Perencanaan Strategi dan Kebijakan untuk Mengukur Keselarasan TI dengan Bisnis Menggunakan SAMM

Seminar on Implementation of Pharmaceutical Service Standards During the Covid-19 Pandemic at Granmed Lubuk Pakam Hospital at 2020
Pharmacist pharmaceutical services for patients related to pharmaceutical preparations in order to improve the quality of pharmaceutical services in hospitals, the implementation of the Minister of Health Regulation no. 72 of 2016 concerning pharmaceutical service standards at Grandmed Hospitals with patient satisfaction during the Covid 19 2021 pandemic. Hospital pharmacy services involve pharmacists in order to supervise and control all activities related to healing treatment of sufferers in the aim of practicing patient safety standards which include Pharmaceutical supplies management activities starting from demand planning to controlling pharmaceutical supplies as well as clinical pharmacy service activities that are oriented towards reducing the risk of drug use errors in hospitals. This type of design was made by collecting data from 59 visiting patients at a certain point in time. Location at GrandMed Hospital Lubuk Pakam. The data obtained were compared with existing standards in the literature and analyzed using the chi-square test. It is known that out of 25 people (42.4%) respondents stated that clinical pharmacy services were not good. Of the 34 people (57.6%) respondents who stated that clinical pharmacy services were good for each employee, they had been given an understanding regarding clinical pharmacy services which had been socialized every morning briefing to maximize serving patients so that patients were satisfied with the services at Grandmed Hospital. This study can be concluded that the implementation of pmk no. 72 of 2016 there is a relationship between clinical pharmacy services and patient satisfaction during the Covid 19 pandemic at Grandmed Lubuk Pakam Hospital in 2021 (pvalue = 0.007)
Limitations of MTT and MTS-Based Assays for Measurement of Antiproliferative Activity of Green Tea Polyphenols
The chemopreventive effect of green tea polyphenols, such as (-)-epigallocatechin-3-gallate (EGCG), has been well demonstrated in cell culture studies. However, a wide range of IC(50) concentrations has been observed in published studies of the anti-proliferative activity of EGCG from different laboratories. Although the susceptibility to EGCG treatment is largely dependent on cancer cell type, the particular cell viability and proliferation assays utilized may significantly influence quantitative results reported in the literature.We compared five widely used methods to measure cell proliferation and viability after EGCG treatment using LNCaP prostate cancer cells and MCF-7 breast cancer cells. Both methods using dyes to quantify adenosine triphosphate (ATP) and deoxynucleic acid (DNA) showed accuracy in the measurement of viable cells when compared to trypan blue assay and results showed good linear correlation (r = 0.95). However, the use of MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) as indicators of metabolically active mitochondria overestimated the number of viable cells by comparison with the ATP, DNA, or trypan blue determinations. As a result, the observed IC(50) concentration of EGCG was 2-fold higher using MTT and MTS compared to dyes quantifying ATP and DNA. In contrast, when cells were treated with apigenin MTT and MTS assays showed consistent results with ATP, DNA, or trypan blue assays.These results demonstrate that MTT and MTS -based assays will provide an underestimation of the anti-proliferative effect of EGCG, and suggest the importance of careful evaluation of the method for in vitro assessment of cell viability and proliferation depending on the chemical nature of botanical supplements
Chatbot Application for the Dissemination of Election Information
The lack of media for election socialization that provides more accessibility for the deaf makes it difficult for them to obtain information given verbally, either by the KPU (The General Elections Commission) or the success team of candidate pairs who will compete. This condition could affect them to unvote. This research aims to design an application with an easy-to-use interface and help deaf people get accurate and accessible news and information. The method used to fulfill application development needs was a descriptive analysis method with a qualitative approach. This study used the Goal-Directed Design method to build an easy-to-use interface for deaf people. According to these applications, the deaf could easily understand election news information because videos are equipped with sign language and subtitles. These applications help the deaf quickly access and obtain the information about the election socialization that will hold and determine whom the candidates will compete in the election such as news feature, ask KPU, listen to reading, etc. The chatbot technology is expected to make the information search process more personal so that deaf people feel more comfortable using the applicatio
