Making inexpensive drugs for the treatment of Human African Trypanosomiasis
Neglected tropical diseases (NTDs) affect the world’s poorest population; mainly in Africa, Asia, and Latin America. According to the WHO, more than one billion people are affected by NTDs. The connection between these areas is the lack of proper sanitation, and the persistence of the diseases, trap the affected countries in the poverty and disease cycle.
African sleeping sickness, a prevalent NTD, is a parasitic infection caused by two parasites from the Trypanosoma brucei species and is transmitted by the tsetse fly or congenitally. The disease is manifested by fever, severe headaches, irritability, extreme fatigue, swollen lymph nodes, and aching muscles and joints. When it reaches the central nervous system it can cause progressive confusion, personality changes, and other neurologic problems. The high-cost and limited number of the drugs contribute to the increase of the cases resulting in a great impact on global health.
The goal of our research is to develop inexpensive and effective drugs using low-cost organic starting materials that target the T. brucei parasite. Our target compounds are Mannich bases containing an aromatic moiety. Previous, but limited, studies have shown that this class of compounds demonstrates promising results against T. brucei. Our one step synthesis involves the use of nitro-substituted acetophenones, formaldehyde, and a variety of amines to make Mannich bases under conventional heating and microwave conditions. By changing the nitro-acetophenones and the amines used in the reaction, we hope to develop a robust drug library we can use to screen against T. brucei
My Religion and My People: Levels of Ingroup Identification and Christian Religious Orientations
People adopt a variety of approaches to practice their religion, but to date there has been no examination as to how identification with one’s community, with Americans, and with all humanity may relate to different religious orientations. Two studies tested the hypotheses that 1) intrinsic religious orientation would be associated with all three levels of ingroup identification, 2) extrinsic religious orientation would be associated with community-level identification only, 3) quest religious orientation would be associated with humanity-level identification only, and 4) orthodox religious orientation would be associated with community- and country-level identification only. Study 2 controlled for several individual difference variables. In both studies, participants (n = 285 for Study 1, n = 507 for Study 2) completed online surveys. The results across both studies showed that identifying with country was associated with orthodox Christianity, and identifying with all humanity was associated with intrinsic Christianity. Implications for research on religion and outgroup prejudice are discussed
Crime Rate Prediction using KNN
Crime is one of the most predominant and alarming aspects in our society and its prevention is a vital task. Crime analysis is a systematic way of detecting and investigating patterns and trends in crime. Thus, it becomes necessary to study various reasons, factors and relationship between different crimes that are occurring and ?nding the most appropriate methods to control and avoid more crimes. The main objective of this project is to classify clustered crimes based on occurrence frequency during different years. Data mining is used broadly in terms of analysis, investigation and discovery of patterns for occurrence of different crimes. In this work, various clustering approaches of data mining are used to analyze the crime data. The K-Nearest Neighbour (KNN) classi?cation is used for crime prediction. The proposed system can predict regions which have high probability for crime rate and can forecast crime prone areas. Instead of focusing on causes of crime occurrence like criminal background of offender, political enmity etcit will focuse mainly on crime factors of each day
Serum methylarginines and spirometry-measured lung function in older adults
Rationale: Methylarginines are endogenous nitric oxide synthase inhibitors that have been implicated in animal models of lung disease but have not previously been examined for their association with spirometric measures of lung function in humans.
Objectives: This study measured serum concentrations of asymmetric and symmetric dimethylarginine in a representative sample of older community-dwelling adults and determined their association with spirometric lung function measures.
Methods: Data on clinical, lifestyle, and demographic characteristics, methylated arginines, and L-arginine (measured using LC-MS/MS) were collected from a population-based sample of older Australian adults from the Hunter Community Study.
The five key lung function measures included as outcomes were Forced Expiratory Volume in 1 second, Forced Vital Capacity, Forced Expiratory Volume in 1 second to Forced Vital Capacity ratio, Percent Predicted Forced Expiratory Volume in 1 second, and Percent Predicted Forced Vital Capacity.
Measurements and Main Results: In adjusted analyses there were statistically significant independent associations between a) higher asymmetric dimethylarginine, lower Forced Expiratory Volume in 1 second and lower Forced Vital Capacity; and b) lower L-arginine/asymmetric dimethylarginine ratio, lower Forced Expiratory Volume in 1 second, lower Percent Predicted Forced Expiratory Volume in 1 second and lower Percent Predicted Forced Vital Capacity. By contrast, no significant associations were observed between symmetric dimethylarginine and lung function.
