    Loss of correlation between HIV viral load and CD4+ T-cell counts in HIV/HTLV-1 co-infection in treatment naive Mozambican patients

    Seven hundred and four HIV-1/2-positive, antiretroviral therapy (ART) naïve patients were screened for HTLV-1 infection. Antibodies to HTLV-1 were found in 32/704 (4.5%) of the patients. Each co-infected individual was matched with two HIV mono-infected patients according to World Health Organization clinical stage, age +/-5 years and gender. Key clinical and laboratory characteristics were compared between the two groups. Mono-infected and co-infected patients displayed similar clinical characteristics. However, co-infected patients had higher absolute CD4+ T-cell counts (P = 0.001), higher percentage CD4+ T-cell counts (P < 0.001) and higher CD4/CD8 ratios (P < 0.001). Although HIV plasma RNA viral loads were inversely correlated with CD4+ T-cell-counts in mono-infected patients (P < 0.0001), a correlation was not found in co-infected individuals (P = 0.11). Patients with untreated HIV and HTLV-1 co-infection show a dissociation between immunological and HIV virological markers. Current recommendations for initiating ART and chemoprophylaxis against opportunistic infections in resource-poor settings rely on more readily available CD4+ T-cell counts without viral load parameters. These guidelines are not appropriate for co-infected individuals in whom high CD4+ T-cell counts persist despite high HIV viral load states. Thus, for co-infected patients, even in resource-poor settings, HIV viral loads are likely to contribute information crucial for the appropriate timing of ART introduction

    O programa nacional de fortalecimento da agricultura familiar no Brasil: uma análise sobre a distribuição regional e setorial dos recursos.

    No Brasil, as políticas públicas para o espaço rural sempre tenderam a priorizar a agricultura patronal, em detrimento dos agricultores familiares. Todavia, os estudos realizados pelos órgãos FAO - INCRA deram subsídio para a criação do Programa Nacional de Fortalecimento da Agricultura Familiar (PRONAF), resultando em um novo direcionamento dos investimentos públicos, os quais passaram a contemplar o segmento dos agricultores familiares. Entende-se o PRONAF como uma política não-compensatória, que, apesar de seus problemas, tem contribuído de fato para mudanças e melhorias no espaço agrário brasileiro. Desde sua criação no final da década de 1990, o PRONAF passou por várias mudanças em sua estrutura administrativa e operacional, a fim de alcançar seus objetivos e adequar-se face a complexa realidade social agrária brasileira. Sendo assim, o presente estudo visa discutir as ações do Estado por meio desse Programa, a partir de suas linhas de atuação, bem como analisar a distribuição de suas concessões de crédito regional e setorialmente. Assim, os procedimentos metodológicos utilizados para a realização deste trabalho compreendem pesquisa bibliográfica e documental, além de pesquisa em fontes secundárias, no intuito de obter dados e informações relevantes para a análise das relações sociais estabelecidas em meio a esse processo de concretização e espacialização desse Programa. Dentre as implicações do PRONAF pode-se notar em âmbito nacional, uma diminuição da disparidade regional brasileira, bem como a preocupação que o Programa tem demonstrado com os aspectos socioculturais locais e regionais, como forma de garantir que seus investimentos perpassem a dimensão econômica, mas valorize outras dimensões, a exemplo dos elementos culturais

    Disparities in allele frequencies and population differentiation for 101 disease-associated single nucleotide polymorphisms between Puerto Ricans and Non-Hispanic Whites

    BACKGROUND. Variations in gene allele frequencies can contribute to differences in the prevalence of some common complex diseases among populations. Natural selection modulates the balance in allele frequencies across populations. Population differentiation (FST) can evidence environmental selection pressures. Such genetic information is limited in Puerto Ricans, the second largest Hispanic ethnic group in the US, and a group with high prevalence of chronic disease. We determined allele frequencies and population differentiation for 101 single nucleotide polymorphisms (SNPs) in 30 genes involved in major metabolic and disease-relevant pathways in Puerto Ricans (n = 969, ages 45–75 years) and compared them to similarly aged non-Hispanic whites (NHW) (n = 597). RESULTS. Minor allele frequency (MAF) distributions for 45.5% of the SNPs assessed in Puerto Ricans were significantly different from those of NHW. Puerto Ricans carried risk alleles in higher frequency and protective alleles in lower frequency than NHW. Patterns of population differentiation showed that Puerto Ricans had SNPs with exceptional FST values in intronic, non-synonymous and promoter regions. NHW had exceptional FST values in intronic and promoter region SNPs only. CONCLUSION.     Development and distribution of the non-indigenous Pacific oyster (Crassostrea gigas) in the Dutch Wadden Sea