Caloric content of Metapenaeus dobsoni

(3) from proximate composition of body tissues. The results showed that any
one of the three methods could be used with reasonable accuracy. Caloric
content can also be worked out from the carbon values. The conversion factor
for Metapenaeus dobsoni is 1 mg carbon = 14.57 calories
Deletion of chromosomal region 8p21 confers resistance to Bortezomib and is associated with upregulated Decoy trail receptor expression in patients with multiple myeloma
Loss of the chromosomal region 8p21 negatively effects survival in patients with multiple myeloma (MM) that undergo autologous stem cell transplantation (ASCT). In this study, we aimed to identify the immunological and molecular consequences of del(8)(p21) with regards to treatment response and bortezomib resistance. In patients receiving bortezomib as a single first line agent without any high-dose therapy, we have observed that patients with del(8)(p21) responded poorly to bortezomib with 50% showing no response while patients without the deletion had a response rate of 90%. In vitro analysis revealed a higher resistance to bortezomib possibly due to an altered gene expression profile caused by del(8)(p21) including genes such as TRAIL-R4, CCDC25, RHOBTB2, PTK2B, SCARA3, MYC, BCL2 and TP53. Furthermore, while bortezomib sensitized MM cells without del(8)(p21) to TRAIL/APO2L mediated apoptosis, in cells with del(8)(p21) bortezomib failed to upregulate the pro-apoptotic death receptors TRAIL-R1 and TRAIL-R2 which are located on the 8p21 region. Also expressing higher levels of the decoy death receptor TRAIL-R4, these cells were largely resistant to TRAIL/APO2L mediated apoptosis. Corroborating the clinical outcome of the patients, our data provides a potential explanation regarding the poor response of MM patients with del(8)(p21) to bortezomib treatment. Furthermore, our clinical analysis suggests that including immunomodulatory agents such as Lenalidomide in the treatment regimen may help to overcome this negative effect, providing an alternative consideration in treatment planning of MM patients with del(8)(p21)
Metabolic Effects of Acute Thiamine Depletion Are Reversed by Rapamycin in Breast and Leukemia Cells
Thiamine-dependent enzymes (TDEs) control metabolic pathways that are frequently altered in cancer and therefore present cancer-relevant targets. We have previously shown that the recombinant enzyme thiaminase cleaves and depletes intracellular thiamine, has growth inhibitory activity against leukemia and breast cancer cell lines, and that its growth inhibitory effects were reversed in leukemia cell lines by rapamycin. Now, we first show further evidence of thiaminase therapeutic potential by demonstrating its activity against breast and leukemia xenografts, and against a primary leukemia xenograft. We therefore further explored the metabolic effects of thiaminase in combination with rapamycin in leukemia and breast cell lines. Thiaminase decreased oxygen consumption rate and increased extracellular acidification rate, consistent with the inhibitory effect of acute thiamine depletion on the activity of the TDEs pyruvate dehydrogenase and 2-oxoglutarate dehydrogenase complexes; these effects were reversed by rapamycin. Metabolomic studies demonstrated intracellular thiamine depletion and the presence of the thiazole cleavage product in thiaminase-treated cells, providing validation of the experimental procedures. Accumulation of ribose and ribulose in both cell lines support the thiaminase-mediated suppression of the TDE transketolase. Interestingly, thiaminase suppression of another TDE, branched chain amino ketoacid dehydrogenase (BCKDH), showed very different patterns in the two cell lines: in RS4 leukemia cells it led to an increase in BCKDH substrates, and in MCF-7 breast cancer cells it led to a decrease in BCKDH products. Immunoblot analyses showed corresponding differences in expression of BCKDH pathway enzymes, and partial protection of thiaminase growth inhibition by gabapentin indicated that BCKDH inhibition may be a mechanism of thiaminase-mediated toxicity. Surprisingly, most of thiaminase-mediated metabolomic effects were also reversed by rapamycin. Thus, these studies demonstrate that acute intracellular thiamine depletion by recombinant thiaminase results in metabolic changes in thiamine-dependent metabolism, and demonstrate a previously unrecognized role of mTOR signaling in the regulation of thiamine-dependent metabolism
