    Role of Sensation Seeking in Sensitivity to d-amphetamine Reinforcement

    Psychomotor stimulant abuse is a significant public health problem. While many individuals experiment with stimulants, there is marked variability in individuals\u27 behavioral and subjective response to these drugs and these differences may be associated with their risk for abuse. One characteristic shown to be associated with drug abuse is sensation seeking, defined as the seeking of novel sensations and experiences and the willingness to take risks for the sake of such experiences. While observational studies have shown that individuals with elevated sensation seeking are more likely to report stimulant use and abuse, less clear is whether subjective and behavioral response to acute stimulant administration may vary as a function of sensation seeking status. We recently completed an outpatient laboratory study in which 37 healthy adults received repeated opportunities to sample and choose between d-amphetamine (d-AMPH; 5, 10, 20 mg/70kg) or placebo. That study provided an opportunity to examine associations between sensation seeking and d-AMPH choice and subjective response under rigorous double-blind experimental conditions. The Zuckerman Sensation Seeking Scale V was administered at intake, providing a Total sensation seeking score as well as four subscales (i.e., Experience Seeking, Disinhibition, Thrill and Adventure Seeking, Boredom Susceptibility). We hypothesized that elevated sensation seeking at intake would be associated with increased preference for d-AMPH over placebo in subsequent choice sessions, as well as greater positive d-AMPH subjective effects. Among males, increased baseline sensation seeking was associated with increased d-AMPH choice and positive subjective effects at the 5 and 10 mg/70 kg doses. Among females we found no significant associations between sensation seeking and d-AMPH choice or subjective effects. Finally, when the association between sensation seeking and other baseline characteristics was examined, there was a significant positive association with lifetime drug use as well as impulsivity. Taken together, our data suggest that elevated sensation seeking in males may be associated with increased sensitivity to d-AMPH reinforcement and positive subjective effects, suggesting increased vulnerability for stimulant use and abuse

    Quantum trajectories and their statistics for remotely entangled quantum bits

    We experimentally and theoretically investigate the quantum trajectories of jointly monitored transmon qubits embedded in spatially separated microwave cavities. Using nearly quantum-noise limited superconducting amplifiers and an optimized setup to reduce signal loss between cavities, we can efficiently track measurement-induced entanglement generation as a continuous process for single realizations of the experiment. The quantum trajectories of transmon qubits naturally split into low and high entanglement classes corresponding to half-parity collapse. The distribution of concurrence is found at any given time and we explore the dynamics of entanglement creation in the state space. The distribution exhibits a sharp cut-off in the high concurrence limit, defining a maximal concurrence boundary. The most likely paths of the qubits' trajectories are also investigated, resulting in three probable paths, gradually projecting the system to two even subspaces and an odd subspace. We also investigate the most likely time for the individual trajectories to reach their most entangled state, and find that there are two solutions for the local maximum, corresponding to the low and high entanglement routes. The theoretical predictions show excellent agreement with the experimental entangled qubit trajectory data.Comment: 11 pages and 4 figure

    First Foods Nutrition Curriculum for New Immigrant Families: A Pilot Study

    Background: Immigrant families arrive in the US from a variety of nutritional landscapes and educational experiences. Early childhood is a key time to intervene to set children on a healthy path. Creating nutritional education programs tailored for immigrant families may improve nutrition and health outcomes. Objective: To evaluate the First Foods curriculum as a tool for knowledge and behavior change for new immigrant families of young children. Methods: Immigrant caregivers of children less than 2 years old were invited to attend First Foods, a 4-class series. Each series was offered in 1 of 5 different languages (Arabic, Dari, Somali, Burmese, and Nepali). Recruitment occurred through community organizations, primary care clinics and Women, Infants, and Children (WIC), and classes were held in King County, Washington. The curriculum was developed and taught by a registered pediatric dietitian with input from general pediatricians, all experienced in the care of immigrant families. Classes were interpreted in the relevant language and course materials were translated. The classes were based on 4 themes -- 1) Child Eating and Development, 2) Eating Together, 3) Food Safety, and 4) Health Living -- and incorporated positive parenting and child development. Attendees completed pre- and post-surveys in their respective languages or in English. Descriptive statistics, chi-squared analyses, t-tests, and a multi-level linear regression model were conducted in Stata v14.0. Results: Participants in the classes included 47 caregivers (91% mothers). Nearly one-third had previously lived in a refugee camp. They had lived in the US a mean 5.5 years (95% CI: 3.8-7.2 years), attended a mean 8.6 years of school (95% CI: 7.1-10.1 years), and had a mean of 2.8 children (95% CI: 2.3-3.3 children). Classes ranged in size from 5 to 14 caregivers. Caregivers reported an improved understanding of 2 out of 4 methods to decrease risk of dental caries (drinking tap water, p = \u3c0.001; going to the dentist, p=0.02). They reported a decreased use of food as a reward from the pre- to the post-survey (p=0.027). Additionally, the caregivers reported increased frequency of considering sugar content in family foods (p=0.033), and decreased frequency of purchasing food at a convenience store, after participating in the curriculum (p=0.001). Conversely, there were several domains where caregivers did not show a change in their response. Conclusion: First Foods, a community-tailored, early childhood feeding curriculum for immigrant parents of young children, improved knowledge and behavior among caregivers from a variety of immigrant communities in some domains.     Integration of gray matter nodules into functional cortical circuits in periventricular heterotopia