    Pacific oysters (Crassostrea gigas) were first observed in the Dutch Wadden Sea near Texel in 1983. The population increased slowly in the beginning but grew exponentially from the mid-1990s onwards, although now some stabilisation seems to be occurring. They occur on a variety of substrates such as mussel beds (Mytilus edulis), shell banks, dikes and poles. After initial settlement spat may fall on older individuals and congregate to dense clumps and subsequently form reefs. Individual Pacific oysters grow 3–4 cm long in their first year and 2–3 cm in their second year. Many mussel beds (Mytilus edulis) are slowly taken over by Pacific oysters, but there are also several reports of mussel spat settling on Pacific oyster reefs. This might in the end result in combined reefs. Successful Pacific oyster spat fall seems to be related to high summer temperatures, but also after mild summers much spat can be found on old (Pacific oyster) shells. Predation is of limited importance. Mortality factors are unknown, but every now and then unexplained mass mortality occurs. The gradual spread of the Pacific oyster in the Dutch Wadden Sea is documented in the first instance based on historical and anecdotal information. At the start of the more in-depth investigation in 2002, Pacific oysters of all size classes were already present near Texel. Near Ameland the development could be followed from the first observed settlement.     Most of the computational study of election problems has assumed that each voter's preferences are, or should be extended to, a total order. However in practice voters may have preferences with ties. We study the complexity of manipulative actions on elections where voters can have ties, extending the definitions of the election systems (when necessary) to handle voters with ties. We show that for natural election systems allowing ties can both increase and decrease the complexity of manipulation and bribery, and we state a general result on the effect of voters with ties on the complexity of control.Comment: A version of this paper will appear in ADT-201

    Studying the Pulsation of Mira Variables in the Ultraviolet

    We present results from an empirical study of the Mg II h & k emission lines of selected Mira variable stars, using spectra from the International Ultraviolet Explorer (IUE). The stars all exhibit similar Mg II behavior during the course of their pulsation cycles. The Mg II flux always peaks after optical maximum near pulsation phase 0.2-0.5, although the Mg II flux can vary greatly from one cycle to the next. The lines are highly blueshifted, with the magnitude of the blueshift decreasing with phase. The widths of the Mg II lines are also phase-dependent, decreasing from about 70 km/s to 40 km/s between phase 0.2 and 0.6. We also study other UV emission lines apparent in the IUE spectra, most of them Fe II lines. These lines are much narrower and not nearly as blueshifted as the Mg II lines.     Screening an in house alkaloids library using Voltage-Sensor Probes for new modulators of voltage-gated sodium channels

    Voltage-gated sodium channels (Nav) are molecular targets of clinically used drugs for treatments of various diseases (epilepsy, chronic pain, cardiac arrhythmia…) and also of numerous animal and plant neurotoxins. The development of easy-to-use screening assays for searching new ligands from chemicals libraries, animal venoms or plant extracts represents a challenge of a great interest to generate therapeutic hits. Here, we used the mammalian GH3B6 pituitary cell line, which constitutively expresses three different neuronal Nav channel isoforms (Nav1.2, Nav1.3 and Nav1.6), to identify novel compounds of pharmacological interest from a library of in-house vegetal alkaloids. The screening is based on a method using Voltage-Sensor Probes (VSPs) that we adapted to detect both activators and blockers of Nav channels. Over the 84 pure alkaloids or plant extracts that were screened, 17 increased the VSP signal. They operated as gating modifier, showing an action mechanism similar to that of batrachotoxin (BTX), known to strongly inhibit Nav channel inactivation. The remaining 67 plant products were assessed for their potency to inhibit BTX-induced VSP signal. We further selected 11 alkaloids as efficient Nav channels inhibitors. We focused our attention on two structural analogs belonging to the aporphine family, liriodenine and oxostephanine, which differ only by a methoxy group. Whereas liriodenine has been already described as a Nav channels blocker, oxostephanine has not been yet documented as an ion channel modulator. In conclusion, the novel VSPs-based screening assay we developed is a suitable method to challenge the discovery and to assess the activity of novel ligands on Nav channels