Conclusions: After adjusting for clinical, demographic, biochemical, and pharmacological confounders, higher serum asymmetric dimethylarginine was independently associated with a reduction in key measures of lung function. Further research is needed to determine if methylarginines predict the decline in lung function
The "Ram Effect": A "Non-Classical" Mechanism for Inducing LH Surges in Sheep
During spring sheep do not normally ovulate but exposure to a ram can induce ovulation. In some ewes an LH surge is induced immediately after exposure to a ram thus raising questions about the control of this precocious LH surge. Our first aim was to determine the plasma concentrations of oestradiol (E2) E2 in anoestrous ewes before and after the "ram effect" in ewes that had a "precocious" LH surge (starting within 6 hours), a "normal" surge (between 6 and 28h) and "late» surge (not detected by 56h). In another experiment we tested if a small increase in circulating E2 could induce an LH surge in anoestrus ewes. The concentration of E2 significantly was not different at the time of ram introduction among ewes with the three types of LH surge. "Precocious" LH surges were not preceded by a large increase in E2 unlike "normal" surges and small elevations of circulating E2 alone were unable to induce LH surges. These results show that the "precocious" LH surge was not the result of E2 positive feedback. Our second aim was to test if noradrenaline (NA) is involved in the LH response to the "ram effect". Using double labelling for Fos and tyrosine hydroxylase (TH) we showed that exposure of anoestrous ewes to a ram induced a higher density of cells positive for both in the A1 nucleus and the Locus Coeruleus complex compared to unstimulated controls. Finally, the administration by retrodialysis into the preoptic area, of NA increased the proportion of ewes with an LH response to ram odor whereas treatment with the α1 antagonist Prazosin decreased the LH pulse frequency and amplitude induced by a sexually active ram. Collectively these results suggest that in anoestrous ewes NA is involved in ram-induced LH secretion as observed in other induced ovulators
Somatic growth dynamics of West Atlantic hawksbill sea turtles: a spatio-temporal perspective

sea turtles (Eretmochelys imbricata) are long-lived, major consumers in coral reef habitats that move over
broad geographic areas (hundreds to thousands of kilometers). We evaluated spatio-temporal effects on
hawksbill growth dynamics over a 33-yr period and 24 study sites throughout the West Atlantic and explored
relationships between growth dynamics and climate indices. We compiled the largest ever data set
on somatic growth rates for hawksbills – 3541 growth increments from 1980 to 2013. Using generalized additive
mixed model analyses, we evaluated 10 covariates, including spatial and temporal variation, that could
affect growth rates. Growth rates throughout the region responded similarly over space and time. The lack
of a spatial effect or spatio-temporal interaction and the very strong temporal effect reveal that growth rates
in West Atlantic hawksbills are likely driven by region-wide forces. Between 1997 and 2013, mean growth
rates declined significantly and steadily by 18%. Regional climate indices have significant relationships with
annual growth rates with 0- or 1-yr lags: positive with the Multivariate El Niño Southern Oscillation Index
(correlation = 0.99) and negative with Caribbean sea surface temperature (correlation = −0.85). Declines
in growth rates between 1997 and 2013 throughout the West Atlantic most likely resulted from warming
waters through indirect negative effects on foraging resources of hawksbills. These climatic influences are
complex. With increasing temperatures, trajectories of decline of coral cover and availability in reef habitats
of major prey species of hawksbills are not parallel. Knowledge of how choice of foraging habitats, prey
selection, and prey abundance are affected by warming water temperatures is needed to understand how
climate change will affect productivity of consumers that live in association with coral reefs
Synthetic biology: Understanding biological design from synthetic circuits
An important aim of synthetic biology is to uncover the design principles of natural biological systems through the rational design of gene and protein circuits. Here, we highlight how the process of engineering biological systems — from synthetic promoters to the control of cell–cell interactions — has contributed to our understanding of how endogenous systems are put together and function. Synthetic biological devices allow us to grasp intuitively the ranges of behaviour generated by simple biological circuits, such as linear cascades and interlocking feedback loops, as well as to exert control over natural processes, such as gene expression and population dynamics
Force spectroscopy in studying infection
Biophysical force spectroscopy tools - for example optical tweezers, magnetic
tweezers, atomic force microscopy, - have been used to study elastic,
mechanical, conformational and dynamic properties of single biological
specimens from single proteins to whole cells to reveal information not
accessible by ensemble average methods such as X-ray crystallography, mass