    Alterations in neuronal circuitry are recognized as an important substrate of many neurological disorders, including epilepsy. Patients with the developmental brain malformation of periventricular nodular heterotopia (PNH) often have both seizures and dyslexia, and there is evidence to suggest that aberrant neuronal connectivity underlies both of these clinical features. We used task-based functional MRI (fMRI) to determine whether heterotopic nodules of gray matter in this condition are integrated into functional cortical circuits. Blood oxygenation level-dependent (BOLD) fMRI was acquired in eight participants with PNH during the performance of reading-related tasks. Evidence of neural activation within heterotopic gray matter was identified, and regions of cortical coactivation were then mapped systematically. Findings were correlated with resting-state functional connectivity results and with performance on the fMRI reading-related tasks. Six participants (75%) demonstrated activation within at least one region of gray matter heterotopia. Cortical areas directly overlying the heterotopia were usually coactivated (60%), as were areas known to have functional connectivity to the heterotopia in the task-free resting state (73%). Six of seven (86%) primary task contrasts resulted in heterotopia activation in at least one participant. Activation was most commonly seen during rapid naming of visual stimuli, a characteristic impairment in this patient population. Our findings represent a systematic demonstration that heterotopic gray matter can be metabolically coactivated in a neuronal migration disorder associated with epilepsy and dyslexia. Gray matter nodules were most commonly coactivated with the anatomically overlying cortex and other regions with resting-state connectivity to heterotopia. These results have broader implications for understanding the network pathogenesis of both seizures and reading disabilities

    Equine-assisted psychotherapy is one of various uses of horses in a therapeutic setting. The following paper provides an overview of this treatment method in therapy. Appropriate terminology and definitions are provided to clarify differences between equine-assisted psychotherapy and other equine therapies. Research provided shows that this treatment method is highly effective, especially for individuals who do not respond to traditional therapies. EAP can be utilized for a variety of conditions including depression, anxiety, PTSD, trauma, and more. The Equine Growth and Learning Association (EAGALA) model is the basis for most treatments involving EAP and has detailed guidelines and processes for formatting therapy sessions. Overall, equine-assisted psychotherapy is highly effective, however, further studies should be conducted involving larger populations to support EAP as a treatment

    Physiological consequences of abnormal connectivity in a developmental epilepsy

    Objective Many forms of epilepsy are associated with aberrant neuronal connections, but the relationship between such pathological connectivity and the underlying physiological predisposition to seizures is unclear. We sought to characterize the cortical excitability profile of a developmental form of epilepsy known to have structural and functional connectivity abnormalities. Methods We employed transcranial magnetic stimulation (TMS) with simultaneous electroencephalographic (EEG) recording in 8 patients with epilepsy from periventricular nodular heterotopia and matched healthy controls. We used connectivity imaging findings to guide TMS targeting and compared the evoked responses to single-pulse stimulation from different cortical regions. Results Heterotopia patients with active epilepsy demonstrated a relatively augmented late cortical response that was greater than that of matched controls. This abnormality was specific to cortical regions with connectivity to subcortical heterotopic gray matter. Topographic mapping of the late response differences showed distributed cortical networks that were not limited to the stimulation site, and source analysis in 1 subject revealed that the generator of abnormal TMS-evoked activity overlapped with the spike and seizure onset zone. Interpretation Our findings indicate that patients with epilepsy from gray matter heterotopia have altered cortical physiology consistent with hyperexcitability, and that this abnormality is specifically linked to the presence of aberrant connectivity. These results support the idea that TMS-EEG could be a useful biomarker in epilepsy in gray matter heterotopia, expand our understanding of circuit mechanisms of epileptogenesis, and have potential implications for therapeutic neuromodulation in similar epileptic conditions associated with deep lesions
