    Immunogenicity of a Prime-Boost Vaccine Containing the Circumsporozoite Proteins of Plasmodium vivax in Rodents

    Plasmodium vivax is the most widespread and the second most prevalent malaria-causing species in the world. Current measures used to control the transmission of this disease would benefit from the development of an efficacious vaccine. in the case of the deadly parasite P. falciparum, the recombinant RTS,S vaccine containing the circumsporozoite antigen (CSP) consistently protects 30 to 50% of human volunteers against infection and is undergoing phase III clinical trials in Africa with similar efficacy. These findings encouraged us to develop a P. vivax vaccine containing the three circulating allelic forms of P. vivax CSP. Toward this goal, we generated three recombinant bacterial proteins representing the CSP alleles, as well as a hybrid polypeptide called PvCSP-All-CSP-epitopes. This hybrid contains the conserved N and C termini of P. vivax CSP and the three variant repeat domains in tandem. We also generated simian and human recombinant replication-defective adenovirus vectors expressing PvCSP-All-CSP-epitopes. Mice immunized with the mixture of recombinant proteins in a formulation containing the adjuvant poly(I.C) developed high and long-lasting serum IgG titers comparable to those elicited by proteins emulsified in complete Freund's adjuvant.     Hemoglobin Uptake by Paracoccidioides spp. Is Receptor-Mediated

    Iron is essential for the proliferation of fungal pathogens during infection. The availability of iron is limited due to its association with host proteins. Fungal pathogens have evolved different mechanisms to acquire iron from host; however, little is known regarding how Paracoccidioides species incorporate and metabolize this ion. In this work, host iron sources that are used by Paracoccidioides spp. were investigated. Robust fungal growth in the presence of the iron-containing molecules hemin and hemoglobin was observed. Paracoccidioides spp. present hemolytic activity and have the ability to internalize a protoporphyrin ring. Using real-time PCR and nanoUPLC-MSE proteomic approaches, fungal growth in the presence of hemoglobin was shown to result in the positive regulation of transcripts that encode putative hemoglobin receptors, in addition to the induction of proteins that are required for amino acid metabolism and vacuolar protein degradation. In fact, one hemoglobin receptor ortholog, Rbt5, was identified as a surface GPI-anchored protein that recognized hemin, protoporphyrin and hemoglobin in vitro. Antisense RNA technology and Agrobacterium tumefaciensmediated transformation were used to generate mitotically stable Pbrbt5 mutants. The knockdown strain had a lower survival inside macrophages and in mouse spleen when compared with the parental strain, which suggested that Rbt5 could act as a virulence factor. In summary, our data indicate that Paracoccidioides spp. can use hemoglobin as an iron source most likely through receptor-mediated pathways that might be relevant for pathogenic mechanisms

    Orientation bias of optically selected galaxy clusters and its impact on stacked weak-lensing analyses

    Weak-lensing measurements of the averaged shear profiles of galaxy clusters binned by some proxy for cluster mass are commonly converted to cluster mass estimates under the assumption that these cluster stacks have spherical symmetry. In this paper, we test whether this assumption holds for optically selected clusters binned by estimated optical richness. Using mock catalogues created from N-body simulations populated realistically with galaxies, we ran a suite of optical cluster finders and estimated their optical richness. We binned galaxy clusters by true cluster mass and estimated optical richness and measure the ellipticity of these stacks. We find that the processes of optical cluster selection and richness estimation are biased, leading to stacked structures that are elongated along the line of sight. We show that weak-lensing alone cannot measure the size of this orientation bias. Weak-lensing masses of stacked optically selected clusters are overestimated by up to 3–6 per cent when clusters can be uniquely associated with haloes. This effect is large enough to lead to significant biases in the cosmological parameters derived from large surveys like the Dark Energy Survey, if not calibrated via simulations or fitted simultaneously. This bias probably also contributes to the observed discrepancy between the observed and predicted Sunyaev–Zel’dovich signal of optically selected clusters
