Characterization of stem rust resistance gene Sr2 in Indian wheat varieties using polymerase chain reaction (PCR) based molecular markers
Stem rust or black rust is one of the most important diseases of wheat worldwide. In India, central, peninsular and southern hill zones are particularly prone to stem rust where favourable environmental conditions exist. The recent emergence of wheat stem rust race Ug99 (TTKSK) and related strains threatens global wheat production as Ug99 overcome resistance gene Sr31 effective for many years. Resistance gene Sr2, derived from cultivar ‘Hope’ is responsible for slow rusting and providing partial but durable resistance against stem rust of wheat. In addition to other unknown minor genes (Sr2 complex), this gene tends to be non-specific and is currently effective against all isolates of Puccinia graminis tritici throughout wheat-growing regions of the world. A set of 135 bread wheat varieties developed in the last forty years for prominent northern, central, peninsular and southern hill regions of India was screened with molecular markers, CsSr2 and GWM533, developed and identified for Sr2 gene. Out of 135 varieties screened, 92 confirmed the presence of Sr2 gene at molecular level. The molecular information of Sr2 gene was corroborated with the available morphological marker data for selected varieties to evaluate the efficacy of these molecular markers in Indian wheat germplasm. It was observed that these two molecular markers in combination provide accuracy in 92% lines for this gene at molecular level with presumed Sr2 status in Indian wheat varieties. It is proposed that the use of CsSr2 and GWM533 will help in gene pyramiding of Sr2 along with other stem rust resistance genes in future wheat varieties to accelerate Indian breeding program for rust resistance.Keywords: Wheat, stem rust, Puccinia graminis f.sp. tritici (Pgt), Sr2 gene, molecular markerAfrican Journal of Biotechnology Vol. 12(18), pp. 2353-235
Mapping age- and sex-specific HIV prevalence in adults in sub-Saharan Africa, 2000-2018.
BACKGROUND: Human immunodeficiency virus and acquired immune deficiency syndrome (HIV/AIDS) is still among the leading causes of disease burden and mortality in sub-Saharan Africa (SSA), and the world is not on track to meet targets set for ending the epidemic by the Joint United Nations Programme on HIV/AIDS (UNAIDS) and the United Nations Sustainable Development Goals (SDGs). Precise HIV burden information is critical for effective geographic and epidemiological targeting of prevention and treatment interventions. Age- and sex-specific HIV prevalence estimates are widely available at the national level, and region-wide local estimates were recently published for adults overall. We add further dimensionality to previous analyses by estimating HIV prevalence at local scales, stratified into sex-specific 5-year age groups for adults ages 15-59 years across SSA. METHODS: We analyzed data from 91 seroprevalence surveys and sentinel surveillance among antenatal care clinic (ANC) attendees using model-based geostatistical methods to produce estimates of HIV prevalence across 43 countries in SSA, from years 2000 to 2018, at a 5 × 5-km resolution and presented among second administrative level (typically districts or counties) units. RESULTS: We found substantial variation in HIV prevalence across localities, ages, and sexes that have been masked in earlier analyses. Within-country variation in prevalence in 2018 was a median 3.5 times greater across ages and sexes, compared to for all adults combined. We note large within-district prevalence differences between age groups: for men, 50% of districts displayed at least a 14-fold difference between age groups with the highest and lowest prevalence, and at least a 9-fold difference for women. Prevalence trends also varied over time; between 2000 and 2018, 70% of all districts saw a reduction in prevalence greater than five percentage points in at least one sex and age group. Meanwhile, over 30% of all districts saw at least a five percentage point prevalence increase in one or more sex and age group. CONCLUSIONS: As the HIV epidemic persists and evolves in SSA, geographic and demographic shifts in prevention and treatment efforts are necessary. These estimates offer epidemiologically informative detail to better guide more targeted interventions, vital for combating HIV in SSA

Effect of sowing date on the incidence of loose smut, flag smut and seed gall of wheat
An experiment was conducted to find out the effect of delay sowing on incidence of loose smut (Ustilago tritici), flag smut (Urocystis agropyri) and seed gall (Anguina tritici) of wheat. Varieties WH 896 and Raj 1555 were free from loose smut at all dates of sowing. At normal date of snwing (25 November) disease incidence varied from 28.18% (HD 2285) to 43.18 % (Sonalika). Due to delay in sowing by 20 days (15 December), the disease incidence was reduced to 21.33-38.18% and delay by 35 days (30 December) it was reduced to 18.62% -32.5 % on all susceptible varieties. At all dates of sowing, varieties WH 283, WI-I 291, WH 896 and Raj 1555 were not infected with flag smut. Other varieties were infected with the disease showing 1.3-14.57, 1.11-11.83 and 0.83-9.14% incidence at the sowing dates of 25 November (normal), 15 December (delay) and 30 December (delay), respectively. All of the varieties tested in this study were infected with seed gall nematode and disease incidence increased with delay in date of sowing. Epidemiology and prediction models for themanagement of rapeseed–mustard diseases: current status and future needs
RESISTANCE GENES AGAINST YELLOW RUST PATHOGEN IN TRITICUM SPELTA: A POSSIBLE NEW Yr RESISTANCE GENE IN ACCESSION IARI276 AND Yr5 PRESENCE CONFIRMED IN PI348764
Yellow/stripe rust caused by Puccinia striiformis f. sp. tritici (Pst) is a major biotic stress in global wheat production. Introgression lines derived from the Triticum spelta accessions PI348764 and IARI276 showed high levels of yellow rust resistance at seedling and adult plant stage. The Yr5 gene located on 2B chromosome was previously the only stripe rust resistance gene described in T. spelta gene pool. By genotyping parental and introgressed material with markers linked with the Yr5 gene, we demonstrate that PI348764 likely carries Yr5, and that it appears to be absent from IARI276. By employing a combination of methods, including screening for adult plant resistance and seedling resistance at multiple field trials; bulked segregant analysis (BSA) on F5 families, and genotyping using wheat Breeders' 35K array; we show that YrIARI276 is a novel stripe rust resistance gene with putative chromosomal locations on 1BL, 1DL, 5AL or 7BL. Furthermore, genetic analysis revealed that YrIARI276 showed a goodness of fit to Mendelian ratios for a single dominant gene.
