrucosa is a habitat-forming octocoral living in the East Atlantic and in the Mediterranean Sea where, under proper circumstances, it can form large populations known as coral forests. Although these coral forests represent vulnerable marine ecosystems of great importance, these habitats are still poorly known, and their monitoring is almost non-existent to date. For this reason, we compared two dierent models to infer the age of E. verrucosa based on nondestructive measurements of the colonies’ size, in order to highlight strengths and weaknesses of the existing tools for a potential application in long-term monitoring. We also applied the two models on a case-study population recently found in the northwest Mediterranean Sea. Our results showed which model was more reliable from a biological point of view, considering both its structure and the results obtained on the case study. However, this model uses solely the height of the colonies as proxy to infer the age, while the total branch fan surface area could represent a more appropriate biometric parameter to monitor the size and the growth of E. verrucosa

The (endo)cannabinoid signaling in female reproduction: What are the latest advances?
Cannabis extracts like marijuana have the highest consumption rate worldwide. Yet, their societal acceptance as recreational and therapeutic drugs could represent a serious hazard to female human reproduction, because cannabis ingredients [termed (phyto)cannabinoids] can perturb an endogenous system of lipid signals known as endocannabinoids. Accumulated evidence on animal models and humans has demonstrated a crucial role of these endogenous signals on different aspects of female reproduction, where they act through an ensamble of proteins that synthesize, transport, degrade and traffic them. Several reports have recently evidenced the potential role of endocannabinoids as biomarkers of female infertility for disease treatment and prevention, as well as their possible epigenetic effects on pregnancy. The purpose of this review is to provide an update of data collected in the last decade on the effects of cannabinoids and endocannabinoids on female reproductive events, from development and maturation of follicles and oocytes, to fertilization, oviductal transport, implantation and labor. In this context, a particular attention has ben devoted to the ovary and the production of fertilizable oocytes, because recent studies have addressed this hot topic with conflicting results among species
Endocannabinoid System Components of the Female Mouse Reproductive Tract Are Modulated during Reproductive Aging
The endocannabinoid (eCB) system has gained ground as a key modulator of several female fertility-related processes, under physiological/pathological conditions. Nevertheless, its modulation during reproductive aging remains unclear. This study aimed to investigate the expression levels of the main receptors (cannabinoid receptor 1,CB1; cannabinoid receptor 2, CB2; G-protein coupled receptor, GPR55; and transient receptor potential vanilloid type 1 channel, TRPV1) and metabolic enzymes (N-acylphosphatidylethanolamine phospholipase D, NAPE-PLD; fatty acid amide hydrolase, FAAH; monoacylglycerol lipase, MAGL; and diacylglycerol lipase, DAGL-α and -β) of this system in the ovaries, oviducts, and uteri of mice at prepubertal, adult, late reproductive, and post-reproductive stages through quantitative ELISA and immunohistochemistry. The ELISA showed that among the receptors, TRPV1 had the highest expression and significantly increased during aging. Among the enzymes, NAPE-PLD, FAAH, and DAGL-β were the most expressed in these organs at all ages, and increased age-dependently. Immunohistochemistry revealed that, regardless of age, NAPE-PLD and FAAH were mainly found in the epithelial cells facing the lumen of the oviduct and uteri. Moreover, in ovaries, NAPE-PLD was predominant in the granulosa cells, while FAAH was sparse in the stromal compartment. Of note, the age-dependent increase in TRPV1 and DAGL-β could be indicative of increased inflammation, while that of NAPE-PLD and FAAH could suggest the need to tightly control the levels of the eCB anandamide at late reproductive age. These findings offer new insights into the role of the eCB system in female reproduction, with potential for therapeutic exploitation
Design of a Microwave Sensor for Measurement of Water in Fuel Contamination
In this paper the modeling and the design of a microwave sensor is proposed for real-time fuel quality monitoring, with particular regard to water-in-fuel measurement. A mechanical and fluid-dynamic design has been performed, evaluating the stiffness, the robustness of the sensor structure and the pressure field acting on it. Furthermore, an electromagnetic study has been carried out to verify the validity of the physical measurement principle. The adopted procedure involves the modeling of the sensor in CAD and the simulation of its behavior. The sensor, based on microwave transmittance, is attractive because of its simplicity and foreseeable low cost
Comparing Haemophilus influenzae type b Conjugate Vaccine Schedules: Systematic Review and Meta-Analysis of Vaccine Trials
Background: The optimal schedule and the need for a booster dose are unclear for
Haemophilus influenzae type b (Hib) conjugate vaccines. We systematically reviewed relative
effects of Hib vaccine schedules.
Methods: We searched 21 databases to May 2010 or June 2012 and selected randomized
controlled trials (RCTs) or quasi-RCTs that compared different Hib schedules (three primary
doses with no booster dose [3p+0], 3p+1 and 2p+1) or different intervals in primary schedules
and between primary and booster schedules. Outcomes were clinical efficacy, nasopharyngeal
carriage and immunological response. Results were combined in random-effects meta-analysis.
Results: Twenty trials from 15 countries were included; 16 used vaccines conjugated to tetanus
toxoid (PRP-T). No trials assessed clinical or carriage outcomes. Twenty trials examined
immunological outcomes and found few relevant differences. Comparing PRP-T 3p+0 with 2p+0
there was no difference in seropositivity at the 1.0μg/ml threshold by six months after the last
primary dose (combined risk difference -0.02, 95%CI -0.10, 0.06). Only small differences were
seen between schedules starting at different ages, with different intervals between primary
doses, or with different intervals between primary and booster doses. Individuals receiving a
booster were more likely to be seropositive than those at the same age who did not.
Conclusions: There is no clear evidence from trials that any 2p+1, 3p+0 or 3p+1 schedule of Hib
conjugate vaccine is likely to provide better protection against Hib disease than other schedules.
Until more data become available, scheduling is likely to be determined by epidemiological and
programmatic considerations in individual settings
Anthropogenic sinkholes of the city of Naples, Italy: an update
In recent years, the study of anthropogenic sinkholes in densely urbanized areas has attracted the attention of both researchers and land management entities. The city of Naples (Italy) has been frequently affected by processes generating such landforms in the last decades: for this reason, an update of the sinkhole inventory and a preliminary susceptibility estimation are proposed in this work. Starting from previous data, not modified since 2010, a total of 270 new events occurred in the period February 2010–June 2021 were collected through the examination of online newspapers, local daily reports, council chronicle news and field surveys. The final consistence of the updated inventory is of 458 events occurred between 1880 and 2021, distributed through time with an increasing trend in frequency. Spatial analysis of sinkholes indicates a concentration in the central sector of the city, corresponding to its ancient and historic centre, crossed by a dense network of underground tunnels and cavities. Cavity-roof collapse is confirmed as one of the potential genetic types, along with processes related to rainfall events and service lines damage. A clear correlation between monthly rainfall and the number of triggered sinkholes was identified. Finally, a preliminary sinkhole susceptibility assessment, carried out by Frequency Ratio method, confirms the central sector of city as that most susceptible to sinkholes and emphasizes the predisposing role of service lines, mostly in the outermost areas of the city
