    Effects of Methotrexate on IL-6alphar, VCAM-1 and NF Kappa B Expression in a Rat Model of Metabolic Syndrome

    Background: In this study, we used Methotrexate (Mtx) to examine the role of immunomodulation on the activation of IL-6 and VCAM-1, which could generate a microenvironment that supports cardiovascular remodelling.Methods: Male WKY and SHR rats were separated into five groups: Control, FFR: WKY rats receiving a 10% (w/v) fructose solution during all 12 weeks, SHR, FFHR: SHR receiving a 10% (w/v) fructose solution during all 12 weeks and FFHR+Mtx (0,3 mg/kg intraperitoneal one injection per day week for 6 weeks) (n = 8 per group). Metabolic variables and systolic blood pressure were measured. Cardiac and vascular remodelling was also evaluated. To assess this, IL-6R and VCAM-1 immunostaining techniques were used.Results: The FFHR experimental model developed metabolic syndrome, vascular and cardiac remodelling, and vascular inflammation because of increased expression of IL-6 and VCAM-1. Chronic treatment with Mtx completely or partiality reversed the variables studied.Conclusions: The results demonstrated an impact on immunomodulation after mtx treatment, which included a reduction in vascular inflammation and a favourable reduction in metabolic and structural parameters.Fil: Renna, Nicolas Federico.     Signal reconstruction in the presence of side information: The impact of projection kernel design

    This paper investigates the impact of projection design on the reconstruction of high-dimensional signals from low-dimensional measurements in the presence of side information. In particular, we assume that both the signal of interest and the side information are described by a joint Gaussian mixture model (GMM) distribution. Sharp necessary and sufficient conditions on the number of measurements needed to guarantee that the average reconstruction error approaches zero in the low-noise regime are derived, for both cases when the side information is available at the decoder or at the decoder and encoder. Numerical results are also presented to showcase the impact of projection design on applications with real imaging data in the presence of side information

    On the design of linear projections for compressive sensing with side information

    In this paper, we study the problem of projection kernel design for the reconstruction of high-dimensional signals from low-dimensional measurements in the presence of side information, assuming that the signal of interest and the side information signal are described by a joint Gaussian mixture model (GMM). In particular, we consider the case where the projection kernel for the signal of interest is random, whereas the projection kernel associated to the side information is designed. We then derive sufficient conditions on the number of measurements needed to guarantee that the minimum meansquared error (MMSE) tends to zero in the low-noise regime. Our results demonstrate that the use of a designed kernel to capture side information can lead to substantial gains in relation to a random one, in terms of the number of linear projections required for reliable reconstruction

    Unconventional secretion of α-Crystallin B requires the Autophagic pathway and is controlled by phosphorylation of its serine 59 residue

    α-Crystallin B (CRYAB or HspB5) is a chaperone member of the small heat-shock protein family that prevents aggregation of many cytosolic client proteins by means of its ATP-independent holdase activity. Surprisingly, several reports show that CRYAB exerts a protective role also extracellularly, and it has been recently demonstrated that CRYAB is secreted from human retinal pigment epithelial cells by an unconventional secretion pathway that involves multi-vesicular bodies. Here we show that autophagy is crucial for this unconventional secretion pathway and that phosphorylation at serine 59 residue regulates CRYAB secretion by inhibiting its recruitment to the autophagosomes. In addition, we found that autophagosomes containing CRYAB are not able to fuse with lysosomes. Therefore, CRYAB is capable to highjack and divert autophagosomes toward the exocytic pathway, inhibiting their canonical route leading to the lysosomal compartment. Potential implications of these findings in the context of disease-associated mutant proteins turn-over are discussed

    Thermogravimetric study of a phase change slurry: effect of variable conditions

    Microcapsules containing Phase Change Materials (MPCM) are widely used for passive systems in energy storage. When MPCM are mixed with a carrier fluid, Phase Change Slurries (PCS) are used for heat transfer fluids in active systems or heat transport systems. The thermal behavior of PCS can be measured as dry or wet basis, resulting in important differences in weight losses. This study explores the optimum conditions for analyzing the thermal behavior of dried PCS by thermogravimetric analysis (TGA) varying the parameter conditions for obtaining peak temperature and heat flow (latent heat). The factors that were taken into account were the atmosphere of study (air and nitrogen) and the heating rate (0.5, 1, 5, and 10 °C·min−1). The best conditions to determine peak temperature are at 1 °C·min−1 and in N2 atmosphere, whereas the decomposition fusion/latent heat of the sample is improved at higher heating velocities towards 10 °C·min−1.The work is partially funded by the Spanish government (ENE2015-64117-C5-2R).     Insights into Thymus Development and Viral Thymic Infections

    T-cell development in the thymus is a complex and highly regulated process, involving a wide variety of cells and molecules which orchestrate thymocyte maturation into either CD4+ or CD8+ single-positive (SP) T cells. Here, we briefly review the process regulating T-cell differentiation, which includes the latest advances in this field. In particular, we highlight how, starting from a pool of hematopoietic stem cells in the bone marrow, the sequential action of transcriptional factors and cytokines dictates the proliferation, restriction of lineage potential, T-cell antigen receptors (TCR) gene rearrangements, and selection events on the T-cell progenitors, ultimately leading to the generation of mature T cells. Moreover, this review discusses paradigmatic examples of viral infections affecting the thymus that, by inducing functional changes within this lymphoid gland, consequently influence the behavior of peripheral mature T-lymphocytes
