    Heritability of peach tree resistance to bacterial leaf spot.

    The objective of this work was to evaluate the broad-sense heritability reaction to bacterial leaf spot (Xanthomonas arboricola pv. pruni), in peach tree populations obtained from directed crosses. Disease severity and defoliation of the genotypes were evaluated in field conditions, with posterior measurement of the healthy leaf area duration (HAD). The observed average heritability (0.51) indicates that the use of the evaluated genitors can be effective for the development of cultivars with higher resistance to the disease.Notas Científicas.     To what extent have the links between ecosystem services and human well-being been researched in Africa, Asia, and Latin America?

    Most studies to date assume that there are multiple relationships between ecosystem services and human well-being, but there are few studies that quantify these relationships. Our objectives were: (1) to investigate the trends and understudied areas within ecosystem services and well-being research; and (2) within these general trends, to analyze to what extent the linkages between ecosystem services and well-being presented in empirical research in Africa, Asia, and Latin America were part of a tested hypothesis, and to assess which conceptual frameworks were used in understanding this interface. The results of this study highlighted that most publications assumed that ecosystem services and well-being were interlinked but did not analyze their relationship as part of the hypothesis to test. While different frameworks on well-being were adopted by empirical research, only one out of the 29 post-Millennium Ecosystem Assessment (MEA) conceptual frameworks that illustrate the linkages between ecosystem services and well-being was documented, and most case studies adopted the MEA. Finally, trade-offs and synergies between ecosystem services and disaggregated well-being were understudied. Considering these knowledge gaps in future studies will help empirical ecosystem services research to simultaneously contribute to improved well-being and environmental sustainability when applied at multiple policy or institutional levels.     Preclinical Evaluation of a Replication-Deficient Intranasal ΔNS1 H5N1 Influenza Vaccine

    We developed a novel intranasal influenza vaccine approach that is based on the construction of replication-deficient vaccine viruses that lack the entire NS1 gene (ΔNS1 virus). We previously showed that these viruses undergo abortive replication in the respiratory tract of animals. The local release of type I interferons and other cytokines and chemokines in the upper respiratory tract may have a “self-adjuvant effect”, in turn increasing vaccine immunogenicity. As a result, ΔNS1 viruses elicit strong B- and T- cell mediated immune responses.), one dose of vaccine delivered intranasally was sufficient for the induction of antibodies against homologous A/Vietnam/1203/04 and heterologous A/Indonesia/5/05 H5N1 strains.Our findings show that intranasal immunization with the replication deficient H5N1 ΔNS1 vaccine candidate is sufficient to induce a protective immune response against H5N1 viruses.     Integrated single-cell genetic and transcriptional analysis suggests novel drivers of chronic lymphocytic leukemia

    Intra-tumoral genetic heterogeneity has been characterized across cancers by genome sequencing of bulk tumors, including chronic lymphocytic leukemia (CLL). In order to more accurately identify subclones, define phylogenetic relationships, and probe genotype-phenotype relationships, we developed methods for targeted mutation detection in DNA and RNA isolated from thousands of single cells from five CLL samples. By clearly resolving phylogenic relationships, we uncovered mutated LCP1 and WNK1 as novel CLL drivers, supported by functional evidence demonstrating their impact on CLL pathways. Integrative analysis of somatic mutations with transcriptional states prompts the idea that convergent evolution generates phenotypically similar cells in distinct genetic branches, thus creating a cohesive expression profile in each CLL sample despite the presence of genetic heterogeneity. Our study highlights the potential for single-cell RNA-based targeted analysis to sensitively determine transcriptional and mutational profiles of individual cancer cells, leading to increased understanding of driving events in malignancy.</p

    Influenza A Virus Induces an Immediate Cytotoxic Activity in All Major Subsets of Peripheral Blood Mononuclear Cells

    A replication defective influenza A vaccine virus (delNS1 virus) was developed. Its attenuation is due to potent stimulation of the innate immune system by the virus. Since the innate immune system can also target cancer cells, we reasoned that delNS1 virus induced immune-stimulation should also lead to the induction of innate cytotoxic effects towards cancer cells.Peripheral blood mononuclear cells (PBMCs), isolated CD56+, CD3+, CD14+ and CD19+ subsets and different combinations of the above subsets were stimulated by delNS1, wild type (wt) virus or heat inactivated virus and co-cultured with tumor cell lines in the presence or absence of antibodies against the interferon system. Stimulation of PBMCs by the delNS1 virus effectively induced cytotoxicity against different cancer cell lines. Surprisingly, virus induced cytotoxicity was exerted by all major subtypes of PBMCs including CD56+, CD3+, CD14+ and CD19+ cells. Virus induced cytotoxicity in CD3+, CD14+ and CD19+ cells was dependent on virus replication, whereas virus induced cytotoxicity in CD56+ cells was only dependent on the binding of the virus. Virus induced cytotoxicity of isolated cell cultures of CD14+, CD19+ or CD56+ cells could be partially blocked by antibodies against type I and type II (IFN) interferon. In contrast, virus induced cytotoxicity in the complete PBMC preparation could not be inhibited by blocking type I or type II IFN, indicating a redundant system of activation in whole blood.Our data suggest that apart from their well known specialized functions all main subsets of peripheral blood cells also initially exert a cytotoxic effect upon virus stimulation. This closely links the innate immune system to the adaptive immune response and renders delNS1 virus a potential therapeutic tool for viro-immunotherapy of cancer