spectroscopy, gel electrophoresis and so on. Here we review the application of
these tools on a range of infection-related questions from antibody-inhibited
protein processivity to virus-cell adhesion. In each case we focus on how the
instrumental design tailored to the biological system in question translates
into the functionality suitable for that particular study. The unique insights
that force spectroscopy has gained to complement knowledge learned through
population averaging techniques in interrogating biomolecular details prove to
be instrumental in therapeutic innovations such as those in structure-based
drug design
Deconstructing, Addressing, and Eliminating Racial and Ethnic Inequities in Prostate Cancer Care
Context
Men of African ancestry have demonstrated markedly higher rates of prostate cancer mortality than men of other races and ethnicities around the world. In fact, the highest rates of prostate cancer mortality worldwide are found in the Caribbean and Sub-Saharan West Africa, and among men of African descent in the USA. Addressing this inequity in prostate cancer care and outcomes requires a focused research approach that creates durable solutions to address the structural, social, environmental, and health factors that create racial disparities in care and outcomes.
Objective
To introduce a conceptual model for evaluating racial inequities in prostate cancer care to facilitate the development of translational research studies and interventions.
Evidence acquisition
A collaborative review of literature relevant to racial inequities in prostate cancer care and outcomes was performed. Existing literature was used to highlight various components of the conceptual model to inform future research and interventions toward equitable care and outcomes.
Evidence synthesis
Racial inequities in prostate cancer outcomes are driven by a series of structural and social determinants of health that impact exposures, mediators, and outcomes. Social determinants of equity, such as laws/policies, economic systems, and structural racism, affect the inequitable access to environmental and neighborhood exposures, in addition to health care access. Although the incidence disparity remains problematic, various studies have demonstrated parity in outcomes when social and health factors, such as access to equitable care, are normalized. Few studies have tested interventions to reduce inequities in prostate cancer among Black men.
Conclusions
Worldwide, men of African ancestry demonstrate worse outcomes in prostate cancer, a phenomenon driven largely by social factors that inform biologic, environmental, and health care risks. A conceptual model was presented that organizes the many factors that influence prostate cancer incidence and mortality. Within that framework, we must understand the current state of inequities in clinical prostate cancer practice, the optimal state of what equitable practice would be, and how achieving equity in prostate cancer care balances costs, benefits, and harms. More robust characterization of the sources of prostate cancer inequities should inform testing of ambitious and innovative interventions as we work toward equity in care and outcomes.

Establishing a global quality of care benchmark report.
BACKGROUND: The Movember funded TrueNTH Global Registry (TNGR) aims to improve care by collecting and analysing a consistent dataset to identify variation in disease management, benchmark care delivery in accordance with best practice guidelines and provide this information to those in a position to enact change. We discuss considerations of designing and implementing a quality of care report for TNGR. METHODS: Eleven working group sessions were held prior to and as reports were being built with representation from clinicians, data managers and investigators contributing to TNGR. The aim of the meetings was to understand current data display approaches, share literature review findings and ideas for innovative approaches. Preferred displays were evaluated with two surveys (survey 1: 5 clinicians and 5 non-clinicians, 83% response rate; survey 2: 17 clinicians and 18 non-clinicians, 93% response rate). RESULTS: Consensus on dashboard design and three data-display preferences were achieved. The dashboard comprised two performance summary charts; one summarising site's relative quality indicator (QI) performance and another to summarise data quality. Binary outcome QIs were presented as funnel plots. Patient-reported outcome measures of function score and the extent to which men were bothered by their symptoms were presented in bubble plots. Time series graphs were seen as providing important information to supplement funnel and bubble plots. R Markdown was selected as the software program principally because of its excellent analytic and graph display capacity, open source licensing model and the large global community sharing program code enhancements. CONCLUSIONS: International collaboration in creating and maintaining clinical quality registries has allowed benchmarking of process and outcome measures on a large scale. A registry report system was developed with stakeholder engagement to produce dynamic reports that provide user-specific feedback to 132 participating sites across 13 countries
